These isolates were physiologiacl characterized to test the more effeicient strains in nitrogen fixation, to select strains more adapted to environmetal stresses such as salinity, alkalinity, temperature. The adapted strains were selected to study the competitiveness of these strains against the standard strain CIAT 899 marked with gus gene under normal conditions, stress of salinity and stress of alkalinity. These strains were also characterized using molecular biology techniques such as REP-PCR, ARDRA of 16S and 23S rDNA, plasmid profiles analysis, sequencing of full length of 16S rDNA, hybridization with nifH gene and amplification of nodC gene. Strain EBRI 26 and Sinorhizobium meliloti strain 2010 were selected as salt tolerant strains to study the proteins involeved in salt tolerance by 2D proteom analysis and MALDI-Tof mass spectrometry.

Book excerpt: Inoculation of legumes with superior nitrogen fixing rhizobia does not always result in the desired increase because of the failure of the inoculation strain to occupy a significant proportion of the nodules. The introduced inoculant strains have to compete with the indiginous, often ineffective nitrogen fixing, rhizobial population at different levels: after their adaptation and multiplication in the soil and host rhizosphere, these strains have to compete on the legume roots for infection sites and nodulation. Although the success of inoculation depends for a great extent on the ratio of inoculant to indigenous rhizobia, the intrinsic characteristics of these rhizobia, and their responses to many abiotic and biotic environmental factors also determine the outcome of inoculation. Common bean (Phaseolus vulgaris L.) is an important crop in Latin America, supplying about 20% of the protein intake per person (Van Rheenen, 1982). The factors which influence the interaction between this legume and its microsymbiont are insufficiently known, and often no response is observed after inoculation of this crop. Recently, Rhizobium spp, such as R.tropici IIB, which nodilate a wide variety of tropical legumes, were indentified. Book excerpt: This study evaluated the effect of maize-legume intercropping on plant growth, N2 fixation, WUE, C assimilation and grain yield under field conditions in Eswatini. The 15N natural abundance and ureide assay were used to assess N2 fixation in the six test legumes (Bambara groundnut, cowpea, dry bean, groundnut, jack bean and soybean). Instantaneous water-use efficiency was estimated as the ratio of photosynthetic rate to stomatal conductance from gas-exchange measurements while the 13C natural abundance technique was used as an integrated estimate of WUE. The six test legumes obtained 39.06 – 70.19% of their N nutrition from symbiosis based on the 15N natural abundance technique, while the ureide assay showed that the legumes could obtain 16.46 – 55.79% of their N from symbiosis. The amount of N-fixed ranged from 12.66 to 66.57 kg ha-1 . In most instances, a high amount of N-fixed by legumes was accompanied by greater shoot DM and high grain yield, which indicates the importance of the symbiotic process in plant growth and grain yield. Plants that recorded high photosynthetic rates and leaf chlorophyll content also had greater WUE, and higher WUE was also associated with increased shoot DM and improved yields. The low shoot C:N ratio (12.67 – 21.44 g.g-1 ) of test legumes would support rapid decomposition. ERIC-PCR fingerprinting of isolates revealed high genetic diversity among the rhizobial symbionts of groundnut, jack bean and soybean from Eswatini. Phylogenetic analysis based on the 16S rRNA, housekeeping genes (atpD, dnaK, glnll and rpoB) and symbiotic genes (nifH and nodC) showed that the three legumes were nodulated by rhizobia belonging to the genus Bradyrhizobium. The groundnut microsymbionts were closely related to B. arachidis, B. iriomotense and B. canariense, while the jack bean and soybean microsymbionts were closely related to B. pachyrhizi and B. elkanii. Symbiotic characterization of bacterial isolates showed high variation in nodule number, nodule dry matter and shoot DM induced by rhizobia in their homologous hosts, name groundnut, jack bean and soybean. Some of the test isolates elicited higher relative symbiotic efficiency (RSE) than the commercial rhizobial strains, indicating that the indigenous rhizobia present in Eswatini soils can potentially be used in the development of inoculants. Interestingly, isolates which recorded high RSE clustered together in the single gene phylograms of the housekeeping genes and in the 16S rRNA phylogram. The findings of this study will assist in improving grain yield of maize in Eswatini through maize-legume intercropping.

