Book excerpt: Modern Methods of Plant Analysis When the handbook Modern Methods of Plant Analysis was first introduced in 1954 the considerations were: 1. the dependence of scientific progress in biology on the improvement of existing and the introduction of new methods; 2. the difficulty in finding many new analytical methods in specialized journals which are normally not accessible to experimental plant biologists; 3. the fact that in the methods sections of papers the description of methods is frequently so compact, or even sometimes so incomplete that it is difficult to reproduce experiments. These considerations still stand today. The series was highly successful, seven volumes appearing between 1956 and 1964. Since there is still today a demand for the old series, the publisher has decided to resume publication of Modern Methods of Plant Analysis. It is hoped that the New Series will be just as acceptable to those working in plant sciences and related fields as the early volumes undoubtedly were. It is difficult to single out the major reasons for success of any publication, but we believe that the methods published in the first series were up-to-date at the time and presented in a way that made description, as applied to plant material, complete in itself with little need to consult other publications. Contributing authors have attempted to follow these guidelines in this New Series of volumes.

This book supplements the web site www.juniperus.org and the reader will find literature citations and a detailed discussion of data that is not present on the web site. The Junipers of the World is now the authoritative reference for Juniperus Prof.Adams has presented arguments for the recognition of species and varieties by showing the data analyses that led to taxonomic decisions. This is one of the few treatments that really integrates data and the reasoning behind taxonomic decisions. In addition, Prof. Adams is candid in admitting that the recognition of some taxa are not well supported and deserve additional study. An essential reference book that will be useful for herbaria, museums, wildlife biologists, Range scientist, foresters, ecologists and all who are involved in identification and study of Juniperus species.

Book excerpt: The second edition of Gas Chromatography and Mass Spectrometry: A Practical Guide follows the highly successful first edition by F.G. Kitson, B.S. Larsen, and C.N. McEwen (1996), which was designed as an indispensible resource for GC/MS practitioners regardless of whether they are a novice or well experienced. The Fundamentals section has been extensively reworked from the original edition to give more depth of an understanding of the techniques and science involved with GC/MS. Even with this expansion, the original brevity and simple didactic style has been retained. Information on chromatographic peak deconvolution has been added along with a more in-depth understanding of the use of mass spectral databases in the identification of unknowns. Since the last edition, a number of advances in GC inlet systems and sample introduction techniques have occurred, and they are included in the new edition. Other updates include a discussion on fast GC and options for combining GC detectors with mass spectrometry. The section regarding GC Conditions, Derivatization, and Mass Spectral Interpretation of Specific Compound Types has the same number of compound types as the original edition, but the information in each section has been expanded to not only explain some of the spectra but to also explain why certain fragmentations take place. The number of Appendices has been increased from 12 to 17. The Appendix on Atomic Masses and Isotope Abundances has been expanded to provide tools to aid in determination of elemental composition from isotope peak intensity ratios. An appendix with examples on "Steps to follow in the determination of elemental compositions based on isotope peak intensities" has been added. Appendices on whether to use GC/MS or LC/MS, third-party software for use in data analysis, list of information required in reporting GC/MS data, X+1 and X+2 peak relative intensities based on the number of atoms of carbon in an ion, and list of available EI mass spectral databases have been added. Others such as the ones on derivatization, isotope peak patterns for ions with Cl and/or Br, terms used in GC and in mass spectrometry, and tips on setting up, maintaining and troubleshooting a GC/MS system have all been expanded and updated. - Covers the practical instruction necessary for successful operation of GC/MS equipment - Reviews the latest advances in instrumentation, ionization methods, and quantitation - Includes troubleshooting techniques and a variety of additional information useful for the GC/MS practitioner - A true benchtop reference - A guide to a basic understanding of the components of a Gas Chromatograph-Mass Spectrometer (GC-MS) - Quick References to data interpretation - Ready source for information on new analyses

This book was released on 2013 with total page pages. Available in PDF, EPUB and Kindle. Book excerpt: Essential oils are amongst the most complex samples an analyst can face in terms of the number of compounds involved. In many cases, minor components are of interest as they can impart a distinctive fragrance character to the oil. Because of the closely related structures and molecular weights among terpenes, positive identification of individual compounds is very difficult with a single chromatographic technique. Further, most of the analytical information is lost when a single technique is used because of the limited peak capacity and the resulting peak overlap. For many years, gas chromatography coupled to mass spectrometry (GC-MS) has been the benchmark technique for qualitative and quantitative analysis of essential oils. Retention indices and mass spectra have to be used in combination for confirmation of the identity of components in an essential oil. Other multidimensional or hyphenated techniques also offer advantages that aid in the identification of essential oil components. This thesis demonstrates the application of a comprehensively coupled supercritical fluid and fast temperature programmed gas chromatograph (SFCxGC) to the analysis of essential oils. An SFCxGC instrument was used to analyse the essential oils of Cymbopogon (lemongrass), Artemisia afra (wilde als), Tagetes minuta (kakiebos) and Pelargonium (geranium) species. The unique application of a porous layer open-tubular (PLOT) column, used in conjunction with supercritical carbon dioxide is demonstrated to effect group separation of polar, oxygenated compounds. This separation and elution of very polar compounds from a silica gel column is believed to occur due to the reduced phase ratio (??) of the system obtained by increasing the volume available to the mobile phase compared to that of a packed column. This separation obtained in the SFC is used to separate essential oils into different chemical classes such as non-polars, ethers, alcohols. Separated chemical classes are re-injected on-line by use of a modulator into a fast, second dimension, temperature programmed GC to effect separation of individual compounds based on their volatility. The entire sample is analysed by both the SFC and GC in such a way that the resolution obtained in the first dimension is conserved by the GC analyses. By using a range of standards, some of the peaks in these oils could be assigned. The identification of compounds was greatly aided by the combination of the two separation dimensions. The comprehensive two-dimensional technique arranges component peaks in a plane from which chemical class and volatility information of each component is readily obtained. The elution pattern within the two-dimensional chromatograms may also be used for direct comparison of oils without identification of the components in the essential oils.

Book excerpt: Given the growing importance of essential oils and waxes, this volume deals with the analysis of a broad spectrum of these compounds from many plant origins. Commercial oils such as olive oil are analysed as are trees such as eucalyptus, mentha, cedar and juniper. In addition, analysis of spices, seasoning, seaweeds, perfumes, liquors and atmospheric monoterpene hydrocarbons are to be found in this book. The volatiles of flower and pollen may be of importance in attraction of bees and other insects to certain plants for pollination purposes; this topic is also discussed. Waxes, both in the soil and as leaf components are analysed and presented in such a way making this book valuable to scientists with varying interests worldwide.
