Despite this fact, no validated biological markers, or biomarkers, currently exist for measuring glutamate pathology in CNS disorders or injuries. Glutamate dysfunction has been associated with an extensive range of nervous system diseases and disorders. Problems with how the neurotransmitter glutamate functions in the brain have been linked to a wide variety of disorders, including schizophrenia, Alzheimer's, substance abuse, and traumatic brain injury. These conditions are widespread, affecting a large portion of the United States population, and remain difficult to treat. Efforts to understand, treat, and prevent glutamate-related disorders can be aided by the identification of valid biomarkers. The Institute of Medicine's Forum on Neuroscience and Nervous System Disorders held a workshop on June 21-22, 2010, to explore ways to accelerate the development, validation, and implementation of such biomarkers. Glutamate-Related Biomarkers in Drug Development for Disorders of the Nervous System: Workshop Summary investigates promising current and emerging technologies, and outlines strategies to procure resources and tools to advance drug development for associated nervous system disorders. Moreover, this report highlights presentations by expert panelists, and the open panel discussions that occurred during the workshop.

Recently, a novel form of GABAA receptor-mediated inhibition, termed “tonic” inhibition, has been described. Whereas synaptic GABAA receptors underlie classical “phasic” GABAA receptor-mediated inhibition (inhibitory postsynaptic currents), tonic GABAA receptor-mediated inhibition results from the activation of extrasynaptic receptors by low concentrations of ambient GABA. Extrasynaptic GABAA receptors are composed of receptor subunits that convey biophysical properties ideally suited to the generation of persistent inhibition and are pharmacologically and functionally distinct from their synaptic counterparts. This book highlights ongoing work examining the properties of recombinant and native extrasynaptic GABAA receptors and their preferential targeting by endogenous and clinically relevant agents. In addition, it emphasizes the important role of extrasynaptic GABAA receptors in GABAergic inhibition throughout the CNS and identifies them as a major player in both physiological and pathophysiological processes.

He persuaded me in 1960 to contribute a review to the Ergebnisse der Physiologie. As originally planned, it was to be relatively short. However, the interest and scope of the whole subject of synapses stimulated me to write a much more comprehensive and extensive account. I was not even then satisfied, particularly as so many new and attractive investigations and ideas were being evolved during and after the writing of this review; and during the writing of this book most interesting developments are occurring in so many centres of research. Through the kind cooperation of my friends I have been given the opportunity to quote and even to illustrate from these new and fascinating developments before their final publication. There would be some justification if the author were to make the claim that this book is the fruit of a life-time of enquiry into the physiology of synapses. In 1927 the subject of Excitatory and Inhibitory Synapses was chosen for investigation in the course leading to the Oxford D. Phil. But there have been such remarkable developments during the last 12 years that in this book very little reference will be made to work earlier than 1951 except in the historical introductions.

It has recently become clear that the strength of this first pain synapse is plastic and modifiable by several modulators, including neuronal and non-neuronal regulators, and studies on the fundamental processes regulating the plasticity of the first pain synapse have resulted in the identification of new targets for the treatment of chronic pain. It was a pleasure to greet the participants in the hope that their valuable contributions will make this meeting a tradition in this town. As one of the hosts of the symposium, I had the sorrowful duty to remind you of the absence of a dear colleague, Professor Graham God dard. His tragic and untimely death represents the irreparable loss of both a friend and an excellent researcher. This symposium is dedicated to his memory. If I compare the topics of the lectures of this symposium with those of the previous one, a striking difference becomes apparent. A dominating tendency of the previous symposium was to attempt to define hippocam pal function or to offer data relevant to supporting or rejecting existing theoretical positions. No such tendency is reflected in the titles of the present symposium, in which most of the contributions deal with hip pocampal phenomena at the most elementary level. Electrical, biochemi cal, biophysical, and pharmacological events at the synaptic, membrane, or intracellular level are analyzed without raising the question of what kind of integral functions these elementary phenomena are a part of.