Book excerpt: The association between root-nodulating bacteria and leguminous plants is advantageous due to their ability to alter atmospheric nitrogen into a useful form in a process known as biological nitrogen fixation (BNF). Research has shown that BNF is the most efficient way to supply the large amounts of nitrogen needed by plants to produce high-yielding crops. As a result, there have been numerous studies into the diversity and identity of the associated nitrogen-fixing bacterial symbionts. Recent advances in molecular microbiology together with the isolation of rhizobia from previously uninvestigated legumes have led to major revisions of rhizobial taxonomy, most notably the inclusion of bacteria from the? -Proteobacteria in the genera Burkholderia and Ralstonia. In this study, the diversity of root nodule bacteria associated with Phaseolus coccineus and Phaseolus vulgaris species in South Africa was investigated. A selection of rhizobial isolates were characterised by SDS-PAGE of whole cell proteins and rep-PCR DNA fingerprint analyses. These results were supplemented by partial 16S rDNA sequencing of a select number of isolates to confirm their identity. Where isolates displayed unexpected genus associations, partial nodA sequencing was performed to determine whether these were incidental contaminants or true nodulators. Based on 16S rDNA sequence analysis, the majority of isolates investigated were fast-growers belonging to the genus Rhizobium. A few isolates showed close relationship to species of the? -Proteobacteria genus, Burkholderia. Both the SDS-PAGE analyses and the combined rep-PCR analyses were able to resolve isolates down to strain level, but the comparison of the SDS-PAGE and 16S rDNA sequencing data confirmed that bacterial discrimination using SDS-PAGE is not useful at the genus level and higher, as isolates showing affinity to Burkholderia were mingled with isolates showing similarity to Rhizobium. These isolates were separate from the Rhizobium isolates in the combined rep-PCR dendrogram. While there were discrepancies between results obtained from SDS-PAGE and rep-PCR analyses, results from the combined rep-PCR analysis correlated with many of the results obtained in the SDS-PAGE analysis. Both geographic location and host plant species appear to have affected the grouping of isolates. Many clusters consisted of isolates from the same location or the same host plant species in both the SDS-PAGE dendrogram as well as the combined rep-PCR dendrogram. The nodA sequencing demonstrated that the majority of isolates tested contain the nodA gene indicating that they are capable of nodulation. There was a large strain diversity observed for the isolates of this study and a number of the root-nodulating bacteria of the Phaseolus spp. appear to constitute several novel nodulating genotypes.

(Latin American common bean) is a leguminous crop that is significant in the life histories of both human civilization and nitrogen-fixing bacteria called rhizobia. These associations are evident from the impacts they have left on the genome of the common bean. As a legume, the common bean has several genes that code for the initiation and maintenance of mutualistic endosymbiosis with rhizobia. Over the course of its domestication and cultivation, the common bean has also undergone simultaneous artificial selection and multiple bottlenecks. As it exists in commercial agriculture, crops of common bean lack within-population genetic diversity and are widely considered poor at facilitating nitrogen fixation. Recent studies suggest nitrogen fertilization to be key in the decline of nodulation in cultivars of common bean. In addition, preliminary research suggests that wild lineages of bean have greater numbers of nodules. However, these data do not directly compare proxies of nitrogen fixation, like nodulation, with genotype and the presence of nitrogen fertilizer. I hypothesized that bean accession would be a more significant factor than nitrogen presence in determining the ability to facilitate nitrogen fixation. Through a randomized block greenhouse experiment that observed the variables of accession and nitrogen treatment, I found that both nitrogen fertilization and genotype were significant contributors to root:shoot ratio, nodule number, and chlorophyll content index (CCI) in common bean. However, bean genotype was more significant in determining ability to facilitate nitrogen fixation.

All lines were rated as susceptible to all isolates even though they were selected based on the moderate resistance they showed in field trials. Thus new screening methods and more cultivars/lines need to be studied in order to develop a set of differentials. Genetic diversity of isolates collected in Puerto Rico, Honduras and the Dominican Republic over ten years from the two newly described subgroups AG 1-1E and AG 1-1F was assayed with four molecular markers. While some isolates were found to be clonal, Universal Rice Primers (URP) markers were the most informative showing differences between countries and years of collection for most of the isolates. This would support the hypothesis that population structure has changed over time and that there could be sexual recombination among a few genotypes. A group of 71 accessions from several species of Phaseolus including wild and landrace collections was tested for resistance to isolates from AG 1-1F, AG 1-1E and AG 1-1F. Fifteen lines were classified as moderately resistant to AG 1-1A but susceptible to the other two subgroups. Line 01-006-3B from P. coccineus was found to be resistant to AG 1-1F, intermediate to AG 1-1E and susceptible to AG 1-1A making it the best line to be considered for common bean breeding.
