This book was released on 2008-02-04 with total page 368 pages. Available in PDF, EPUB and Kindle. Book excerpt: Fluorescent nucleic acid probes, which use energy transfer, include such constructs as molecular beacons, molecular break lights, Scorpion primers, TaqMan probes, and others. These probes signal detection of their targets by changing either the intensity or the color of their fluorescence. Not surpr- ingly, these luminous, multicolored probes carry more flashy names than their counterparts in the other fields of molecular biology. In recent years, fluor- cent probes and assays, which make use of energy transfer, have multiplied at a high rate and have found numerous applications. However, in spite of this explosive growth in the field, there are no manuals summarizing different p- tocols and fluorescent probe designs. In view of this, the main objective of Fluorescent Energy Transfer Nucleic Acid Probes: Designs and Protocols is to provide such a collection. Oligonucleotides with one or several chromophore tags can form fluor- cent probes capable of energy transfer. Energy transport within the probe can occur via the resonance energy transfer mechanism, also called Förster tra- fer, or by non-Förster transfer mechanisms. Although the probes using Förster transfer were developed and used first, the later non-Förster-based probes, such as molecular beacons, now represent an attractive and widely used option. The term “fluorescent energy transfer probes” in the title of this book covers both Förster-based fluorescence resonance energy transfer (FRET) probes and probes using non-FRET mechanisms. Energy transfer probes serve as molecule-size sensors, changing their fluorescence upon detection of various DNA reactions.

Book Synopsis Handbook of Molecular Microbial Ecology II by : Frans J. de Bruijn
Download or read book Handbook of Molecular Microbial Ecology II written by Frans J. de Bruijn and published by John Wiley & Sons. This book was released on 2011-09-27 with total page 632 pages. Available in PDF, EPUB and Kindle. Book excerpt: The premiere two-volume reference on revelations from studying complex microbial communities in many distinct habitats Metagenomics is an emerging field that has changed the way microbiologists study microorganisms. It involves the genomic analysis of microorganisms by extraction and cloning of DNA from a group of microorganisms, or the direct use of the purified DNA or RNA for sequencing, which allows scientists to bypass the usual protocol of isolating and culturing individual microbial species. This method is now used in laboratories across the globe to study microorganism diversity and for isolating novel medical and industrial compounds. Handbook of Molecular Microbial Ecology is the first comprehensive two-volume reference to cover unculturable microorganisms in a large variety of habitats, which could not previously have been analyzed without metagenomic methodology. It features review articles as well as a large number of case studies, based largely on original publications and written by international experts. This second volume, Metagenomics in Different Habitats, covers such topics as: Viral genomes Metagenomics studies in a variety of habitats, including marine environments and lakes, soil, and human and animal digestive tracts Other habitats, including those involving microbiome diversity in human saliva and functional intestinal metagenomics; diversity of archaea in terrestrial hot springs; and microbial communities living at the surface of building stones Biodegradation Biocatalysts and natural products A special feature of this book is the highlighting of the databases and computer programs used in each study; they are listed along with their sites in order to facilitate the computer-assisted analysis of the vast amount of data generated by metagenomic studies. Such studies in a variety of habitats are described here, which present a large number of different system-dependent approaches in greatly differing habitats. Handbook of Molecular Microbial Ecology II is an invaluable reference for researchers in metagenomics, microbial ecology, microbiology, and environmental microbiology; those working on the Human Microbiome Project; microbial geneticists; and professionals in molecular microbiology and bioinformatics.
