In fact, mitochondria biological activities have progressively expanded to include not only various bioenergetic processes but also important biosynthetic pathways, calcium homeostasis and thermogenesis, cell death by apoptosis, several different signal transduction pathways mainly related to redox control of gene expression and so on. This functional and structural complexity may undergo important derangements so to justify the definition of ‘mitochondrial medicine’, which should include all the clinical consequences of congenital or acquired mitochondrial dysfunctions. There are actually a growing number of studies which assign a significant pathogenic role to damaged mitochondria in different diseases: ischemia/reperfusion injury, neurodegenerative diseases, cancer with its dramatic sequelae (i.e, metastasis), metabolic syndrome, hyperlipidemias, just to mention a few of the most important pathologies. In this context, a further aspect that should not be disregarded is the interaction of pharmacological agents with mitochondria, not only in regard of the toxicological aspects but, above all, of the potential therapeutic applications. In fact, it is interesting to note that, while the properties of different so-called “mitoxicants” are well-known, the subtle linkages between drugs and mitochondria is still in need of a real pharmacological and therapeutic control at the clinical level. This lack of consideration can often lead to an underestimation of unwanted toxic effects but also of desirable therapeutic activities. A reevaluation of the potential clinical role of mitochondria could give a new light on some yet obscure aspects of human pathophysiology.

Life-long nutrition studies are not possible in humans because of their long survival. The answer is to reduce our calorie intake, improve our diet, and exercise more. But calorie restriction is extremely difficult to maintain for long periods. How then can we solve this problem? Adult C. elegans are reproductive for about the rst week of adulthood followed by approximately two weeks of post-reproductive adulthood prior to death. Life span is most commonly measured in the laboratory by maintaining the worms on the surface of a nutrie- agar medium (Nematode Growth Medium, NGM) with E. coli OP50 as the bacterial food source (REF). Alternative culture conditions have been described in liquid media; however, these are not widely used for longevity studies. Longevity of the commonly used wild type C. elegans hermaphrodite (N2) varies ? from 16 to 23 days under standard laboratory conditions (20 C, NGM agar, E. coli OP50 food source). Life span can be increased by maintaining animals at lower ambient temperatures and shortened by raising the ambient temperature. Use of a killed bacterial food source, rather than live E. coli, increases lifespan by 2–4 days, and growth of adult animals in the absence of bacteria (axenic growth or bac- rial deprivation) increases median life span to 32–38 days [3, 23, 24]. Under both standard laboratory conditions and bacterial deprivation conditions, wild-derived C. elegans hermaphrodites exhibit longevity comparable to N2 animals [25].
