Book excerpt: Le but d'une opération de mélange de poudres est d'atteindre l'homogénéité, c'est-à-dire une situation où tous les constituants sont distribués de façon uniforme dans le mélange. Les moyens utilisés actuellement pour évaluer l'efficacité de cette étape, ne sont pas satisfaisants, à cause du biais qu'ils apportent. Par conséquent, une méthode alternative se développe dans l'industrie pharmaceutique, sous l'impulsion de la Food and drug administration : la démarche "Process analytical technology", qui permet de suivre en ligne et en temps réel le mélange. Plusieurs technologies peuvent être utilisées pour effectuer ce suivi, dont la spectroscopie proche infra rouge (PIR). Nous avons testé deux spectromètres, Acousto-Optic Tunable Filter et à barrette de diodes, permettant d'acquérir des spectres PIR caractéristiques de l'état du mélange. Nous avons comparé ces deux matériels et évalué le point d'arrêt sur un mélange avec une faible concentration en principe actif. Au contraire, l'étude d'un mélange avec deux constituants de structure chimique proche n'a pas permis de déterminer l'atteinte de l'homogénéité. Enfin, le suivi d'un mélange non critique a montré que la combinaison de différents traitements appliqués sur les spectres obtenus, semblait la solution la plus intéressante pour connaître le point d'arrêt. Une étude approfondie permettrait d'évaluer la significativité des résultats obtenus.

De nos jours, les procédés de fabrication comportant des étapes discontinues connaissent des limites en termes de productivité et de qualité. Il est donc nécessaire de faire appel à des procédés innovants. Afin de démontrer la conformité et l'homogénéité permanente de la poudre tout au long du procédé de fabrication par ce nouveau système, il est indispensable de réaliser des étapes de calibrations et d'essais afin de s'assurer d'un résultat final de qualité. L'objectif est donc d'améliorer la capabilité de l'humidité résiduelle des produits d'un atelier de granulation humide. Pour cela, nous nous sommes appuyée sur la méthodologie Lean Six Sigma pour réaliser ce projet. Cette mise en place rentre également dans une démarche de Quality by Design, puisque le but est d'explorer les caractéristiques de la poudre, de mieux connaître le procédé de séchage en l'explorant aux limites et en identifiant ses paramètres critiques.

Book excerpt: La démarche PAT a été initiée dans les années 2000 par la FDA auprès des industries et des laboratoires pharmaceutiques. Son objectif est de mettre en place des outils efficaces et performants pour contrôler et optimiser les procédés de production. La spectroscopie proche infrarouge est un outil analytique particulièrement approprié pour la mise en place du PAT. Elle permet de réaliser des analyses rapides, non destructives, sans préparation et sans contact. Cette étude menée au sein de l'entreprise Merial a pour objectif de montrer que la SPIR peut être une méthode alternative à la CLHP pour quantifier des substances en solution. Ce mémoire expose les principes et la marche à suivre pour établir et analyser un modèle de quantification. Il explique la préparation des solutions puis les prétraitements à réaliser et se termine enfin par l'analyse des modèles obtenus et le choix du meilleur modèle. Ce travail a permis de mettre au point un modèle de quantification pour trois des quatre substances présentes dans la solution étudiée. Pour la quatrième substance de par sa faible concentration dans le produit fini, aucun modèle de quantification n'a pu être mis au point malgré de nombreux essais réalisés. La spectroscopie proche infrarouge permet de réaliser des analyses quantitatives ou qualitatives comme ce travail l'a démontré. Elle est capable de donner des informations sur les propriétés physiques et chimiques des produits. Son développement sera optimal lorsque les industries pharmaceutiques et les autorités de tutelle auront acquises suffisamment d'expérience dans ce domaine.
