Book excerpt: The Streptomycetes are industrially widely used microorganisms due to their ability to produce numerous different chemical compounds. These show very varied effects upon other living systems, and result from profound and subtle biochemical and morphological differentiation during the streptomycete life cycle. It is therefore not surprising that research on the genetics of antibiotic biosynthesis and differentiation in this group is currently progressing rapidly in many countries. Intimately connected with the production of antibiotics is resistance to them; analysis of this is giving further information about the origin and evolution of this class of genes and their hypothesized spread among other microorganisms. Another interesting feature of the Streptomyces group is their mycelial growth. Also, their ecologically important utilization of high molecular weight compounds requires enzymes to be transported outside the cell to hydrolyze non-diffusible substrates. Finally, we have as yet limited understanding of the various mechanisms of genome rearrangement observed in some of these species; deletions and/or amplifications of enormous amounts of DNA can occur without seriously affecting the viability of the organism under laboratory conditions. The present volume, which includes contributions addressing the above subjects and others, originates from a meeting on "Genetics and Product Formation in Streptomyces" sponsored by the Federation of European Microbiological Societies in Erfurt on May 1-6 1990. Compared to previous ones of this kind held in 1979, 1983 and 1987 in Weimar, one can point to impressive progress in the study and applications of Streptomyces genetics.

Book excerpt: Microbial natural products have been an important traditional source of valuable antibiotics and other drugs but interest in them waned in the 1990s when big pharma decided that their discovery was no longer cost-effective and concentrated instead on synthetic chemistry as a source of novel compounds, often with disappointing results. Moreover understanding the biosynthesis of complex natural products was frustratingly difficult. With the development of molecular genetic methods to isolate and manipulate the complex microbial enzymes that make natural products, unexpected chemistry has been revealed and interest in the compounds has again flowered. This two-volume treatment of the subject will showcase the most important chemical classes of complex natural products: the peptides, made by the assembly of short chains of amino acid subunits, and the polyketides, assembled from the joining of small carboxylic acids such as acetate and malonate. In both classes, variation in sub-unit structure, number and chemical modification leads to an almost infinite variety of final structures, accounting for the huge importance of the compounds in nature and medicine. - Gathers tried and tested methods and techniques from top players in the field - In depth coverage of ribosomally-synthesised and Non-ribosomally-synthesised peptides - Provides an extremely useful reference for the experienced research scientist

Available in PDF, EPUB and Kindle. Book excerpt: Penicillins and cephalosporins have a long history in combating bacterial infections. Despite new infectious diseases and occurring resistance, beta-lactam antibiotics will for many years to come continue to play a prominent role in our therapeutic arsenal. This book covers the industrial development of the chemical and biochemical processes used to manufacture these products, as well as looking ahead to possible future processes. The interplay between synthetic organic chemistry with the understanding and application of enzymes, modeling of fermentation processes and integration through (bio-) chemical process engineering is illustrated. In-depth scientific approaches to biocatalysis and biocatalyst development including enzyme kinetics, enzyme crystal studies and semi-rational enzyme mutations are also presented. Metabolic pathway analysis and modeling of fermentation process are treated as well as molecular precision in synthetic approaches to beta-lactams, their precursors and derivatives. Process technology studies including new reactor concepts, possible short-cut routes and improved down-stream-processing methods complete a broad view on the scope and limitations of the presently developed industrial processes including an intriguing insight into future process possibilities. This book represents an excellent case study on the transformation of traditional, stoïchiometric, organic synthesis and classical fermentations into modern (bio-) catalysis and biosynthesis based on insights in metabolic pathways and enzyme actions.

Book excerpt: This is the second edition of the text "Bioreaction Engineering Principles" by Jens Nielsen and John Villadsen, originally published in 1994 by Plenum Press (now part of Kluwer). Time runs fast in Biotechnology, and when Kluwer Plenum stopped reprinting the first edition and asked us to make a second, revised edition we happily accepted. A text on bioreactions written in the early 1990's will not reflect the enormous development of experimental as well as theoretical aspects of cellular reactions during the past decade. In the preface to the first edition we admitted to be newcomers in the field. One of us (JV) has had 10 more years of job training in biotechnology, and the younger author (IN) has now received international recognition for his work with the hottest topics of "modem" biotechnology. Furthermore we are happy to have induced Gunnar Liden, professor of chemical reaction engineering at our sister university in Lund, Sweden to join us as co-author of the second edition. His contribution, especially on the chemical engineering aspects of "real" bioreactors has been of the greatest value. Chapter 8 of the present edition is largely unchanged from the first edition. We wish to thank professor Martin Hjortso from LSU for his substantial help with this chapter.

Written by experts in their specialist fields the series creates a unique service for the active research chemist, supplying regular critical in-depth accounts of progress in particular areas of chemistry. For over 80 years the Royal Society of Chemistry and its predecessor, the Chemical Society, have been publishing reports charting developments in chemistry, which originally took the form of Annual Reports. However, by 1967 the whole spectrum of chemistry could no longer be contained within one volume and the series Specialist Periodical Reports was born. The Annual Reports themselves still existed but were divided into two, and subsequently three, volumes covering Inorganic, Organic and Physical Chemistry. For more general coverage of the highlights in chemistry they remain a 'must'. Since that time the SPR series has altered according to the fluctuating degree of activity in various fields of chemistry. Some titles have remained unchanged, while others have altered their emphasis along with their titles; some have been combined under a new name whereas others have had to be discontinued. The current list of Specialist Periodical Reports can be seen on the inside flap of this volume.
