Book excerpt: New general methodology for the stereoselective synthesis of unnatural alpha-amino acids has been developed. Early work focussed on investigating methods for the generation of chiral allylic alcohols using cross-metathesis of a simple enone with various terminal alkenes, followed by an asymmetric ketone reduction. The allylic alcohols were then converted to protected allylic amines via Overman rearrangement chemistry. Oxidative alkene cleavage and hydrolysis of these intermediates generated a range of alpha-amino acid targets. Attempts were also made to apply the developed methodology to the synthesis of a simple alpha,alpha-disubstituted alpha-amino acid target. The Overman rearrangement was also applied to the generation of a late-stage intermediate which could be used in the synthesis of (2S,3S)-capreomycidine, a component of a number of peptides which exhibit antibacterial activity.

Available in PDF, EPUB and Kindle. Book excerpt: The purpose of this book is to review and critically evaluate the best new methods to synthesize alpha-amino acids in optically active form. There is so much new literature on amino acid synthesis that the experimentalist will undoubtedly have difficulty in selecting the most appropriate methodology for constructing the amino acid of immediate interest. This book is a guide for steering the scientist through the maze of existing reports on the subject and contains the most up-to-date critical reviews of methods of asymmetric synthesis of amino acids. In areas that are relatively new conceptually and less studied experimentally, an effort has been made to review the most salient works with an eye towards future development. Over 330 schemes and figures are presented with references for rapid visual retrieval of information. The book will be of great value to academic and industrial organic research chemists, especially those concerned with medicinal and agricultural chemistry, as well as to graduate and post graduate students, biochemists and biologists.

Available in PDF, EPUB and Kindle. Book excerpt: Covers all facets of the synthesis of ß-amino acids As evidenced by an exponential increase in the literature published on the subject, interest in ß-amino acids has grown over the past several years. With major pharmaceutical applications, these amino acids are now studied across multiple lines of research, including combinatorial chemistry, medicinal chemistry, molecular design, proteomics, and others. This Second Edition of Enantioselective Synthesis of ß-Amino Acids updates reviews included in the First Edition while also covering new developments since its publication. The book presents detailed discussions of the most important methods for the synthesis of ß-amino acids. In most cases, the lead chemist who originally developed a method provides an authoritative description of it. In addition, Enantioselective Synthesis of ß-Amino Acids, Second Edition: * Features introductory overviews on the structural types of relevant ß-amino acid targets and salient ß-amino acids present in natural products * Dedicates several chapters to advances in the synthesis of oligomers from ß-amino acids * Includes general and practical procedures for the preparation of ß-amino acids in each chapter * Discusses the most important methods that have been recently developed for the asymmetric synthesis of cyclic and open-chain ß-amino acids * Includes a report on the preparation of libraries of enantiopure ß-amino acids using combinatorial approaches The only book of its kind available today, Enantioselective Synthesis of ß-Amino Acids, Second Edition offers upper-level students and professionals an essential resource for pharmaceutical development, medicinal chemistry, and biochemistry.

Thoroughly revised and updated, this enlarged second edition offers a plethora of valuable information on methods and reagents in stereoselective synthesis. Methods have been selected for high efficiency and selectivity; mechanistic aspects are treated succinctly, with a strong emphasis on practical applications. For this new edition, material has been added on * homogeneous diastereoselective hydrogenations * enantioselective oxidations * novel, efficient chiral auxiliaries Much of the information given is presented in figures and tables, which makes the book a valuable reference work for the practically minded organic chemist. From reviews of the first edition: 'The extensive material in the volume should prove particularly useful to anyone involved in synthetic chemistry or teaching a course in organic chemistry.' Journal of Medicinal Chemistry 'With nearly 1400 references cited, the book contains a wealth of information and should be a useful addition to the chemist's library.' The American Scientist

Book excerpt: The thermal and metal-catalysed aza-Claisen rearrangement of allylic trichlororacetimidates has found widespread application in the synthesis of nitrogen-containing molecules including alkaloids, antibiotics and unnatural amino acids. We have recently investigated the use of this reaction for the rearrangement of chiral molecules. This has led to the development of a highly diastereoselective, ether-directed palladium(II)-catalysed process which has been used for the synthesis of ß-hydroxy-a-amino acids. To expand the scope and understanding of the rearrangement, a number of analogues of 1 have been synthesised and subjected to a palladium(II)-catalysed rearrangement. Gizzerosine ((S)-2-amino-9-(4-imidazolyl)-7-azanonanoic acid) is a potent agonist for the H2-receptor. It brings about gizzard erosion and ulceration in broiler chickens. This disease is colloquially known as Â'Black VomitÂ' due to bleeding of lesions and subsequent regurgitation. Its biological profile suggests it could be a promising drug candidate for gastric achlorhydria and osteoporosis, although only small amounts can be isolated from fish meal for biological studies. A short and highly efficient synthesis has been developed in ten steps and 31% overall yield. Protein-protein inteactions are the key to organising cellular processes in space and time. Protein-protein interactions are involved in viral fusion and so are promising targets for anti-viral drugs. Photoactivatable amino acids have been used to identify these interactions. Activation by ultraviolet light induces covalent cross-linking (known as photo cross-linking) of the interacting proteins which can be detected with high specificity by simple western blotting. Significant progress has been made towards the development of a stereoselective synthesis of photoactivatable amino acids incorporating a diazirine ring, such as photo-leucine, which generate a reactive carbene after the light induced loss of nitrogen.

Book excerpt: In recent years, organo-fluorine chemistry has made a marked impact on the design and synthesis of a large variety of biologically active molecules, such as steroids, carbohydrates, amines, amino acids, peptides and other natural products. Naturally occurring amino acids play a pivotal role in living systems, and therefore synthetic fluorine-containing amino acids have been of significant interest to researchers working towards the understanding and modification of physiological processes. Fluorine-containing Amino Acids: is the first volume devoted to the synthesis and properties of fluorine-containing amino acids pays special attention to the preparation of enantiomerically pure acids (which are essential to the modern pharmaceutical industry) deals with a rapidly expanding field of research has been written by experienced researchers who are responsible for many developments in the field highlights the interdisciplinary nature of this topic Fluorine-containing Amino Acids is the only dedicated reference in this subject and will be essential for researchers in synthetic organic, peptide, natural product, and medicinal chemistry and biochemistry.
