Book excerpt: The effects of classic enzyme inhibitors, phenols, and extracts from wood on activity of cellulases from two brown-rot and two white-rot fungi were investigated. The cellulases from the fungi varied in their tolerances to the inhibitors. This suggested structural differences in the enzymes. C sub x cellulases, which break down soluble cellulose, were not inhibited in any of the fungi by sulfhydryl-binding compounds and metal-complexing agents. Thus, sulfhydryl groups apparently are not an active site of C sub x, and the enzyme does not require metal. Most phenols inhibited cellulase only when oxidized, and generally inhibited C sub x of the white-rot fungi more than C sub x of the brown-rot fungi. The purpose of the study is to investigate the inhibition of cellulases from wood-decaying fungi by using the following: classical enzyme inhibitors, phenolics, and extracts from wood.

The objective of the study is to determine the influence of the morphological characteristics of pulp fiber from 12 western U.S. softwood species, representative of a wide range of fiber. The effect of the morphology of the wood pulp fiber as distinguished from that of the wood fiber is emphasized because differences in performance of fiber-based products are traced to the pulp fiber. Interrelationships of fiber morphology and pulp sheet properties are discussed. The results indicate that it is not possible to fully characterize the performance of a pulp by a single morphological characteristic and that the morphological characteristics significantly influence strength properties of sheets.

Book excerpt: The oil crisis during the 1970s turned interest towards the utilization of renewable resources and towards lignocellulosics in particular. The 1970s were also the cradle period of biotechnology, and the years when biotechnical utilization of lignocellulosic waste from agriculture and forestry gained priori ty. This was a logical conclusion since one of nature's most important biologi cal reactions is the conversion of wood and other lignocellulosic materials to carbon dioxide, water and humic substances. However, while biotechnology in other areas like medicine and pharmacology concerned production of expen sive products on a small scale, biotechnical utilization and conversion of ligno cellulosics meant production of inexpensive products on a large scale. Biotechnical utilization of lignocellulosic materials is therefore a very difficult task, and the commercial utilization of this technology has not progressed as rapidly as one would have desired. One reason for this was the lack of basic knowledge of enzyme mechanisms involved in the degradation and conversion of wood, other lignocellulosics and their individual components. There are also risks associated with initiating a technical development before a stable platform of knowledge is available. Several of the projects started with en thusiasm have therefore suffered some loss of interest. Also contributing to this failing interest is the fact that the oil crisis at the time was not a real one. At present, nobody predicts a rapid exhaustion of the oil resources and fuel production from lignocellulosics is no longer a high priority.
