Book excerpt: More than ever, the introduction of new methods or techniques serves to stimulate progress into understanding the structure and function of the nervous system. This axiom is exemplified by recent techniques that have revolutionized several branches of neurochemistry and promise to remain dominant for many years. Such developments underscore the need to remain abreast of new research strategies and provide further justification for the present series. The use of high performance liquid chromatography combined with bioassay methods provides a powerful technique for iso lation and assay of trace amounts of neuropeptides. Two chapters in the present volume deal with this subject: one (Stenn and Lewis) describes the assay of enkephalins, and the other (Chretien and Seidah), on lipo tropic peptides, includes procedures for structural analysis by microse quencing. These methods rival earlier ones for peptide separations in speed, sensitivity, and cost and have general applicability in most labo ratories. High performance liquid chromatography has also largely sup planted earlier and more tedious procedures for the assay of catechola mines, as described in Chapter 7 by Causon. As in earlier volumes, we have striven to retain a balance between studies on intact tissues and those on subcellular components.

Book excerpt: Modern Methods of Plant Analysis When the handbook Modern Methods of Plant Analysis was first introduced in 1954 the considerations were: 1. the dependence of scientific progress in biology on the improvement of existing and the introduction of new methods; 2. the difficulty in finding many new analytical methods in specialized journals which are normally not accessible to experimental plant biologists; 3. the fact that in the methods sections of papers the description of methods is frequently so compact, or even sometimes so incomplete that it is difficult to reproduce experiments. These considerations still stand today. The series was highly successful, seven volumes appearing between 1956 and 1964. Since there is still today a demand for the old series, the publisher has decided to resume publication of Modern Methods of Plant Analysis. It is hoped that the New Series will be just as acceptable to those working in plant sciences and related fields as the early volumes undoubtedly were. It is difficult to single out the major reasons for success of any publication, but we believe that the methods published in the first series were up-to-date at the time and presented in a way that made description, as applied to plant material, complete in itselfwith little need to consult other publications. Contributing authors have attempted to follow these guidelines in this New Series of volumes.

Book excerpt: This book originates from a symposium held at the London Hospital Medical College under the auspices of Applied Chromatography Systems Ltd. to discuss the place of HPLC in the endocrinology field. Many of the authors of the present book were speakers at this symposium. It seemed to us that many endocrinolo gists did not at that time fully appreciate the value of HPLC, and this book was designed to publicise the potential value of this technique. A survey of methods used in the steroid field Cp 185) confirmed the view that HPLC is not being used as widely, particularly in research described in clinical journals, as might be expected. We hope that this book will illustrate, albeit in a few selected areas of endocrinology, just how versatile and powerful a technique HPLC is, and en courage those who have not yet experienced it to have a go. The beginner does not need to buy expensive instrumentation - all that is required is a pump, injector, column and detector - the rest can come later! All the authors have practical experience in the use of HPLC in the particular area they discuss. All readers who discover apparent errors or who feel that the treatment of a topic of interest can be improved upon are encouraged to contact the editors. All criti cism, especially if constructive, is welcomed. We are still learning, and other peoples' experience is always valuable.

Book excerpt: The first book to offer a blueprint for overcoming the challenges to successfully quantifying biomarkers in living organisms The demand among scientists and clinicians for targeted quantitation experiments has experienced explosive growth in recent years. While there are a few books dedicated to bioanalysis and biomarkers in general, until now there were none devoted exclusively to addressing critical issues surrounding this area of intense research. Target Biomarker Quantitation by LC-MS provides a detailed blueprint for quantifying biomarkers in biological systems. It uses numerous real-world cases to exemplify key concepts, all of which were carefully selected and presented so as to allow the concepts they embody to be easily expanded to future applications, including new biomarker development. Target Biomarker Quantitation by LC-MS primarily focuses on the assay establishment for biomarker quantitation—a critical issue rarely treated in depth. It offers comprehensive coverage of three core areas of biomarker assay establishment: the relationship between the measured biomarkers and their intended usage; contemporary regulatory requirements for biomarker assays (a thorough understanding of which is essential to producing a successful and defendable submission); and the technical challenges of analyzing biomarkers produced inside a living organism or cell. Covers the theory of and applications for state-of-the-art mass spectrometry and chromatography and their applications in biomarker analysis Features real-life examples illustrating the challenges involved in target biomarker quantitation and the innovative approaches which have been used to overcome those challenges Addresses potential obstacles to obtain effective biomarker level and data interpretation, such as specificity establishment and sample collection Outlines a tiered approach and fit-for-purpose assay protocol for target biomarker quantitation Highlights the current state of the biomarker regulatory environment and protocol standards Target Biomarker Quantitation by LC-MS is a valuable resource for bioanalytical scientists, drug metabolism and pharmacokinetics scientists, clinical scientists, analytical chemists, and others for whom biomarker quantitation is an important tool of the trade. It also functions as an excellent text for graduate courses in pharmaceutical, biochemistry and chemistry.

Book excerpt: This sixth volume in the book series Progress in HPLC-HPCE examines the enhancement of high- performance liquid chromatography through the development of an advanced mode of electrochemical detection (ECD) --- the coulometric array detection --- from its initial, yet problematic, amperometric (thin-layer) design to the highly sensitive, selective and stable coulometric (flow-through) design. Unlike amperometric electrodes, the coulometric electrode is 100% efficient and measures signals from all of the analyte passing through it, which leads to improved sensitivity as well as unique selectivity. The coulometric electrode array offers the resolution of the photodiode array with the extreme sensitivity of an electrochemical detector.

This compilation is categorized into six major topics — mass spectrometry in research; mass spectrometry of inorganic compounds; instruments and techniques; theory and correlation of mass spectra; mass spectra and analysis; and ionization and dissociation. Under these major topics, parts of the papers discussed include field ionization mass spectroscopy; initial kinetic energy discrimination effects in crossed-field ion sources; mass spectrometric study of CaO and Ta; and spark source mass spectrometry as an analytical technique. The book also covers discussions on initial energy of hydrocarbon fragment ions; determination of the structure of alkaloids by mass spectrometry; and application of a time-of-flight mass spectrometer to the examination of ion-molecule interactions. This volume is a useful reference to students and researchers conducting work on mass spectrometry.
