Chronic exposure to jet fuel has been shown to adversely affect human liver function, to cause emotional dysfunction, to cause abnormal electroencephalograms, to cause shortened attention spans, and to decrease sensorimotor speed. Currently, there are no standards for personnel exposure to jet fuels of any kind, let along JP-8 jet fuel. Kerosene based petroleum distillates have been associated with hepatic, renal, neurologic and pulmonary toxicity in animals models and human occupational exposures. The U.S. Department of Labor, Bureau of Labor statistics estimates that over 1.3 million workers were exposed to jet fuels in 1992. Thus, jet fuel exposure may not only have serious consequences for USAF personnel, but also may have potential harmful effects upon a significant number of civilian workers. Short-term (7 day) JP-8 jet fuel exposure causes lung injury as evidenced by increased pulmonary resistance, a decrease in bronchoalveolar lavage concentrations of substance P, increased wet lung/body weight ratio, and increased alveolar permeability. Long-term exposures, although demonstrating evidence of lung recovery, results in injury to secondary organis such as liver, kidneys and spleen.

Book excerpt: Experiments were performed exposing mice to varying doses of either JP-8 or S-8 jet fuel for 1h/day for 7 days, using the new exposure apparatus with in-line monitoring of jet fuel concentrations (aerosol and vapor). Six controls, 6 JP-8 exposed and 6 S-8 exposed animals examined. There were no significant effect on body weight due to JP-8 or S-8 exposure, and no effect on immune cell viability from any immune organ due to JP-8 or S-8 exposure. There was a 53% decrease in Spleen weight due to S-8 exposure and a corresponding 40% decrease due to JP-8 exposure. There was a 36% decrease in Thymus weight due to S-8 exposure and a corresponding 60% decrease due to JP-8 exposure. There was a 64% decrease in Thymus cell numbers due to S-8 exposure and a corresponding 46% decrease due to JP-8 exposure. There was a 20% decrease in Spleen cell numbers due to JP-8 exposure only. No significant change in composition in any immune organ due to JP-8 or S-8 exposure. Both JP-8 and S-8 exposures using the new exposure chamber resulted in changes in immune function.

Book excerpt: During our preliminary year of experimentation we have made an initial examination Off the immunotoxicological effects of JP-8 jet fuel exposure. Inbred C57BL6 mice were exposed to varying concentrations (either 500, 1000 or 2500 mg/m3) of aerosolized JP-8 jet fuel for a period of 7 days with an exposure period of 1 hour per day. Animal exposure was performed via nose-only presentation while the animals were held in individual subject loading tubes. The tubes were nose cone fitted to receiving adapters that originated from a common anodized aluminum exposure chamber. Nose only exposure was utilized to minimize ingestion of jet fuel during self grooming. Animals were rotated on a daily basis through the 12 adapter positions on the exposure chamber. This rotation was done to minimize proximity to the jet fuel source as a variable in exposure concentration or composition. Exposure concentration was determined by a seven stage --cascade impactor, and were measured after each exposure (1,2). 24 hours after the last exposure the animals were sacrificed and examined for changes in immune system composition and function. The major immune system organ systems (i.e., spleen, thymus, lymph nodes, blood and bone marrow) were recovered and examined for changes in organ weight total cell numbers, immune cell components (by differential histochemical staining).

Will exposure to JP-8 interfere with the immune response to recall antigens and block the protective effect of prior immunization? 3) How can we prevent JP-8 induced immune suppression? Once a better understanding of the suppressive mechanism is obtained, we will begin to design rational steps aimed at preventing its immune suppressive effect. 4) What components in JP-8 induce dermal immunotoxicity? JP-is produced from commercial Jet-A by use of an additive package containing diethylene glycol monomethyl ether, (DiEGME) an anti-icing compound. Because some have suggested DiEGME is a dermal immunotoxin we will concentrate on determining if application of DiEGME induces immune suppression. The specific aims of this proposal are designed to gain an understanding of the mechanism(s) involved in JP-8-induced immune suppression. The long-term of this work is to use the knowledge obtained here to design a procedure to prevent JP-8-induced immune suppression and limit its potential harmful effect on USAF personnel.

Book excerpt: Military personnel during the Gulf War were exposed to various compounds including N, N-diethyl-m-toluarmide (DEET), pyridostigmine bromide (PYR), and W-8 jet fuel. The effects of concurrent exposure to these agents co%could potentially result in significant health effects or contribute to the manifestations of Gulf War Syndrome. Adenopathy, fever, joint pain, and skin rash are reported by veterans and these signs may be linked to immune dysfunction. Recent studies indicate that neurotoxicity increases during concurrent exposure to both PYR and DEET with other agents. Due to known interdependencies of the immune and nervous systems, it is hypothesized that the effects of PYR and DEET may affect the immune system. Additionally, single low-level exposures to W-8 have been reported to significantly affect T- and B-lymphocyte populations. Thus, the possibility exists that combinations of DEET and PYR could exacerbate the effects of W-8 or cause a predisposition to other immune conditions such as autoimmune disease. Therefore, this investigation will evaluate the effects of concurrent exposure to these common military agents on immunological parameters (i.e., natural killer cell activity, macrophage function, etc.) and - susceptibility to diseases such as cancer, infections, and autoimmune disease.
