Book excerpt: Understanding a species' hierarchical genetic variation may provide insights into its ability to cope with current and future stressors, such as climate change, exploitation, and degradation of habitat. Analyzing the events that led to speciation and population diversification may give a framework for predicting responses and adaptations to future changes, imparting critical information for conservation managers. I resolve the evolutionary history and population genetic patterns of the walleye Sander vitreus using molecular and morphological characters and a variety of analyses, including sequences from three nuclear and three mitochondrial (mt) DNA regions and nine nuclear DNA microsatellite (μsat) loci. Results show that walleye and its sister species the sauger S. canadensis diverged ~15.4 Mya (5.2-27.1 Mya = 95% highest posterior density (HPD)), with modern walleye haplotypes differentiating ~10.6 Mya (6.9-14.3 HPD), when climate changes were occurring across North America. Walleye exhibit significant genetic structure (mean FST mtDNA control region=0.24, μsat=0.10) and substantial genetic diversity (mean control region=0.53, μsat=0.68) across its range, at broad- and fine-scales. Contemporary patterns correspond to a genetic isolation by geographic distance hypothesis, with northern populations reflecting differential contribution from three distinct Pleistocene glacial refugia. Comparisons of historic versus modern samples from Lake Erie reveal that allelic frequencies have changed over ~70 years (mean FST control region=0.26, μsat=0.17), with modern populations having higher diversity (mean control region=0.67, μsat=0.72 vs. control region=0.05, μsat=0.47). Today's higher diversity may be due to population rebounds following closure of the walleye fishery in 1970. Paratypes of the historic blue pike did not differ morphologically or genetically from walleye, indicating that the blue pike was not a separate taxon. Additional fine-scale results within the highly degraded Huron-Erie Corridor reveal the seven spawning groups show a mixture of connectivity and divergence. This study also discerns the effects of habitat augmentation on the genetic structure of walleye spawning groups, finding that populations are distinct and have similar levels of genetic diversity (mean control region=0.73, μsat=0.72). Some genetic exchange occurred at an augmentation site, suggesting that walleye from other locations arrived to spawn on the newly available habitat. Such migration could change the genetic composition of the original spawning group. The results of this dissertation provide insight to the processes that shaped the evolution and population genetic patterns of today's walleye, including past climate change, Pleistocene glaciations, and anthropogenic stressors. Future research identifying the adaptations underlying the genetic diversity and divergence patterns discerned here and their respective genetic contributions to fitness will aid efforts to sustain natural populations in the face of ongoing climate change and other anthropogenic stressors.

Book excerpt: Many migratory animals time reproduction using environmental cues (e.g., photoperiod) that signal favorable conditions for offspring (e.g., food availability). However, environmental change can decouple these cues from the conditions relevant to offspring success. In turn, variation in reproductive behaviors may be critical to population persistence in a changing environment. For example, while natal homing can lead to valuable local adaptations in a stable environment, straying to new breeding sites can buffer against environmental change. Among exploited species, knowledge of reproductive behavior, such as spawn timing (phenology) and spawning site selection, has proven useful to fisheries management by influencing angling dynamics, estimates of biological reference points, and stock delineations. Thus, a well-developed understanding of reproductive phenology, breeding site selection, and fishery dynamics could help agencies effectively conserve and manage fisheries in ecosystems experiencing environmental change. Accordingly, my dissertation research sought to better understand the reproductive behaviors and angling dynamics of Lake Erie’s walleye population. In collaboration with other researchers and agency biologists, I used acoustic telemetry, creel surveys, environmental monitoring data, and quantitative techniques to discern the spawning behaviors and angling dynamics of this population. We assessed the timing of residency in spawning sites, focusing on how environmental cues and individual characteristics drive observed variation (Chapter 2), developed a quantitative framework to estimate the frequencies of each potential spawning site selection strategy (Chapter 3), demonstrated sex based-differences in spawning behavior and male-biased walleye harvest during the spawning season (Chapter 4), and modeled the impact of fishery characteristics and angler behaviors on recreational catch outcomes (Chapter 5). Each chapter generated new insights to the benefit of fisheries management through advances in our understanding of reproductive ecology and angling dynamics. In Chapter 2, we found that the timing of residency in spawning sites differed between sexes and among spawning locations and was controlled by photoperiod and temperature. The varying responses to warming suggest that reproductive resilience is promoted by conserving local spawning populations and a broad age structure. The findings from Chapter 3 demonstrated that, although spawning site fidelity and annual spawning are common, apparent skipped spawning also occurs (~ 14% of observations). Accounting for this potential skipped spawning led to revised biological reference points, owing to adjusted estimates of spawning stock biomass. Additionally, although few walleye strayed, the pattern of straying indicated that some stocks mix heavily. In Chapter 4, we found strong evidence for sex-specific spawning behaviors, with males occupying spawning grounds longer than females, which helped explain male-biased walleye harvest on the spawning grounds. Lastly, in Chapter 5, modeling creel survey data showed that angler decisions influenced catch outcomes more than abundance of the targeted walleye population. The findings from this chapter can help anglers maximize their probability of fishing success, given desired catch outcomes. The improved understanding of Lake Erie walleye reproductive behavior, angling dynamics, and the relationships between them, should help agencies worldwide keep their valued fish populations sustainable in the face of continued environmental change.

