Book excerpt: Maternal programming can have numerous detrimental effects on offspring development, growth, and health. To improve livestock production efficiency, it is necessary to determine the mechanisms that can result in reduced growth, health, and product quality. We hypothesized that 1) restricted- and over-feeding during gestation would inhibit offspring muscle development, and 2) high maternal milk production would affect factors involved in offspring growth and immunity. To investigate the effects of poor maternal nutrition on offspring muscle development and growth, pregnant Western White-faced ewes (n = 82) were randomly assigned to one of three diets at day 30 of gestation through parturition; a control- (100% NRC requirements; CON), restricted- (60% NRC; RES), or over-fed (140%; OVER) diet. At day 45, 90, and 135 of gestation and within 24 hours of birth, ewes and fetuses were euthanized for collection of offspring longissimus dorsi, semitendinosus, and triceps brachii (n = 10 to 15 fetuses per treatment per time point). Muscle sections were immunostained with Pax7 antibody and data were analyzed using PROC MIXED in SAS. An interaction of maternal diet by time point was observed in the semitendinosus (ST) and triceps brachii (TB), where Pax7(+) cells were decreased in RES offspring at day 45, 90, and 135 in the ST (P ≤ 0.02) and day 90 in the TB (P ≤ 0.002). Within OVER offspring, Pax7(+) cells were decreased at day 90 in the ST (P ≤ 0.02) and day 45 and 90 in the TB (P ≤ 0.04). No effect of maternal diet by time point was observed in the LD (P = 0.57). An interaction of maternal diet by litter size was also observed, where singletons and twins had reduced Pax7(+) cells in the ST and TB (P ≤ 0.02) but triplets had an increased percentage of Pax7(+) cells in the LD (P ≤ 0.04). In conclusion, maternal restricted- and over-feeding negatively affects prenatal muscle development over time but these effects are muscle specific. To investigate the effects of maternal milk production during gestation on calf growth and immunity, morphometric measurements and blood samples were obtained from dairy calves born to high (average lactation of 14,865 kg of milk; n = 17) or low (average lactation of 10,069 kg of milk; n = 18) milk producing cows within 24 hours of birth. Offspring will be referred to as HIGH or LOW, respectively. Blood samples were analyzed to determine concentrations of insulin, glucose, haptoglobin, interferon gamma, and other biochemical factors relating to growth and immunity. An effect of maternal lactation by gender was observed for skull width (P = 0.09), nose-occipital length (P = 0.03), glucose (P = 0.05), direct bilirubin (P = 0.07), calcium (P = 0.08), and magnesium (P = 0.07). Additionally, an effect of lactation was observed for total protein and globulin (P ≤ 0.01). No effect of lactation by gender, lactation, or gender was observed for insulin (P = 0.53), serum IgG (P = 0.23), colostrum IgG (P =32), or haptoglobin (P = 0.24). In conclusion, maternal lactation status can affect several factors relating to offspring growth and health. Future studies are required to further assess the effects of maternal programming on offspring development, growth, and health to improve current livestock production.

We used two rodent cohorts to model impacts of maternal diet on offspring microbiota composition and metabolic health. 1) Sprague-Dawley dams consumed high fat (HF) or chow diets during pregnancy and offspring were raised by dams consuming the same or opposite diet; fecal samples were collected for microbiota analysis on postnatal day (PND) 21. 2) C57BL/6 dams received low fat (LF), HF, or HF supplemented with glycan (HFG) diets; offspring were weaned onto the HF or LF diet. Offspring fecal samples were collected at PND21, PND35, and PND90. The cross-fostering study indicates postnatal, rather than prenatal, influence drives early microbiota composition, while the C57BL/6 study suggests maternal and post-weaning diets shape microbiota composition in adult offspring.

