Available in PDF, EPUB and Kindle. Book excerpt: PH and ion-selective microelectrodes are rapidly finding an increasing number of applications in the study and control of living (and nonliving) systems. They are unique in their capacity to measure chemical species without altering natural or controlled environmental conditions. Furthermore, these potentiometric tools measure the activity of the chemical species in contrast to conventional ones that measure total concentration. The "Workshop on IonueSelective Microelectrodes" is designed to provide an insight into the principles, theory, fabrication, techniques, present limitations, goals, and applications of some of these tools. The importance and types of microelectrodes and guidelines for their application in biological systems are discussed by Berman. Their present limitations are reviewed by Durst. He warns that their use in analyzing living matter should be approached with caution because of the ill-defined nature of biologic systems. Techniques are presented next for the fabrication of pH (Hebert), antimony (Green and Giebisch, and Malnic et al.), oxygen (Whalen), then single-barrelled (Wright, Walker and Ladle, Morris and rnjevic) and double barrelled (Zeuthen et al., and Khuri) potassium and chloride liquid ion-exchanger microelectrodes. Difficulties with and fabrication of reference and glucose electrodes are covered, respectively, by Durst and Wright, and Bessman and Schultz. Applications of pH and ion-selective microelectrodes are described in microanalysis (Wright), measurement of intracellular ion activity and calculation of equilibrium potentials (Brown and Kunze), and then studies of the kidney (Wright, Malnic et al., and Khuri), brain (Zeuthen et al., Morris and Krnjevic), frog heart (Walker and Ladle), and human skeletal muscle (Filler and Das). In addition, actual and potential clinical applications, respectively, of pH (Filler and Das) and glucose (Bessman and Schultz) electrodes are reviewed.

It consists of eleven invited review papers and selected sidcussions from the Symposium on Ion-Selective Electrodes held at National Bureau of Standards on January 30-31, 1969. The publication provides a comprehensive survey of the field and should prove very valuable in advancing the state of the art. The chapters include discussions on the theory, characteristics, and methodology of all types of ion-selective electrodes; their use in thermodynamic, kinetic, and complex-ion studies; standards; pure and applied research in various biomedical areas of interest; industrial analysis and control systems; and applications to diverse analytical problems. Both the present status and future potential of these sensors are discussed in a wide range of scientific disciplines, from the physical sciences to biomedicine. Extensive bibliographies provide an excellent survey of the literature for anyone using or anticipating the use of these electrodes. (Author).

Book excerpt: We continue in this second volume the plan evident in the first; i.e., of presenting a number of well-rounded up-to-date reviews of important developments in the exciting field of ion-selective electrodes in analytical chemistry. In this volume, in addition to the exciting applications of ISE'S to biochemistry systems represented by the description of enzyme electrodes, there is featured the most recent development in ISE'S, namely, the joining of the electrochemical and solid state expertise, resulting in CHEMFETS. The scholarly survey of the current status of ISE'S will undoubtedly be welcomed by all workers in the field. Book excerpt: Membrane Electrodes considers the significant developments in the field of sensing probes, with an emphasis on membrane electrodes. This book is organized into three parts encompassing 11 chapters. Part I is an introduction to the variety of ion-selective membrane electrodes that have been constructed and with which experiments have been conducted. This part deals first with the thermodynamic principles and other concepts underlying the description of the behavior of electrolyte solutions, followed by a discussion on the various theories of membrane potential applicable to a variety of solid and liquid membrane electrodes. Part II describes the preparation, properties, and uses of the various solid and liquid membrane electrodes. Part III presents glass membrane electrodes as a prelude to the description of other membrane systems in which glass electrodes are invariably used as the primary sensing device. This book will prove useful to students, technologists, and researchers in various fields of science and technology.

Berson in 1959 opens up a new avenue in ultra sensitive analysis of trace substances in complex biological systems. In recognition of the enormous contributions of RIA to basic research in biology and to routine clinical tests in laboratory medicine, R.S. Yalow, the co-developer of RIA, was awarded, in 1977, the Nobel Prize for Medicine and Physiology. The basic principle of RIA is elegantly simple. It is based on a specific, competitive binding reaction between the analyte and the radio-labeled analog of the analyte for the specific antibody raised to the analyte. The combination of high specificity and affinity of an antibody molecule makes it a very versatile analytical reagent capable of reacting specifically with analytes at a very low concentration in a complex solution such as serum. The sensitivity of RIA is provided by using a radioactive tracer.

Book excerpt: New methods of locally delivering chemicals to biological systems have emerged in recent years, but nearly all are limited by their reliance on finite capacities of therapeutic stock which require physical access for restoration or replacement. In this thesis, I examine the ion-selective membrane (ISM), a filter that discriminately transports single ions within mixed electrolytes, whose operation in ion delivery platforms does not suffer from the fundamental restriction of finite capacity. Specifically, I discuss the optimal design of the ISM and its operating conditions, as well as its implementation for peripheral neuro-modulation. Up to this point, limitations in available characterization methods, both theoretical and practical, have severely restricted its technical development and the exploration of in vivo applications. In this thesis, I (1) began by revisiting the operation of ISM systems from a first-principles perspective, constructing a highly detailed physicochemical transport model to describe intra-membrane processes, (2) developed a new experimental technique for direct, real-time monitoring of ion concentration polarization which allowed me to evaluate those processes under relevant conditions, (3) established a rapid-manufacturing process for fabricating nerve cuff electrodes compatible with ISM coating and tested it in vivo, and (4) built off this foundational work to fabricate a Ca2-selective nerve cuff electrode suitable for electrochemical operations, implanted in vivo on the sciatic nerve of a rat. I discovered over-limiting phenomena that arise in ISM systems under electrical polarization, which have broad implications for all of its ion filtering applications. Finally, from my electrophysiological studies, I found evidence that Ca2+ depletion with the nerve cuff had a discriminatory effect on distinct fascicles within the rat sciatic nerve, potentially offering a new strategy for the selective activation of therapeutic targets.
