Book excerpt: Neuropeptides is one of the disciplines in the field of Neuroscience that has been largely developed from the seventies and one that is foreseen to develop to possibly unimaginable heights in the near future, since neuropeptides are widespread distributed in the central and peripheral nervous systems and they are involved in many pathologies (Parkinson s disease, Alzheimer s disease, schizophrenia, depression&). Growing numbers of researches (cell biologists, biochemists, geneticists, histologists, pharmacologists, physiologists, psychiatrists, pathologists, immunologists, neurobiologists, psychologists&) are currently involved in this discipline and increasingly sophisticated methods for its study are becoming available. Our main aims in writing this book on neuropeptides were the following: a) to review the general data that exist on neuropeptides; b) to show the results found in several research works on the distribution and functions of neuropeptides; and c) to show the future perspectives on neuropeptide research. In this sense, the text has been divided in four parts. The first entitled Introduction to Neuropeptides includes the general data on neuropeptides such as: concepts of neuropeptide, neurotransmitter, neuromodulator and neurohormone, families of neuropeptides, phylogeny and ontogeny of neuropeptides, biosynthesis of neuropeptides, release and receptors of neuropeptides, enzymatic degradation of neuropeptides, methodologies used in the study of neuropeptides and distribution of neuropeptides (Chapter 1). The second part called Distribution of Neuropeptides includes the results obtained by different research groups on the distribution of neuropeptides in the cat spinal cord, brainstem, diencephalon and cerebral cortex, as well as on the distribution of neuropeptides in the dog brainstem and diencephalon and in the alpaca brainstem (Chapters 2-7). The third part deals with the Functions of the Neuropeptides and includes a review chapter on such functions (involvement of neuropeptides in pain, analgesia, thermoregulation, glucoregulation, food and water intake, immunoregulation, wakefulness and sleep, learning and memory) (Chapter 8) and a specific chapter on the implication of neuropeptides in cardiovascular control (Chapter 9). The aim of the fourth part was to show the Pathophysiology of the Neuropeptides . Thus, there is a chapter in which the involvement of neuropeptides in several pathologies such as asthma, arthritis, Alzheimer s disease, Parkinson s disease, alcoholism, schizophrenia, depression, stress and suicide is reviewed. In such chapter, the implication of neuropeptides in aging will be also reviewed (Chapter 10). Moreover, in the fourth part three specific chapters will be focused on the involvement of neuropeptides in migraine and Parkinson s and Alzheimer s disease (Chapters 11, 12), as well as on the implications of neuropeptides and NK-1 receptor antagonists in cancer research (Chapter 13). This book is directed not only to researchers interested in the field of neuropeptides, but to Life and Health Sciences Ph.D. students. For this reason, we have included three review chapters on the general concerning of neuropeptides (Chapter 1), the functional implications of neuropeptides (Chapter 8) and on the pathophysiology of the neuropeptides (Chapter 10). We hope that this text help to students to be enthusiasm in the study of neuropeptides and to introduce them in this field, since much remains to be studied on the distribution and functions of neuropeptides. Agriculture, both animal and plant, succeeds only to the extent that arthropod and helminth consumers, vectors and pathogens can be kept at bay. Humans and their companion animals are also plagued by invertebrate parasites. The deployment of chemical agents for these purposes inevitably elicits the selection of resistant populations of the targets of control, necessitating a regular introduction of new kinds of molecules. Experience in other areas of chemotherapy has shown that a thorough understanding of the biology of disease is an essential platform upon which to build a discovery program. Unfortunately, investment of research resources into understanding the basic physiology of invertebrates as a strategy to illuminate new molecular targets for pesticide and parasiticide discovery has been scarce, and the pace of introduction of new molecules for these indications has been slowed as a result. An exciting and so far unexploited area to explore in this regard is invertebrate neuropeptide physiology. This book was assembled to focus attention on this promising field by compiling a comprehensive review of recent research on neuropeptides in arthropods and helminths, with contributions from many of the leading laboratories working on these systems.

Meanwhile, neuropsychiatry was keeping a vigilant eye on the outcome of this concerted effort with the untold hope that a genetic defect in neurotransmitter metabolism would ac count for the pathogenesis of certain psychiatric ill nesses. Thus, when neurochemists in the early sixties began to study the feasibility of measuring the metabolism of brain neurotransmitters in vivo, clinical biochemists eagerly adopted these methods to their needs and sought to verify whether inborn errors of transmitter biogenesis were a cause for at least certain forms of depression, mania and schizophrenia. Undoubtedly, it is still too early to evaluate the outcome of these studies. However, current opinion holds that gross inborn errors in transmitter metabolism do not anpear to be operative as a primary cause of psychia tric disorders. Though monoamine metabolism appears to be defective in certain groups of psychiatric disorders, the cause of these changes can at best be associated with changes in patterns of neuronal firing. It is generally believed that these persistent changes are determined by a number of unknown factors operative in various psychia tric illnesses. In the attempt to identify the molecular nature of these unknown factors, the focus of current research is directed toward transmitter receptors.

They have been implicated in the regulation of a wide range of physiological processes, including feeding-related and motivated behaviors, and also environmental adaptations. In this work, improved mass spectrometry-based analytical platforms were developed and applied to the crustacean systems to characterize signaling molecules. This dissertation begins with a review of mass spectrometry-based neuropeptide studies from both temporal- and spatial-domains. This review is then followed by several chapters detailing a few research projects related to the crustacean neuropeptidomic characterization and comparative analysis. The neuropeptidome of crayfish, Orconectes rusticus is characterized for the first time using mass spectrometry-based tools. In vivo microdialysis sampling technique offers the capability of direct sampling from extracellular space in a time-resolved manner. It is used to investigate the secreted neuropeptide and neurotransmitter content in Jonah crab, Cancer borealis, in this work. A new quantitation strategy using alternative mass spectrometry data acquisition approach is developed and applied for the first time to quantify neuropeptides. Coupling of this method with microdialysis enables the study of neuropeptide dynamics concurrent with different behaviors. Proof-of-principle experiments validating this approach have been carried out in Jonah crab, Cancer borealis to study feeding- and circadian rhythm-related neuropeptide changes using micoridialysis in a time-resolved manner. This permits a close correlation between behavioral and neurochemical changes, providing potential candidates for future validation of regulatory roles. In addition to providing spatial information, mass spectrometry imaging (MSI) technique enables the characterization of signaling molecules while preserving the temporal resolution. A novel MSI-based platform is developed by interfacing with microdialysis sample collection and is applied to the study of in vivo neuropeptide degradation profiles in an off-line ‘real-time’ fashion. This unique platform provides novel insights into neuropeptide inactivation/elimination process, which is an essential step to understand peptidegic signaling pathway. In addition to neuropeptides, this platform has been further modified to study secreted neurotransmitters, and small molecule metabolites that might interact with neuropeptides in crustacean for the first time. This work not only reports improvements upon neuropeptides identification and quantitation by developing improved analytical strategies, but also provides important evidence for the potential roles of several neuropeptides in regulating food intake and circadian rhythm behaviors. Novel platform that integrates MALDI-MSI with microdialysis in exploring neurochemical changes in dynamic biological events is also presented, which could be potentially applied to many other systems in future research. Collectively this dissertation research develops new analytical methods and improves our fundamental understanding of neuropeptide signaling.
