The methods have been assembled and the protocols drafted by a number of scientists associated with the UNEP-GEF funded Conservation and Sustainable Management of Below-Ground Biodiversity Project, executed by the Tropical Soil Biology and Fertility (TSBF) Institute of the International Center for Tropical Agriculture (CIAT). The methods provide a standardized basis for characterizing soil biodiversity and current land uses in terrestrial natural, semi-natural and agroecosystems in tropical forests and at forest margins. The aim is to assess soil biodiversity against current and historic land use practices both at plot and landscape scales and, further, to identify opportunities for improved sustainable land management through the introduction, management or remediation of soil biota, thus reducing the need for external inputs such as fertilizers and pesticides. Nevertheless, very little is known about individual representatives, the diversity and community composition and ecological functioning of these important organisms. For instance, soil protists are commonly lumped into a single functional unit, i.e. bacterivores. This work tackles missing knowledge gaps on soil protists and common misconceptions using multi-methodological approaches including cultivation, microcosm experiments and environmental sequencing. In a first part, several new species and genera of amoeboid protists are described showing their immense unknown diversity. In the second part, the enormous complexity of soil protists communities is highlighted using cultivation- and sequence-based approaches. In the third part, the present of diverse mycophagous and nematophagous protists are shown in functional studies on cultivated taxa and their environmental importance supported by sequence-based approaches. This work is just a start for a promising future of soil Protistology that is likely to find other important roles of these diverse organisms.

Book excerpt: The virtual impossibility of extracting the many different species from a habitat with equal efficiency by a single method (e.g. Nef, 1960). 1.1 Population estimates Population estimates can be classified into a number of different types; the most convenient classification is that adopted by Morris (1955), although he used the terms somewhat differently in a later paper (1960). 1.1.1 Absolute and related estimates The animal numbers may be expressed as a density per unit area of the ground of the habitat. Such estimates are given by nearest neighbour and related techniques (Chapter 2), marking and recapture (Chapter 3), by sampling a known fraction of the habitat (Chapter 4-6) and by removal sampling and random walk techniques (Chapter 7). Absolute population The number of animals per unit area (e.g. hectare, acre). It is almost impossible to construct a budget or to study mortality factors without the conversion of population estimates to absolute figures, for not only do insects often move from the plant to the soil at different developmental stages, but the amount of plant material is itself always changing. The importance of obtaining absolute estimates cannot be overemphasized.