Book excerpt: The population genetic structure of three of the largest walleye spawning groups in Lake Erie is tested for consistency over time and space spanning 14 years, based on nine high-resolution nuclear DNA microsatellite loci. Previous genetic studies focused on a one-time genetic "snapshot", with an earlier study by our laboratory finding that the genetic structure of three Lake Erie spawning groups along the southern shore - Maumee River, Sandusky River, and Van Buren Bay reefs - appeared similar in 2003, whereas most other spawning groups across Lake Erie were genetically distinctive. The present study analyzes the stability of genetic similarity patterns within and among 726 walleye spawning at these three sites across years and age cohorts in 1995, 1998, 2003, 2007, and 2008. Genetic patterns are evaluated using pairwise FST analog and contingency tests, AMOVA partitioning, and Bayesian assignment tests. Results reveal overall year-to-year consistency in genetic structure of walleye spawning at the three sites, with some annual variation in the Van Buren Bay reef group. Greater genetic divergence from the other groups is detected in the Van Buren Bay spawning group, which reflects greater geographic separation. Walleye spawning in the Sandusky and Maumee Rivers are genetically distinguishable from each other when data from all years are combined, which suggests possible sample size effect (i.e., annual sample sizes likely were not large enough to detect their genetic differentiation). No significant differences occur among age cohorts, between the sexes, or among sampling dates within spawning runs. Results demonstrate the importance of sampling over several years of walleye spawning runs in order to resolve fine-scale genetic relationships within an open lake system.

However, since experiential legacies are difficult to measure in populations, how experiential legacies of individuals affect cohort- and population-level outcomes (i.e., buffering or amplifying population responses) remains difficult to discern. The objective of my dissertation research is to evaluate the extent to which experiential legacies affect individual performance and alter population dynamics. By exploring the importance of individual life events to populations, we can better understand the interconnectedness of life stages and better anticipate how environmental change may alter population and community dynamics. Specifically, I examined individual experiential legacies across a range of animal species with the goal of identifying generalizable patterns in response to early-life nutritional stress (Chapter 2), and then I focused on experiential legacies within individuals (Chapter 3), cohorts of individuals (Chapter 4), and a population consisting of multiple cohorts (Chapter 5) in Lake Erie Walleye (Sander vitreus). Lake Erie Walleye present a population for which understanding the long-term impacts of early-life experiences may be particularly valuable due to current human-induced environmental changes within its ecosystem. Since experiential legacies can produce unanticipated changes in the kinds as well as proportions of subsequent (i.e., later in life) phenotypes, examining patterns across multiple species may expose underlying trends. Patterns in experiential legacies across 81 studies of 65 animal species demonstrated generally consistent negative or neutral impacts of early nutritional stress on later-life phenotypes, indicative of energy depletion as a mechanism for the long-term consequences of early-life conditions (Chapter 2). Yet, overall, my results emphasize the existence of complicated interactions among a suite of phenotypic responses in determining individual performance. Within Lake Erie Walleye, I found evidence of experiential legacies from early-life experiences using laboratory experiments, but I also observed indications of the strong influence of maternal legacies and of carryover effects from more recent experiences using long-term data on cohorts. Nutritional quality of food during early life (a 10-d period starting when Walleye could first feed) was positively correlated with Walleye juvenile sizes and this correlation with size continued even after all treatments were fed on a high-quality standardized diet for an additional 27-d (Chapter 3). Beyond the period examined in my laboratory experiments, though, in an analysis of field data, I found that maternal effects were more influential than sizes or densities achieved during the first few months of life to annual growth in Walleye cohorts at ages 3-5 (Chapter 4). Thus, early-life experiences can produce experiential legacies in Lake Erie Walleye, but those experiences may be overwhelmed by the lingering influence of other factors such as maternal effects. Additionally, at the cohort-level, growth in the previous year negatively affected recent growth at ages 3-5, which may be indicative of compensatory growth in Walleye and could reduce variation among cohorts in size-at-age over a longer period of time. At the population-level, I modeled how specific experiential legacies may be more or less beneficial under different environmental conditions for Lake Erie Walleye, which demonstrates characteristics of a periodic life history (i.e., high fecundity, low early-life survival, old age at maturity), as well as for populations representing equilibrium (low fecundity, low early-life survival, old age at maturity) and opportunistic (high fecundity, low early-life survival, young age at maturity) life histories (Chapter 5). Across experiential legacies and these three life history strategies, early-life environments were primarily responsible for differences in simulated population growth rates and demography, with more frequent “good” early-life environments increasing population growth rates, increasing variation in population growth rates, and decreasing the proportion of older ages in the populations. However, when early-life environments were likely to be good and later-life environments were likely to be poor, experiential legacies that create lifetime trajectories (i.e., early-life conditions establish phenotypes that persist throughout life) were beneficial to population growth for all life histories. While other modeled experiential legacies did not demonstrate any specific benefit to Walleye, experiential legacies that create environmental specialization (i.e., later-life phenotypes perform best when early-life and later-life environments are similar) and later stressor resilience (i.e., poor early-life environments allow later-life phenotypes to perform well in poor environments) were beneficial for the equilibrium population when early-life environments were frequently poor, potentially due to the importance of adult survival for the equilibrium population. Overall, these simulations indicated that variation in experiential legacies across populations could be due to combinations of life history characteristics and frequencies of specific environmental conditions in early and later life. My results at the individual, cohort, and population level demonstrate how exploring experiential legacies can provide a deeper understanding of population patterns. Across species, experiential legacies may be related to how energy is allocated in early life (Chapter 2). Within Lake Erie Walleye, I observed that early-life nutritional conditions continued to affect juvenile Walleye sizes after nutritional conditions became standardized, supporting the idea of early-life energy allocation driving later performance (Chapter 3). Despite these results in young Walleye, the early-life growth environment did not appear to be the most important factor driving later cohort-based Walleye growth, potentially due to compensatory growth maintaining stable size-at-age and maternal provisioning legacies (Chapter 4). Regardless of the experiential legacy that Lake Erie Walleye or other species experience, the early-life environment appears to be extremely influential in driving population growth; however, certain experiential legacies may be more common in specific environmental scenarios for species with specific life histories due to the potential advantages those experiential legacies provide (Chapter 5). While many questions remain, my research has improved our understanding of patterns and implications of experiential legacies in general, as well as the degree to which legacies of early life influence the response of Lake Erie Walleye to its environment.

