Book excerpt: The epidermal growth factor receptor (EOFR) is a cell-surface protein that relays signals from the extracellular environment into the cell by binding specific polypeptide hormones followed by activation of intracellular signal transduction pathways. Although rarely an oncogene, the ability of EGFR-mediated signaling to generate diverse responses including growth, differentiation, stress response, apoptosis suppression, and altered mobility makes this protein a potentially powerful tumor promoter. The association between higher EOFR expression and poorer prognosis in breast cancer and the frequency of higher EGFR levels in more aggressive/metastatic breast tumors reinforce this possibility. Our research aims to uncover a link between chemotherapeutic exposure and increased expression of EOFR in breast cancer cells with the hope of explaining why higher levels of EGFR are common to more advanced breast tumors. We have shown that exposure of MCF-7, T-47D, and ZR-75-1 breast cancer cells to the anti-metabolite compound methotrexate causes an up-regulation of EGFR receptor expression. Our work demonstrates that the EGFR up-regulation usually occurs at both the niRNA level and protein level (with increased expression on the cell surface) and that this may be accompanied by changes in the expression of EGFR ligands. We also demonstrate that the methotrexate-induced EOFR expression in these cells alters EGF-mediated phosphorylation of ERK and AKT (causing changes in the specificity, timing and intensity of EOFR signaling through these pathways in a cell specific manner). ERK and AKT signaling pathways have been shown to mediate anti-apoptotic effects. We therefore hypothesize that increased EOFR expression and signaling in these cells provides a survival advantage by suppression of chemotherapy induced apoptosis, possibly explaining the prevalence of EGFR over-expression in drug resistant cell lines and more aggressive breast tumors.

Book excerpt: Mammary lobuloalveolar development during pregnancy is controlled by the endocrine action of prolactin, estrogen and progesterone, but currently little is known of the transcriptional events that occur in response to these hormones. This study examines the transcriptional response of the mammary gland to the direct action of prolactin. Mammary tissue recombination experiments demonstrated that lobuloalveolar development is dependent upon the presence of prolactin receptors in mammary epithelium but not the stroma. Comparison of transcript profiles derived from recombined glands with and without epithelial prolactin receptors, from glands without epithelium, and from SCp2 cells treated with and without prolactin, allowed a small set of epithelial-specific prolactin-modulated transcripts to be identified. This included the epithelial specific Ets transcription factor Elf5. Elf5 expression decreased in Prlr(+/- ) mammary glands but there was no change in Prlr expression in Elf5(+/- ) mammary glands, indicate that ElfS acts downstream of prolactin. The expression of milk protein genes decreased in both Prlr(+/- ) and Elf5(+/- ) mammary glands, consistent with a role for Elf5 in mediating prolactin-induced epithelial differentiation, however, the expression of a number of growth factors decreased in Prlr(+/- ) mammary glands but increased in Elf5(+/- ) mammary glands. Thus prolactin induces the expression of Elf5, which performs a pivotal role in mediating the switch from prolactin induced epithelial cell proliferation to differentiation within the mammary gland during pregnancy.

Book excerpt: "Triple Negative Breast Cancer (TNBC) is the most resistant subtype of breast cancer. The lack of expression of estrogen, progesterone, and human epidermal growth factor 2 receptors in TNBC leads to the limited use of targeted therapy. Therefore, treatment with chemotherapy remains the main approach for treating TNBC with low therapeutic benefits and increased risk of resistance. Pro-differentiation based therapies by reversing breast cancer cells into an epithelial phenotype have recently been studied as a promising therapeutic approach for TNBC. Indeed, the laboratory of Dr. Ali showed that restoring prolactin pro-differentiation pathway reversed the mesenchymal aggressive phenotype of TNBC into the epithelial form with less aggressive behavior, less invasiveness, and lower metastasis. In addition, they identified a TNBC subgroup of patients that preserved prolactin receptor (PRLR) expression to show better overall survival and better prognosis. By metagenes cluster analysis, the TNBC-PRLR subtype is characterized by the up regulation of luminal-like differentiation genes which are Forkhead Box A1 (FOXA1) and androgen receptor (AR). Our aim is to determine the effect of FOXA1 on the epithelial mesenchymal transition (EMT) of breast cancer cells and how it may modulate the antitumorigenic role of prolactin and its signaling pathway in breast cancer. To achieve our objectives, we measured the expression of FOXA1 at the mRNA and protein levels after treating TNBC-PRLR cell lines with prolactin. We also produced CRISPR knock out of FOXA1 in these cell lines and measured the protein level of both E-cadherin, an epithelial marker and vimentin as a mesenchymal marker. As expected, the knock-out cells showed a decrease in the level of E-cadherin and an increase in vimentin compared to the parental cells. Finally, we conclude that FOXA1 can be a potential pro-differentiation marker in breast cancer cells. Further studies are needed to investigate this role of FOXA1 and its use as a target for personalized medicine in triple negative breast cancer"--
