Book excerpt: Viral synergism occurs when two or more unrelated viruses simultaneously infect the same plant and the multiplication of one of the viruses is enhanced. This is generally associated with no change(s) in multiplication of the other viruses involved. Synergism also results in intensification of symptoms. In mixed-infection, viruses may also interact in an antagonistic manner, where one virus suppresses the replication or accumulation of another virus. This phenomenon is uncommon, and only two cases have been reported where the coat protein (CP) accumulation of one of the viruses has decreased. A number of synergistic interactions studied involve viruses belonging to the Potyviridae family. The increase in CP accumulation of the non-potyviruses in such an interaction has been attributed to the effect of the helper-component proteinase (HC-Pro) of potyviruses. Plant antiviral defense mechanism called "gene silencing". HC-Pro is known as a strong suppressor of gene silencing and represents the first identified and characterized plant viral suppressor of gene silencing. The ability of Soybean mosaic virus (SMV), a member of the Potyviridae family, to interact synergistically with Bean pod mottle virus (BPMV) and Cowpea mosaic virus (CPMV) in mixed-infection in soybean has been demonstrated, but no change in the level of accumulation of CP of SMV was reported. In addition to SMV, soybean is infected by many other potyviruses or non-potyviruses, including Alfalfa mosaic virus (AMV). This research was aimed at studying the interaction of SMV with AMV in mixed-infection in soybean. Two biologically distinct SMV strains and three AMV isolates were used in this study and their interactions in mixed-infection in two different cultivars of soybeans (Williams 82 and Lee 68) were investigated. It was demonstrated that (a) mixed-infection between AMV and SMV can be easily established, irrespective of sequential or simultaneous inoculation of the two viruses; (b) based on CP accumulation and disease phenotype, AMV interaction with SMV is synergistic resulting in enhancement in symptom severity and AMV CP accumulation; (c) synergistic interaction of AMV with SMV is strain and cultivar independent; (d) interaction of SMV with AMV is antagonistic, which is also strain and soybean cultivar-independent.

Book excerpt: Soybean Mosaic Virus (SMV) is a prevalent viral pathogen transmitted by aphids and via seed, causing significant yield loss and quality reduction. There are seven SMV strain groups (G1 to G7) and three resistance loci (Rsv1, Rsv3, and Rsv4) reported in soybean. New sources of SMV resistance would be valuable for breeding cultivars with durable resistance to multiple strains. The objectives of this research were to analyze genetic diversity of SMV-resistant soybean germplasm at the molecular level; to pyramid three genes from different sources for durable SMV resistance; and to identify and map new genes/alleles for differential reactions to SMV strains. One hundred and fourteen SMV-resistant germplasm genotypes collected worldwide were screened with 98 simple sequence repeat (SSR) markers. The 114 germplasm genotypes were grouped into four clusters with the largest group containing most of Asian genotypes. Rsv1 alleles were found to be most common, while Rsv3 and Rsv4 were rare among all the SMV resistant germplasm tested. PI 96983 (Rsv1) was crossed with Columbia (Rsv3, Rsv4) to pyramid all three SMV resistance genes with the aid of SSR markers. The molecular screening of a F2 plant population derived from PI 96983 x Columbia showed good fit to the expected genotypic ratios at each marker locus; one out of 70 plants was identified to contain three resistance genes in homozygous condition (Rsv1+Rsv3+ Rsv4), while 26 additional plants also contained three genes but at the heterozygous condition. F2:3 lines inoculated with G1 and G7 strains confirmed the presence of specific alleles and SMV resistance. A new SMV resistant parent PI 61944 with unknown gene(s) (Rsv-?) was crossed with the susceptible parent Essex (rsv), and resistant lines PI 96983 (Rsv1), L29 (Rsv3), and V94-5152 (Rsv4) to investigate the inheritance and allelism of SMV resistance. F2 plants were screened with selected SSR markers, and the F2:3 lines were screened with SMV G1 and G7. SMV screening and molecular analysis revealed that the PI 61944 carries a new allele at the Rsv3 locus on linkage group (LG) B2. This research has identified genetic diversity among SMV-resistant soybean germplasm, combined three genes for durable resistance in soybean, and discovered a new SMV resistance allele. These germplasm will be useful in soybean breeding programs where SMV resistance is an objective. This research also demonstrated that SSR technology is extremely useful in marker assisted selection (MAS).

Seven SMV strains, G1 - G7, and three independent multi-allelic loci for SMV resistance, Rsv1, Rsv3, and Rsv4, have been identified. In the initial study, 299 soybean germplasm lines were genotyped for Rsv4 region, inoculated with SMV-G1 and G7 strains, and classified into several resistance groups. The Glyma.02g121400 locus was sequenced from ten soybean accessions, and alignment of the sequences revealed three SNPs displaying 100% polymorphic consistency when a soybean genotype carrying the Rsv4 gene was present. A cross between V94-5152 {u00D7} Lee 68 was made to create linkage map revealing a distance of 3.6 cM between the Rsv4 and the closest SNP. Five Rsv4 candidate genes have been proposed in this region. In the second study, three SMV R-genes were pyramided by crossing J05 and V94-5152. The gene-pyramided line GP20, was crossed with Williams 82, F2 plants were genotyped and collated with phenotypic data of F2:3 lines inoculated with SMV-G1 and G7 strains. The results confirmed a successful incorporation of three genes into one soybean line. In the third study, soybean germplasm PI 438307 was crossed to Essex for the inheritance study, and to three differential parents for the allelism test. F2 population and F2:3 lines derived from all four cross combinations were screened with SMV-G7 strain. Additionally, F2 generation of PI 438307 x Essex were genotyped with two SSRs. The results revealed that resistance to SMV in PI 438307 is controlled by a single dominant gene at the Rsv4 locus. PI 438307 plants exhibited a unique symptoms; therefore, a new allele Rsv4-v was assigned to SMV resistance in PI 438307. In the final study, PI 96983 and York were crossed to evaluate allelomorphic relationship between Rsv1 and Rsv1-y. To break possible linkage, 3000 F2-plant population was phenotyped using the SMV-G1 strain. Occurrence of susceptible and segregating lines indicated tight linkage between two genes positioned in a distance of 2.2 cM. The Rsv2 symbol was proposed to be assigned instead of Rsv1-y. Results from this research may accelerate breeding efforts to develop multi-virus resistant crops.