Book excerpt: Abstract: To allow use of otolith microchemistry in determining natal origins of freshwater fish, we must understand how quickly environmental trace elements leave their signatures in larval otoliths. This experiment was designed to determine at what point in the early life history of the walleye (Sander vitreus) a chemical signal is first recorded and whether uptake rates are rapid enough to record a natal site signal. In April 2008, eggs and sperm from spawners in the Maumee River, Ohio were transported to our laboratory at the Ohio State University for fertilization. Fertilized eggs were incubated in McDonald style jars with water spiked with different concentrations of strontium. At hatch, larvae were randomly transferred into tanks with an experimental design of two replicate tanks for each of the three levels of strontium concentration (300 ug/L, 900 ug/L, and 1500 ug/L) crossed with two temperatures (8°C and 13°C) that represented a range of expected conditions in Lake Erie and its tributaries. Egg samples were taken 2 and 4 days before hatch and on the day of hatch. Larval samples were taken every other day, beginning with day of hatch, for 20 days. Otoliths were removed and processed with a combination of laser ablation and solution based inductively coupled plasma mass spectrometry. Initial results show that by 20 days post-hatch, larval otoliths show a treatment-related strontium signal. We are continuing analyses to identify the earliest point at which we can distinguish strontium treatments.

Book excerpt: As sequel to Aquatic Animal Nutrition – A Mechanistic Perspective from Individuals to Generations, the present treatise on organic macro- and micronutrients continues the unique cross fertilization of aquatic ecology/ecophysiology and aquaculture. This treatise considers proteins and their constituents, carbohydrates from mono- to polysaccharides, fatty acids from free acids to fat, and waxes. It becomes obvious that these organic nutrients are more than only simple fuel for the metabolism of animals; rather, their constituents have messenger and controlling function for the actual consuming individual and even for succeeding generations. This aspect will become particularly clear by putting the organisms under consideration back into their ecosystem with their interrelationships and interdependencies. Furthermore, micronutrients, such as vitamins and nucleotides as well as exogenous enzymes, are in the focus of this volume with known and still-to-be-discovered controlling physiological and biomolecular functions. Aquatic Animal Nutrition – Organic Macro and Micro Nutrients addresses several gaps in nutritional research and practice. One major gap is the lack of common research standards and protocols for nutritional studies so that virtually incomparable approaches have to be compared. This applies also to the studied animals, since most approaches disregard intraspecific variabilities and the existence of epimutations in farmed individuals. Furthermore, recalling the Mechanistic Perspective from Individuals to Generations, dietary benefits and deficiencies have effects on succeeding generations. In most studies, this long-term and sustainable aspect is overruled by pure short-term production aspects. By comparing nutritional behavior and success of fishes and invertebrates, Aquatic Animal Nutrition points out different metabolic pathways in these animal groups and discusses how, for instance, fishes would benefit when having some successful metabolic pathway of invertebrates. Application of novel genetic techniques will help turn this vision into reality. However, a widely missing link in the current nutritional research is epigenetics regarding transgenerational heritages of acquired morphological and physiological properties. To increase public acceptance, nutritional optimization of farmed animals based on this mechanism, rather than genetical engineering, appears promising.
