After about 1930, when it became possible to examine the dynamic aspects of a cell seriously and to increase the breadth of our knowledge of the chemicals which are involved, there was a feeling that the elucidation of a pathway or the use to which a compound or a process was put biologically had a ubiquitous character. Among the reasons for believing this was the constancy of the amino acid building blocks for proteins. Not only were the same general structures found regardless of organismic type, but the stereochemistry was the same. This sort of observation led to the idea of the "unity of nature. " Few people, of course, thought a complete unity existed, because it was already known in the latter part of the nineteenth century that the sterols, the polysaccharides, and the pigments of various organisms could be substantially different, but only recently, during the last decade or so, has the fuH scope ofthe difference-as weil as ofthe similarity-begun to emerge. Of particular importance, it has now become evident that a large hiatus exists between some types of organism. Even more important, perhaps, and certainly more unexpected, are the substantial gaps which have been discovered within otherwise similar organisms. The evolutionary process is presumably behind what we observe, and therefore an understanding of the rules and regulations which govern similarity and difference should eventually lead to an elucidation of evolution itself.

However, such extractions yield a complex mixture of different lipid classes which have to be purified further for quantitative analysis. Moreover, the crude lipid extract will be contami nated by other hydrophobic molecules, e.g. by intrinsic membrane proteins. Of the various types of separation processes, thin layer and column chromatography are most useful for intact lipids. High performance liquid chromatography (HPLC) is also rapidly becoming more popular, especially for the fractionation of molecular species of a given lipid class. The most powerful tool for quantitation of the majority of lipids is gas liquid chromatography (GLC). The method is very sensitive and, if adapted with capillary columns, can provide information with regard to such subtle features as the position or configuration of substitutions along acyl chains. By coupling GLC or HPLC to a radioactivity detector, then the techniques are also very useful for metabolic measurements. Although research laboratories use generally sophisticated analytical methods such as GLC to analyse and quantify lipid samples, chemical derivatie:ations are often used in hospitals. For these methods, the lipid samples are derivatized to yield a product which can be measured simply and accurately-usually by colour. Thus, total triacylglycerol, cholesterol or phospholipid-phosphorus can be quantitated conveniently without bothering with the extra information of molecular species, etc. which might be determined by more thorough analyses. These include the involvement of phosphatidylcholine in biological membranes, as a component of plasma lipoprotein, as a transporter of choline in the body, and also as a "reserve", and possibly only source of unesterified choline. Moreover, numerous clinical studies have recently been conducted, to evaluate the possible uses of externally supplied lecithin in the treatment of some intractible neuropsychiatric disorders (e.g. tardive dyski.nesia, Alzheimer's disease, etc.) and other conditions (e.g. hypercholesteremia). Results to date are encouraging, yet equivocal. This is due, in part, to the fact that the field of phospholipid methodology is highly complex. There is much confusion in the literature, and many ambiguities still remain in the interpretation of experimental findings. This is particularly so for phenomena involving phospholipid function in the central nervous system. This book incorporates the proceedings of the Fourth International Colloquium on Lecithin, which took place in Chicago, Illinois, USA, on September 15-17, 1986. The purpese of this colloquium was to review, in a comprehensive manner, basic principles as weli as current information about the technology, biochemistry, physiology and therapeutic potential of lecithin. Over 108 individuals from all over the world participated in the sessions. The meeting was subdivided into oral presentations and panel discussions (see Table of Contents). It was sponsored and financed by the Lucas Meyer GmbH (Hamburg), for which the editors are most grateful.
