Book excerpt: The concept that hormones influence tumor growth originated in 1889 with the proposal of Albert Schinzinger who suggested that breast cancer is related to the ovaries. Several years later, Sir George Beatson observed that remission of disseminated breast cancer could be achieved in premenopausal patients by performing bilateral oophorectomy. As a result of the contri butions of Hedley Atkins, Charles Huggins and others, additive and ablative hormonal therapies have been widely used for the treatment of advanced breast cancers for several decades. Model systems to study the effects of hormones on growth and regression of breast tumors have been available for many years; however, the complexities of the hormonal environment have rendered in vivo studies difficult in man and experimental animals. Recently, the availability of long-term cultures of breast cancer cells has stimulated many investigators to use these cell lines to unravel the mechanisms of hormone action. Because of the extreme diversity and complexity of advances regarding the endocrinology of the breast and breast cancers, a multi-authored review was deemed necessary. It has been gratifying to receive contributions from many noted scholars. In Volume I of this monograph, the influence of steroid hormones and their antagonists upon normal and neoplastic tissues of the mammary gland are presented. In Volume II, the effects of peptide and other hormones are reviewed.

Available in PDF, EPUB and Kindle. Book excerpt: The papers in this volume were presented at the Symposium on Hormones and Cancer held March 24-26, 1980, at the Worcester Foundation for Experimental Biology, Shrewsbury, MA. The meeting was organized to review recent advances in basic and clinical research work on hormone-responsive tumors of the reproductive system. The association between protracted hormone action and cancer of the reproductive system is now irrefutable. Yet we still do not know how hormones initiate and promote neoplastic transformation of their target cells. A major effort is cur rently being directed at understanding the hormonal regulation of neoplastic cells, especially those arising from the breast, uterus, pituitary gland and prostate. The symposium brought together leading experts whose research is focused on this impor tant area of human health. Although many questions remain to be answered by future studies, the material covered in this volume provides an up-to-date overview of the current status of work in this area. The subjects range from the mechanism of hormone action to the treatment of hormone-dependent tumors, and topics include estrogen, progestin, androgen and glucocorticoid action~ hormone receptors~ specific protein responses~ developmental effects of estrogens and antiestrogens~ the DES syndrome~ regu lation of the cell cycle~ therapeutic effects of antiestrogens, aromatase inhibitors, iodoestrogens, progestins on estrogen dependent tumors~ characterization of retinoid-binding sites~ and the utility of various tumor markers.

Book excerpt: The International Foundation for Biochemical Endocrinology is incorporated as a nonprofit research and educational organization. It is dedicated to the dissemination of knowledge, cooperative research programs, and cultural interaction on an international basis. The Foundation is concerned with both basic research and practical applications of biological knowledge to the betterment of humanity. Among our interests are global resource management, human reproduction, hormonal regulation of normal and cancer cells, study of aging and degenerative diseases, brain peptides, peptide neurotransmitter compounds, mechanism of action of hormones, peptide hormone synthesis, and recombinant DNA techniques. This monograph is the ninth sponsored by the Foundation in the Biochemical Endocrinology series. The previous four have been: Hormonally Active Brain Peptides: Structure and Function (1982), K. W. McKerns and V. Pantie, eds.; Reproductive Processes and Contraception (1981), K. W. McKerns, ed.; Synthesis and Release of Adenohypophyseal Hormones (1980), M. Jutisz and K. W. McKerns, eds.; and Structure and Function of the Gonadotropins (1978), K. W. McKerns, ed. These have all been published by Plenum Press.

Book excerpt: In the past decade there has been a growing public interest and resurgence in research in the field of hormonal carcinogenesis. This is due to the widespread use of therapeutic hormonal agents worldwide and to the increasing awareness of the causal association of hormones, both endogenous and exogenously administered, and a variety of human cancers. These associations include estrogens in uterine, cervical, vaginal, liver, testicular, prostatic, and possible breast cancers; progesterone and progestational hormones in breast cancer; androgens and anabolic steroids in hepatic and prostatic cancers. Additionally, gonadotrophins playa role in the etiology of ovarian and testicular cancers and thyroid-stimulating hormones in thyroid cancers. Therefore, hormonal carcinogenesis encompasses the study of both natural and synthetic hormonal agents, including growth factors and other peptide and protein factors, which contribute substantially to the etiology of both human and animal neoplasms, benign or malignant. Hormones may be involved in all aspects of neoplastic transformation, including initiation, promotion, and progression, and the inhibition of these processes. There are a number of important issues in women's health that need to be addressed. More than 40 million U. S. women are menopausal, and these women have a life expectancy of over 30 years after the menopause. When these figures are multiplied worldwide, the numbers become staggering. After the menopause, estrogen replacement therapy (ERT) is the choice of most women in industrialized countries.

Available in PDF, EPUB and Kindle. Book excerpt: The link between the pineal gland and cancer is a rapidly emerging research field due to promising experimental and clinical trials with melatonin. The pineal gland acts as a transducer of environmental light to regulate rhythmic processes, including reproductive function in seasonally breeding animals and the entrainment of circadian rhythms, such as the sleep-wake cycle, in man. This book elucidates the physiological significance of the pineal gland and surveys phenomena and mechanisms of pineal - tumor interaction at the neuroendocrine, neuroimmune, neural, and molecular levels. Yet unidentified low-molecular-weight pineal substances with tumor-inhibiting capacity, a possible involvement of melatonin in electromagnetic field effects on cancer, and the oncotherapeutic potential of melatonin are also addressed. The encouraging results should incite further research to elucidate the exact nature of the link between the pineal gland and cancer for the benefit of patients.
