Integrated Virus Detection describes this technology and provides many examples of applications including a chapter on viruses found in honeybees with descriptions of seasonal and yearly variation. This straightforward technology can be used to detect known, unknown, and unsequenced viruses collected from environmental and other complex biological sources. This book summarizes more than ten US patents issued for the invention of the IVDS, which is the common name of the electrospray–differential mobility analyzer method. The IVDS is powering mankind’s ability to rapidly detect, measure, and monitor viruses as well as virus-like particles. Three facts make rapid detection possible: virus size, which ranges from 20 to 800 nm.; disparity in each virus species’ particle size thus allowing size data to be used for detection and preliminary identification; and the fact that virus particle density is distinct from other nanoparticles. The IVDS can ascertain the absence of virion particles, thus presenting compelling evidence of a true negative, which is useful in verifying decontamination and other processes. Book excerpt: The detection and analysis of viruses have been goals of science for more than 70 years, following the first real evidence that a new type of microorganism was responsible for diseases in man and animals. These new microbes were smaller than bacteria, which have now been well documented, classified, and studied. Their small size made classifying the new microbes more difficult, and the field of virology has been advanced by biochemical techniques rather than by direct examination. Advancements in electron microscopy in more recent times have made advances in this area, and much has been reported on the physical features of more than 21 virus families. All these historic techniques are time consuming and require special knowledge and specialized chemicals and preparations to be successful. Capitalizing on the physical characteristics, it was possible to separate the families and count the individual viruses in a new and dramatic way, using easily obtained materials and simple-to-operate techniques. The only materials used in the Integrated Virus Detection System (IVDS) are a buffer, CO2 gas, and butanol. The work in this report presents the advances in the concentration and counting phases of IVDS. Results indicate a practical, easy to use device, which can count viruses in near real time. The JVDS can, in its final configuration, be expected to analyze a sample, count the viruses present, and give a preliminary identification for all viruses.

Book excerpt: A new physically based methodology - The Integrated Virus Detection System (IVDS) - was used to characterize a high concentration, 10.2 mg protein/ml, sample preparation of MS2 Bacteriophage with a reported 10(exp 14) pfu/ml (DPM14) virus count in a common TNME buffer. Virus counts were made using the IVDS instrument following serial dilution. Results indicated virus counts of 1.5 x 10(exp 5) for the neat sample (DPM14), followed by 6.5 x 10(exp 4) viruses (DPM13), 1.2 x 10(exp 4) viruses (DPM12), 9.3 x 10(exp 2) viruses (DPM11), 88 viruses (DPM10), and 5 viruses (DPM9), respectively. Lower concentrations display a consistent multiplier and were consistent with target dilutions. Increases in virus concentration appear to decrease the multiplier. Variation is considered to be due to aggregation. Results demonstrate a consistent and simple-to-use methodology. Results further indicate that the IVDS instrument can be used for characterization of other virus preparations with equal ease and similar results.

The MS2 samples were grown and purified using various techniques, which showed ESI-MS samples responding differently from IVDS samples. The LC-MS of the specific biomarker of the MS2 bacteriophage from an infected Escherichia coli (E. coli) sample was characterized in the presence of E. coli proteins. The significant impact of the sample matrix was observed upon the identification of MS2 using a database search. Escherichia coli infected with MS2 showed a different score from other uninfected scores. Only purified MS2, using CsCl and analyzed by LS-MS, showed a positive match in the database search. However, the variation in the MS2 sample matrix had no effect on the characterization of MS2.

The need for a rapid and trustworthy virus detector remains an urgent and continuing requirement for commanders requiring an assurance that they have early warning from these threats. This short report presents a historical summary, with references, of methods used to extract, purify, and concentrate viruses. Working with viruses is not an easy job. The requirement for detection was identified nearly 60 years ago and just now is seeing an approach with the evolution of the Integrated Virus Detector System (IVDS) device to undertake this exacting and difficult task. This work expounds a new methodology - IVDS. The IVDS utilizes a physical process to detect viruses. This is a departure from the traditional approaches that historically have utilized obscure chemical reactions, complicated reagents and other exacting and difficult procedures for detection. The IVDS is based in four physical stages which are collection, separation, purification, and detection. Development and risks are discussed. An IVDS development schedule and costs are given that could lead to an advance prototype in two years. The IVDS system promises a very sensitive, broad spectrum "generic" virus detector free from reagent based reactions, simple to operate, and providing a capability for continuous monitoring and recording of virus levels in the outdoor bioaerosol.

Book excerpt: This is the third report in a series of reports that investigates the survivability of the MS2 virus in harsh environments. The first two reports study the effects of pH on the virus. In this report, we study the effect of elevated temperatures on the virus. The stability of the virus was studied by counting the number of virus particles using the Integrated Virus Detection System (IVDS). Test results show that the virus particles are relatively stable for a limited time (up to 0.50 hr) at temperatures as high as 63 degrees C. Longer exposure or exposure to higher temperatures, results in quick breakdown of the virus particles.

This suggests that viruses might be able to survive such harsh environmental conditions.
