Book excerpt: Cell Culture and Somatic Cell Genetics of Plants, Volume 6: Molecular Biology of Plant Nuclear Genes focuses on the spectacular and rapid advances in the molecular biology and genetics of plants. This book consists of 19 chapters. Chapters 1 to 5 describe the most commonly used approaches for the genetic transformation of plants. The light-inducible and tissue-organ-specific genes are discussed in Chapters 6 to 11. In Chapters 12 to 14, the genes regulating phytohormone synthesis, heat shock proteins, and nodulation in legume roots are treated, while in Chapters 15 to 16, the relationship between chromatin structure and gene expression and molecular biology of plant RNA viruses are analyzed. The development of transgenic plants resistant to viruses, insects, and herbicides is dealt with in the last three chapters. This volume is suitable for plant molecular biologist, genetic engineers, and researchers concerned with plant cell and tissue culture.

Available in PDF, EPUB and Kindle. Book excerpt: This book provides detailed information on methodologies used in biological, serological and nucleic acid based assays for the detection, diagnosis and management of plant viruses. The content is divided into six main parts, the first of which presents techniques used in the biological characterization and transmission of viruses, while Part II covers purification and techniques concerning the physico-chemical properties of viruses. In turn, Part III focuses on in vitro expression, production of polyclonal and monoclonal antibodies; and on various serological assays including precipitin tests, ELISA, blot immunoassays, immunosorbent electron microscopy and lateral flow immunoassays. Part IV addresses the isolation of DNA and RNA from plants and nucleic acid based assays such as dot-blot, polymerase chain reaction, real-time PCR, loop-mediated isothermal amplification, rolling circle amplification, recombinase polymerase amplification, and next-generation sequencing approaches. Part V discusses cloning, sequencing, sequence analysis and the production of infectious clones, while the last part (Part VI) provides biotechnological approaches for the management of viruses. Given its scope, the book will be a valuable resource for every laboratory, student and teacher, and for everyone interested in plant virology, plant pathology, plant biology and molecular biology, offering them a practical manual on various aspects of plant viruses.

Available in PDF, EPUB and Kindle. Book excerpt: Concern about the environmental consequences of the widespread use of pesticides has increased, and evidence of pesticide-resistant virus vectors have continued to emerge. This volume presents a timely survey of the mechanisms of plant resistance and examines current developments in breeding for resistance, with particular emphasis on advances in genetic engineering which allow for the incorporation of viral genetic material into plants. Discusses the mechanisms of innate resistance in strains of tobacco, tomato, and cowpea; various aspects of induced resistance, including the characterization and roles of the pathogenesis-related proteins; antiviral substances and their comparison with interferon; and cross-protection between plant virus strains. Also presents several papers which evaluate the status of genetic engineering as it relates to breeding resistant plants. Among these are discussions of the potential use of plant viruses as gene vectors, gene coding for viral coat protein, satellite RNA, and antisense RNA, and practical issues such as the durability of resistant crop plants in the field.

Available in PDF, EPUB and Kindle. Book excerpt: There are over a million different species of insects, and individually they 8 outnumber humans by more than 10 to 1. Moreover, some insects live in close association with both plants and higher animals and naturally exchange viruses with them. It has even been speculated that viruses in general may have radiated through the plant and animal kingdoms from common insect origins. Be that as it may, since insects play pivotal roles in the biosphere, both to the benefit and detriment of mankind, they and the viruses that infect them are important subjects for study. Insects are infected by a diverse medley of viruses, and this volume focuses on those insect virus families that are found primarily or exclusively in insects. All major families of insect-selective viruses are covered except for the baculo viruses, which were described in a separate volume of The Viruses series. Included in this volume are the established families of insect viruses, the newly recognized ascovirus family, and the nudiviruses, which probably represent a separate family but currently remain unclassified. The coverage of arboviruses is limited to a single chapter that focuses on their potential utility as vectors and in insect control. Omitted for practical reasons are several individual insect viruses that have been described sporadically in the literature and probably represent novel virus families.

This book was released on 2012-12-06 with total page 372 pages. Available in PDF, EPUB and Kindle. Book excerpt: The development of recombinant DNA technology has made a marked impact on molecular virology. The cleavage of viral DNA genomes with restriction enzymes and the cloning of such DNA fragments in bacterial p1asmids has led to the amplification of selected viral DNA fragments for sequencing and gene expression. RNA virus genomes which can be transcribed to their cDNA form were also cloned in bacterial p1asmids, facilitating the study of RNA virus genes. With the elucidation in recent years of the promoter sequence of various viral genes and the expression of these genes in bacteria or yeast, the understanding of many viral gene functions has made great progress. Cloning and expression of viral genes in mammalian cells was made possible by the construction of shuttle plasmid vectors which carry the origins of DNA replication from bacteria and/or mammalian viruses. The expression of viral genes in bacteria, yeast and eukaryotic cells gives reason to hope that it will be possible to produce viral antigens in large quantities for use as human or animal vaccines. The present volume attempts to capture for the reader some of the high lights of recombinant DNA research in the field of animal and plant viruses.
