Book excerpt: To date, numerous candidate genes have been associated with autism spectrum disorder (ASD) with many of these genes known to have important roles in synaptic function and the development of neural circuits. This suggests that certain neurobiological processes could be commonly altered in ASD. Therefore, although there is a great deal of clinical and genetic heterogeneity in ASDs, there may be convergent deficits in key molecular mechanisms which underlie the disease. Although this is encouraging, a lack of appropriate human-based models of complex neurodevelopmental disorders has greatly hindered investigations of convergent neurobiology in ASD. Patient-specific induced pluripotent stem cells (iPSCs) can be used to model brain region-specific neuronal development in genetic backgrounds known to result in certain pathological conditions. This ability to model neurodevelopment in vitro permits the characterization of disease mechanisms that manifest themselves during the early steps of brain development in ASD, facilitating the identification of common molecular mechanisms disrupted in this disorder. Importantly, the identification of common pathways in ASD that are disrupted in the early stages of disease development could serve as important candidate targets for therapeutic intervention. Patient-specific iPSC lines were derived from the whole blood of individuals affected with idiopathic ASD, and subsequently differentiated into cortical neurons for up to 135 days in culture. The findings of transcriptome and gene network analysis indicate that iPSC-derived neurons from our cohort of ASD individuals have altered expression of genes associated with synaptic function, Wnt signaling, neuronal differentiation, and various processes involving the extracellular matrix (i.e. axon guidance and cell migration). Furthermore, our data show that genes previously implicated in ASD and intellectual disability are significantly overrepresented in gene coexpression networks related to synaptic function that are transcriptionally co-regulated by Wnt. Finally, we demonstrate that neurons derived from our sample cohort of idiopathic ASD individuals have excessive neurite outgrowth at an early stage of cortical neurogenesis. In sum, our data indicate that convergent molecular disturbances in ASD impact synaptic development and function, metabolism, and cellular molecular interactions involving the extracellular matrix. Furthermore, our results from transcriptome and gene-network analysis both indicate considerable transcriptional dysregulation during earlier stages of neuronal development.

Here, using a computation-assisted interaction proteomics approach in neural cells including primary neurons, we isolate high-confidence binding partners of proteins linked to ASD and ID. As part of these studies, we uncover the brain-enriched, coiled-coil domain protein huntingtin-associated protein 1 (Hap1) as a novel functional binding partner of the tuberous sclerosis complex (TSC) protein Tsc1. We validate and map the Hap1-Tsc1 interaction, and find that Hap1 and Tsc1 form a complex endogenously in the brain. Hap1 knockdown in primary hippocampal neurons triggers the specification of supernumerary axons, and in utero knockdown of Hap1 in mice profoundly impairs the positioning of pyramidal neurons in the hippocampus in vivo. Importantly, the Hap1 knockdown-induced phenotypes in primary neurons and in vivo recapitulate the phenotypes induced by Tsc1 knockdown. We also define a mechanism by which Hap1 regulates Tsc1 function. We observed that exogenous Hap1 promotes the abundance of soluble, stable Tsc1 expressed in cells. Hap1 knockdown in neurons reduces Tsc1 abundance and accordingly stimulates the activity of mTORC1, as reflected by phosphorylation of the ribosomal protein S6. Importantly, inhibition of mTORC1 signaling suppresses the Hap1 knockdown-induced axon phenotype in hippocampal neurons. Collectively, these findings define a novel relationship between Hap1 and Tsc1 that regulates neuronal Tsc1 abundance, pyramidal neuron development, and neuronal mTORC1 signaling, with important mechanistic implications for our understanding of neurodevelopmental disorders of cognition.

Many neurodevelopmental disorders are caused by dysregulation of the mechanisms that promote the formation of normal synaptic connections. Here, I show that dysregulation of neuronal stress pathways during development negatively impacts synaptic growth and functionality. First, I explore the Drosophila model of Fragile X Syndrome, a common and debilitating disorder that causes intellectual disability and autism spectrum disorders in humans. I show that chronic, excess signaling through the Wallenda/DLK stress pathway drives synaptic dysfunction, circuit dysregulation, and abnormal behaviors. Second, I show that the NAD+ synthesizing enzyme, dNmnat, which is an axoprotective factor, must be degraded at the synapse to promote synaptic transmission at the developing larval neuromuscular junction. In total, this work shows that the pathways that govern neuronal stress responses must be downregulated during development to promote normal nervous system function. This highlights a new paradigm in which to consider neurodevelopmental disorders: chronic upregulation of stress pathways may contribute to the lifelong symptoms associated with these disorders. This implies that not all neuronal dysfunction is permanently set during development. Thus, there is hope for the application of therapies in neurodevelopmental disorders that target active signaling through neuronal stress pathways.
