To better understand how estrogen-responsive genes are regulated, our laboratory identified a number of proteins that associate with the DNA-bound ERalpha. Our studies demonstrate that the nonmetastatic protein 23 homolog 1 (NM23-H1) interacts with ERalpha, increases ERalpha-ERE complex formation, influences ERalpha-mediated transcription and associates with the promoter region of the endogenous estrogen-responsive progesterone receptor (PR) gene. Furthermore, we show that a second protein, Apurinic/apyrimidinic endonuclease 1 or redox factor-1 (Ape1/Ref-1), interacts with ERalpha, promotes the ERalpha-ERE interaction, influences ERalpha-mediated transactivation, and selectively associates with endogenous, estrogen-responsive genes in MCF-7 cells. Our findings suggest that NM23-H1 and Ape1/Ref-1 are instrumental in modulating expression of estrogen-responsive genes. Interestingly, we demonstrate that Ape1/Ref-1 and NM23-H1, as well as the oxidative stress proteins Cu/Zn superoxide dismutase (SOD1), thioredoxin (Trx) and protein disulfide isomerase (PDI) are overexpressed in human breast cancer tissues. These studies provide a novel link between DNA repair, redox regulation and modulation of ERalpha-mediated transcription.

Who would have imagined that we could synthesize a nucleotide sequence that binds a specific steroid receptor and acts as a controlling element for gene expression? No one; but as is evident from the results reported in several chapters, this technique is yielding a wealth of information. Using these and other techniques it has become apparent that gene transcription is controlled by interactions between transacting factors and DNA recognition sequences (response elements). These transacting factors appear to be members of a large gene family that includes steroid hormone receptors, transcription factors, protooncogenes and homeobox proteins. Thus a great deal has been learned, but as usual, questions remain. Many of these questions are posed by the findings and observations found in several chapters in this volume. Non hormone binding forms of steroid receptors and their relevance to receptor down regulation, recycling and biological response remain a mystery. The quantitative relationship between receptor binding and biological response still presents agonizing problems. The growth of many human breast carcinomas is regulated by the female hormone estrogen through the action of the estrogen receptor protein. The logic of our approach was to develop small, cell-permeable molecules that prevent the activation of downstream genes by the DNA-binding protein estrogen receptor. A series of pyrrole/imidizole polyamides have synthesized in the laboratory of Dr. Peter Dervan at The California Institute of Technology and supplied to our laboratory. These polyamides were designed to bind the 6 bp half-site recognized by estrogen receptor. Standard DNase footprinting methods were used to measure the binding affinities of the synthetic ligands for their target sequences. A series of polyamides were screened for binding affinities and sequence specificity. We have used recombinant human estrogen receptor protein in DNA binding studies with the same target ERE sequences. Using DNase footprinting methods and gel mobility shift assays, we optimized conditions for ER-DNA interactions and we have shown that the ERE-binding polyamides inhibit ER binding to EREs. Future studies will examine whether these compounds are effective inhibitors of ER-dependent gene transcription in breast carcinoma cells in culture.
