Schneider xi Introduction The term metabolic compartmentation, as related to the chemistry of the brain, appeared in print for the first time about 15 years ago. The concept was developed in the laboratory of Dr. Heinrich Waelsch as a result of studies related to the metabolism of glutamic acid and glutamine. It was welcomed by a number of neurochemists who felt that the concept had validity and would help explain metabolic phenomena that were otherwise quite puzzling. The concept gradually achieved general acceptance, and by 1971 the amount of information that could profit by being examined from the point of view of metabolic compartmentation had increased sufficiently to warrant a symposium on the subject ("Metabolic Compartmentation in the Brain," Balazs and Cremer, eds. , MacMillan, 1973). Almost all the participants at that initial symposium were present at the second, on which this volume is based.

Book excerpt: The NATO Advanced Study Institute held in the Hotel Metropole Llandrindod Wells, Powys, Wales from July 26th to August 4th 1981 provided an excellent platform for presentation and appraisal of our current knowledge of neurotransmitter systems. The proceed ings comprise the present volume and these were arranged to allow study of the pathways and the interactions of specific transmitter systems, one with another, in the various sub-regions of the central nervous system. In addition several of the key factors involved in neurotransmitter release were highlighted. These include the structural and molecular organisation of the nerve terminal, its organelles (such as vesicles, neurotubules and synaptic densities) and the mechanisms involved in neurotransmitter release including the participation of calcium. The organisation of transmitter synthesis, storage and transport were discussed together with their linkage to other streams of metabolism, and the cellular compartmentation of these processes. In addition to the principal lectures given at the Institute, many part1c1pants presented short reports for discussion and these are all included in this volume. The success of the Study Institute was due to the careful planning of the scientific programme to allow a smooth flow and integration of the topics included and this is reflected in the organisation of this book. Book excerpt: Synaptic transmission demands the operation of a highly specialized metabolic machinery involving the transfer of metabolites and neurotransmitters between neurons, astrocytes and microvessels. In the last years, important advances have occurred in our understanding of the mechanisms underlying cerebral activation, neuroglial coupling and the associated neurovascular response. Briefly, exacerbated oxygen consumption in stimulated neurons is thought to trigger glycolytic lactate and glucose transfer from astrocytes which, in turn, obtain these fuels from the microvasculature. Neurotransmitter release is made possible by a combination of transcellular cycles exchanging metabolites between these three compartments, returning eventually the synapsis to its pre-firing situation in the resting periods. In spite of the enormous progresses achieved in recent years, the drivers determining the predominant direction of the fluxes, their quantitative contribution and their energy requirements, have remained until today incompletely understood, more particularly under the circumstances prevailing in vivo. In many instances, progress derived from the implementation of novel methodological approaches including advanced neuroimaging and neurospectroscopy methods. Book excerpt: The concept of chemical transmission in the central nervous system has taken some time to be generally accepted, but an increasing number of compounds are now being recognized as hav ing a transmitter role in the brain. The acetylcholine system was the first to be discovered in the periphery and its charac teristic features of storage of transmitter in vesicles in the nerve terminal, its electrically-evoked release and rapid extra neuronal breakdown were considered to be necessary criteria for any neurotransmitter candidate. The subsequent elucidation of the noradrenergic system made it apparent that rapid enzymatic breakdown was not essential for a released transmitter, and the possibility of high-affinity re-uptake processes became establ ished as an alternative means of terminating the synaptic actions of a transmitter. With the eventual acceptance of the amino acids as excitat ory or inhibitory transmitters, the requirement for a transmit ter to be present in a low concentration overall (although locally concentrated in specific terminals) also had to be discarded. This necessitated the additional concept of specif ic metabolic pools with different functions being located in different cells or within different regions of the same cell. Some localization of glutamate and aspartate remote from excit able membranes is clearly essential since their overall brain concentrations would be sufficient to maximally depolarize the majority of neurones in the brain. The concept of separate metabolic pools has been supported by stUdies on turnover rate (see Chapter 5).

As a sign of the interest and success of the partici pants I can mention that all participants have sent in their contributions. Perhaps editors of volumes would anpreciate that the very last came in no later than two months past the deadline.