Book Synopsis Preimplantation Genetics by : A. Kuliev
Download or read book Preimplantation Genetics written by A. Kuliev and published by Springer Science & Business Media. This book was released on 2012-12-06 with total page 314 pages. Available in PDF, EPUB and Kindle. Book excerpt: Yury Verlinskyand Anver Kuliev Reproductive Genetics Institute, Illinois Masonic Medical Center, 836 W. Wellington chicago, IL 60657 Although introduction of a first trimester prenatal diagnosis by chorionic viIIus sampling (CVS) has considerably improved the possibility for prevention of genetic diseases, it requires a selective abortion in case of an affected fetus. Following the direction of an earlier prenatal diagnosis and to avoid the need for abortion, preimplantation genetic diagnosis has been initiated based on polar body removal and pre-embryo biopsy. The First International symposium on Preimplantation Genetics, Chicago, September 17-19, 1990, was organized to explore these important developments, to review the state of knowledge in the field, and to address existing problems to be solved for developing and improving current approaches for preimplantation diagnosis of genetic disorders. A growing interest in the subject was obvious from the wide attendance of the meeting: over 250 scientists from 19 countries participated. This was the first attempt to put together the advances in different areas of basic and applied research relevant to Preimplantation Genetic Diagnosis, with the multidisciplinary scientific program including the sessions on embryology, micromanipulation and biopsy, genetic analysis of gametes and pre-embryos, IVF, gene expression and gene therapy, and ethical and legal issues. The deliberations of the Symposium presented in the above mentioned sessions, which comprise the contents of correspond ing sections of the Proceedings, open a newarea in medical research based on the interaction of IVF and New Genetics.

Book Synopsis Use of Novel Techniques to Quantify Phenotypes in Biological Treatment Process by : K. G. Robinson
Download or read book Use of Novel Techniques to Quantify Phenotypes in Biological Treatment Process written by K. G. Robinson and published by IWA Publishing. This book was released on 2004-01-01 with total page 220 pages. Available in PDF, EPUB and Kindle. Book excerpt: This research attempts to evaluate nitrification treatment performance in combined carbon/nitrogen municipal wastewater reactors using traditional physical/chemical methods and modern molecular techniques. Bench scale activated sludge reactors were operated at different SRTs under varying DO levels and temperatures over a 21-month period. Real-time PCR assays were used to determine cell concentrations of total bacterial 16S rDNA, a gross measure of biomass content, the amoA gene, a measure of ammonia-oxidizing bacteria (AOB), and the Nitrospira 16S rDNA gene, a measure of nitrite-oxidizing bacteria (NOB). As expected, gravimetric biomass and total bacterial 16S rDNA levels increased with increasing SRT. Ammonia oxidation rates and N. oligotropha-type AOB concentrations did not follow similar trends with respect to changes in SRT, temperature, and DO nor were they highly correlated. The concentration of available nitrite and SRT were positively correlated with Nitrospira cell densities, while DO concentration and temperature were negatively correlated with NOB levels. The percentage of the total population comprised of AOB and NOB obtained with the real-time PCR assays were compared to predicted values estimated from design equations using typical kinetic parameters. While the percentages of NOB measured using the real-time PCR assay corresponded very well with the predicted values, the measured percentages of AOB were much lower than those estimated from the design equations, suggesting that N. oligotropha-type AOB were not the dominant ammonia-oxidizing species in these reactors. Book Synopsis Principles of Fluorescence Spectroscopy by : Joseph R. Lakowicz
Download or read book Principles of Fluorescence Spectroscopy written by Joseph R. Lakowicz and published by Springer Science & Business Media. This book was released on 2013-11-11 with total page 487 pages. Available in PDF, EPUB and Kindle. Book excerpt: Fluorescence methods are being used increasingly in biochemical, medical, and chemical research. This is because of the inherent sensitivity of this technique. and the favorable time scale of the phenomenon of fluorescence. 8 Fluorescence emission occurs about 10- sec (10 nsec) after light absorp tion. During this period of time a wide range of molecular processes can occur, and these can effect the spectral characteristics of the fluorescent compound. This combination of sensitivity and a favorable time scale allows fluorescence methods to be generally useful for studies of proteins and membranes and their interactions with other macromolecules. This book describes the fundamental aspects of fluorescence. and the biochemical applications of this methodology. Each chapter starts with the -theoreticalbasis of each phenomenon of fluorescence, followed by examples which illustrate the use of the phenomenon in the study of biochemical problems. The book contains numerous figures. It is felt that such graphical presentations contribute to pleasurable reading and increased understand ing. Separate chapters are devoted to fluorescence polarization, lifetimes, quenching, energy transfer, solvent effects, and excited state reactions. To enhance the usefulness of this work as a textbook, problems are included which illustrate the concepts described in each chapter. Furthermore, a separate chapter is devoted to the instrumentation used in fluorescence spectroscopy. This chapter will be especially valuable for those perform ing or contemplating fluorescence measurements. Such measurements are easily compromised by failure to consider a number of simple principles.