Increasing obesity rates are particularly troubling because much evidence has been reported that maternal diet before and during pregnancy can increase the likelihood of obesity in the next generation, through epigenetic and obesity-related developmental changes. Extremely low fat maternal diets have also been shown to have transgenerational effects on health, including evidence for a "thrifty phenotype" hypothesis that supposes the intrauterine conditions prepare the offspring for the outside world based on the nutritional environment available during fetal development; therefore, offspring exposed to low fat intrauterine environments but much higher fat diets outside of this environment are more at risk for obesity and other metabolic dysfunction. In order to test the transgenerational relationship between maternal diet and offspring health, Danio rerio females were fed either a high fat diet (HFD), control diet, or low fat diet (LFD) for several months and crossed with males on the control diet. This study reports that significant differences in offspring early motor behaviour were observed were observed in offspring sired from HFD and LFD mothers, both from the control and from each other. These results indicate possible effects on skeletal muscle and neural development. Significant differences in appetite were also observed between control and HFD, and between HFD and LFD. These results may indicate changes in the secretion of satiety-related hormones in these offspring. HFD and LFD females also showed evidence of decreased fertility and increased offspring mortality rates. These data indicate that maternal diet before and during pregnancy may have far reaching effects on the development and general health of the offspring.

One-carbon nutrients involved in epigenetic processes represent a possible underlying mechanism. Unfortunately, there is limited research in humans to support animal findings, and the few studies available have been limited to birth defects and weight outcomes in early infancy. The aim of this project was to examine the associations between maternal diet (diet quality indicators and one-carbon nutrients) during the period surrounding pregnancy and long-term weight outcomes in the offspring.Methods: We examined 2,729 mother-child pairs from the Nursesu2019 Health Study II and the offspring cohort Growing Up Today Study 2 (GUTS 2). Children, 12-14 years at baseline were 21-23 years at the last follow-up. Overweight was defined for youth less than 18 years of age using sex- and age-specific body mass index (BMI) cut-offs recommended by the International Obesity Task Force and for those 18 years or older as BMI u226525 kg/m2. Dietary information was collected by a validated 131-item food frequency questionnaire (FFQ) and questions on supplement use. Maternal dietary patterns were calculated from FFQs using three diet quality scores u2013 the alternate Healthy Eating Index (aHEI), Alternate Mediterranean Diet (aMED) and Dietary Approach to Stop Hypertension (DASH). Log-binomial models were used to estimate relative risks (RRs) and 95% confidence intervals.Results: In models adjusted for sex, gestational age at delivery and maternal total energy intake, greater maternal adherence to aMED and DASH, but not aHEI, was associated with lower overweight risk in the offspring (RRQ5 vs Q1=0.82 [0.70-0.97] for aMED and 0.86 [0.72-1.04] for DASH) (P for trend

Book excerpt: To date, limited evidence suggests that maternal preconception and pregnancy dietary patterns are associated with reduced risks of adverse maternal and child health outcomes such as gestational diabetes mellitus and preterm birth. However, there are insufficient studies examining preconception and pregnancy dietary patterns on birth weight outcomes such as gestational-age birth weight for any conclusion to be made. Additionally, there is a relative paucity of studies investigating preconception, postpartum, and beyond postpartum dietary patterns. Recent research has advanced from examining each modifiable lifestyle factor singly (e.g. diet, physical activity, smoking) towards a holistic approach of examining multiple lifestyle factors using composite healthy lifestyle scores. Two large-scale mother-offspring cohorts in Ireland and the United States have shown that adherence to a healthy lifestyle during pregnancy and beyond pregnancy is associated with reduced risks of adverse offspring birth outcomes and childhood obesity. These findings are consistent with data obtained from cross-sectional studies based in China and Tanzania.

SACN concludes that there is cause for concern about the later health consequences of compromised or excessive nutrient supply during early fetal and infant life. It notes that in the context of reproduction, the impact of current dietary patterns on women and girls is of particular concern, and considers that improving the nutritional status of women of childbearing age, infants and young children has the potential to improve the health of future generations. The Committee also offers recommendations for future research in this area, particularly emphasising the need for data which better characterise dietary patterns and patterns of pre and postnatal growth.
