Book excerpt: Menander (c. 341-291 BC) was the foremost innovator of Greek New Comedy, a dramatic style that moved away from the fantastical to focus upon the problems of ordinary Athenians. This collection contains the full text of 'Old Cantankerous' (Dyskolos), the only surviving complete example of New Comedy, as well as fragments from works including 'The Girl from Samos' and 'The Rape of the Locks', all of which are concerned with domestic catastrophes, the hazards of love and the trials of family life. Written in a poetic style regarded by the ancients as second only to Homer, these polished works - profoundly influential upon both Roman playwrights such as Plautus and Terence, and the wider Western tradition - may be regarded as the first true comedies of manners.

Book excerpt: There are numerous excellent reviews on fragment-based drug discovery (FBDD), but there are to date no hand-holding guides or protocols with which one can embark on this orthogonal approach to complement traditional high throughput screening methodologies. This Methods in Enzymology volume offers the tools, practical approaches, and hit-to-lead examples on how to conduct FBDD screens. The chapters in this volume cover methods that have proven to be successful in generating leads from fragments, including chapters on how to apply computational techniques, nuclear magnetic resonance, surface plasma resonance, thermal shift and binding assays, protein crystallography, and medicinal chemistry in FBDD. Also elaborated by experienced researchers in FBDD are sample preparations of fragments, proteins, and GPCR as well as examples of how to generate leads from hits. Book excerpt: This first systematic summary of the impact of fragment-based approaches on the drug development process provides essential information that was previously unavailable. Adopting a practice-oriented approach, this represents a book by professionals for professionals, tailor-made for drug developers in the pharma and biotech sector who need to keep up-to-date on the latest technologies and strategies in pharmaceutical ligand design. The book is clearly divided into three sections on ligand design, spectroscopic techniques, and screening and drug discovery, backed by numerous case studies.

Available in PDF, EPUB and Kindle. Book excerpt: A sentence of first-order logic is satisfiable if it is true in some structure, and finitely satisfiable if it is true in some finite structure. The question arises as to whether there exists an algorithm for determining whether a given formula of first-order logic is satisfiable, or indeed finitely satisfiable. This question was answered negatively in 1936 by Church and Turing (for satisfiability) and in 1950 by Trakhtenbrot (for finite satisfiability). In contrast, the satisfiability and finite satisfiability problems are algorithmically solvable for restricted subsets—-or, as we say, fragments—-of first-order logic, a fact which is today of considerable interest in Computer Science. This book provides an up-to-date survey of the principal axes of research, charting the limits of decision in first-order logic and exploring the trade-off between expressive power and complexity of reasoning. Divided into three parts, the book considers for which fragments of first-order logic there is an effective method for determining satisfiability or finite satisfiability. Furthermore, if these problems are decidable for some fragment, what is their computational complexity? Part I focusses on fragments defined by restricting the set of available formulas. Topics covered include the Aristotelian syllogistic and its relatives, the two-variable fragment, the guarded fragment, the quantifier-prefix fragments and the fluted fragment. Part II investigates logics with counting quantifiers. Starting with De Morgan's numerical generalization of the Aristotelian syllogistic, we proceed to the two-variable fragment with counting quantifiers and its guarded subfragment, explaining the applications of the latter to the problem of query answering in structured data. Part III concerns logics characterized by semantic constraints, limiting the available interpretations of certain predicates. Taking propositional modal logic and graded modal logic as our cue, we return to the satisfiability problem for two-variable first-order logic and its relatives, but this time with certain distinguished binary predicates constrained to be interpreted as equivalence relations or transitive relations. The work finishes, slightly breaching the bounds of first-order logic proper, with a chapter on logics interpreted over trees.

Available in PDF, EPUB and Kindle. Book excerpt: This volume was created initially from a symposium of the same name presented at the International Primatological Society's XVIII Congress in Adelaide. South Australia. 6-12 January 2000. Many of the authors who have contributed to this text could not attend the symposium. so this has become another vehicle for the rapidly growing discipline of Fragmentation Science among primatologists. Fragmentation has quickly become a field separate from general ecology. which underscores the severity of the situation since we as a planet are rapidly losing habitat of all types to human disturbance. Getting ecologists. particularly primatologists. to admit that they study in fragments is not easy. In the field of primatology. one studies many things. but rarely do those things (genetics. behavior. population dynamics) get called out as studies in fragmentation. For some reason "fragmentation primatologists" fear that our work is somehow "not as good" as those who study in continuous habitat. We worry that perhaps our subjects are not demonstrating as robust behaviors as they "should" given fragmented or disturbed habitat conditions. I had a colleague openly state that she did not work in fragmented forests. that she merely studied behavior when it was clear that her study sites. everyone of them. was isolated habitat. Our desire to be just another link in the data chain for wild primates is so strong that it makes us deny what kinds of habitats we are working in. However.

Book excerpt: The fragment molecular orbital (FMO) method is a fast linear-scaling quantum-mechanical method employed by chemists and physicists all over the world. It provides a wealth of properties of fragments from quantum-chemical calculations, a bottomless treasure pit for data mining and machine learning. However, there is no user-friendly description of its usage in the widely employed quantum-chemical open-source software GAMESS, nor is there any book covering the usage of GAMESS in general. This leaves very many interested users to their own devices to get through a variety of problems with very cryptic descriptions of keywords in the program manual and no guide whatsoever as to what options should be set for particular scientific tasks. This book is the panacea to many frustrations. The main focus of the book is to build a solid bridge connecting FMO users to GAMESS, by giving a helpful introduction of various FMO methods as needed for particular problems found in computational chemistry, and describing in detail how to do these simulations and understand the results from the output of the program. The book also covers parallelization strategies for attaining high parallel efficiency in massively parallel computations, and provides means to analyze performance and design a solution for overcoming performance bottlenecks. A special section is devoted to dealing with problems in executing GAMESS, arising from computational environment and user errors. Finally, 14 carefully selected types of applications are discussed in detail, describing the input keywords and explaining where to find the main results in the text-based output.

Available in PDF, EPUB and Kindle. Book excerpt: From its origins as a niche technique more than 15 years ago, fragment-based approaches have become a major tool for drug and ligand discovery, often yielding results where other methods have failed. Written by the pioneers in the field, this book provides a comprehensive overview of current methods and applications of fragment-based discovery, as well as an outlook on where the field is headed. The first part discusses basic considerations of when to use fragment-based methods, how to select targets, and how to build libraries in the chemical fragment space. The second part describes established, novel and emerging methods for fragment screening, including empirical as well as computational approaches. Special cases of fragment-based screening, e. g. for complex target systems and for covalent inhibitors are also discussed. The third part presents several case studies from recent and on-going drug discovery projects for a variety of target classes, from kinases and phosphatases to targeting protein-protein interaction and epigenetic targets.
