Book excerpt: Les nanoparticules métalliques manufacturées (NPs) sont des polluants émergents dont la concentration augmente dans les sols en raison de leur utilisation croissante dans de nombreux produits commerciaux de la vie courante (cosmétiques, aliments, peintures...). Des études in vitro ont montré la toxicité des NPs pour les microorganismes, mais il existe encore peu de données sur l'écotoxicité et le devenir de ces contaminants dans les sols. L'objectif de cette thèse est donc d'évaluer l'influence des paramètres abiotiques du sol sur (i) les caractéristiques physico-chimiques et le transfert des NPs, et (ii) sur la toxicité des NPs pour les communautés microbiennes du sol, en particulier pour des groupes fonctionnels microbiens impliqués dans le cycle du carbone et de l'azote. Nous avons mis en évidence que les propriétés du sol influençaient l'agrégation et la charge de surface des NPs de dioxyde de titane (TiO2) et d'oxyde de cuivre (CuO). Dans les six sols agricoles étudiés, nous avons observé un transport très faible des NPs testées lors d'une expérimentation en colonnes de sols. Nous avons mis en évidence une absence de toxicité des NPs de TiO2 sur les communautés microbiennes, sauf dans un sol limono-argileux à forte teneur en matière organique. Dans ce sol, des effets négatifs ont été observés après 90 jours d'exposition sur les activités microbiennes, sur l'abondance des microorganismes nitrifiants et la diversité des bactéries et des archées. Des études complémentaires en colonnes de sol, ont permis de mettre en évidence des effets délétères des NPs plus importants que la nitrification lors d'une contamination chronique au TiO2 que lors d'une contamination aigüe.

Certains de ces nanomatériaux sont déjà commercialisés. Cet essor des nanosciences n'a malheureusement pas été accompagné par les études toxicologiques adéquates. Le but de mon travail est donc d'apporter des éléments de réponse à cette problématique. Dans cette optique, l'analyse des interactions biologiques entre des nanoparticules d'oxyde de zinc (ZnO) semiconducteur utilisé dans les crèmes solaires, et Escherichia coli, une bactérie présente dans l'environnement, a été réalisée. Le comportement toxique des ZnO a été évalué dans deux milieux : un milieu de croissance classique, le Luria Bertani (LB), et un milieu environnemental, l'eau de Seine. Les résultats obtenus dans ce travail ont mis en évidence la nécessité d'une approche pluridisciplinaire pour étudier l'impact toxique des nanoparticules. Ainsi, des effets bactériostatique et bactéricide des nanoparticules ont été démontrés, mais la comparaison des effets biologiques observés lors d'une incubation avec des ions Zn2+, suggère une dissolution partielle des nanoparticules. Cette hypothèse a été confirmée par l'étude de la solubilité des nanoparticules dans les différents milieux. La composition du milieu joue indéniablement un rôle dans le potentiel toxique des ZnO. L'implication d'un stress oxydatif dans les mécanismes de toxicité des nanoparticules a également été observée en utilisant une souche mutée pour l'enzyme de la superoxyde dismutase.

Dans ce travail de thèse, nous avons étudié les effets des propriétés de surface et de taille de nanoparticules d’oxyde de fer, supposées être biocompatibles et qui sont utilisées dans le cadre de certaines applications biomédicales, telles que l'imagerie et l'hyperthermie magnétique. Des nanoparticules de maghémite de 6 à 50 nm ont été synthétisées, de forme sphérique ou en forme de nano-fleurs et fonctionnalisées de façon à ce qu’elles possèdent une charge de surface positive ou négative. Les propriétés physicochimiques de ces particules ont été mesurées : leur taille, leur surface spécifique, leur densité de charge de surface et leur stabilité dans le milieu de culture cellulaire ont été établies. L’influence de ces paramètres sur la mortalité de cellules alvéolaires humaines cancéreuses a été testée. Les résultats montrent que la fonctionnalisation de surface des nanoparticules d'oxyde de fer a une forte influence sur leurs interactions avec les cellules et leur toxicité, en jouant notamment sur leur stabilité dans le milieu de culture cellulaire. Par ailleurs, les nanoparticules chargées positivement sont plus toxiques que celles possédant une charge de surface négative. Les nano-fleurs ne sont pas plus toxiques que les nanoparticules sphériques et possèdent des propriétés remarquables d’agents de contraste IRM et en tant que vecteurs d’hyperthermie magnétique : leur utilisation dans le domaine biomédical est donc prometteuse
