Book excerpt: La thèse vise à élaborer une méthodologie permettant d’évaluer l’évolution microbienne possible de déchets solides, majoritairement minéraux et contenant une certaine fraction de matière organique, en fonction de scénarios donnés. Le travail propose d’abord une synthèse bibliographique des aspects fondamentaux de la biodétérioration de déchets essentiellement minéraux et la synthèse des procédures existantes d’évaluation bio-physico-chimique des déchets. Dans une deuxième phase, l’étude cherche à examiner la pertinence et l’adaptabilité des tests existants d’évaluation de la biodégradabilité et plus globalement de suivi du comportement des déchets dans différents scénarios de gestion (valorisation ou stockage). A partir de ces observations, des adaptations ou de nouvelles procédures ont été mises au point afin de proposer une méthodologie générale permettant d’évaluer la biodégradabilité de déchets majoritairement minéraux contenant une fraction plus ou moins importante de matière organique. Pour cela, cinq déchets minéraux différents de part leur nature, leur process de fabrication, leur état de maturation, ..., ont été sélectionnés. De nombreuses analyses physico-chimiques (analyses élémentaires des majeurs et mineurs, analyses de la fraction carbonée, essais de lixiviation, essais ANC, ...) et biologiques (analyses quantitatives de la microflore et de sa biodiversité, essais de comestibilité, de biodégradabilité intrinsèque et de biolixiviation) ont été effectuées afin de caractériser le plus largement possible ces déchets. Cette étude montre clairement que la réalisation de l’ensemble de ces essais n’est pas indispensable pour étudier le comportement d’un déchet dans un scénario donné mais aussi que deux déchets a priori assez similaires peuvent se comporter différemment. Le choix des essais à réaliser doit donc être fait en fonction des objectifs recherchés et adapté à chaque déchet en fonction du scénario de gestion. La synthèse de ce travail montre que dans la plupart des cas, les essais de longues durées sont à privilégier car l’activité microbienne nécessite des phases d’adaptation parfois très longues (par exemple, une production de méthane a été mise en évidence après 500 jours d’incubation). Bien que le temps est important dans le développement de l’activité microbienne, l’analyse statistique des données a montré qu’il ne jouait pas le rôle principal dans la variabilité des résultats obtenus mais que les différences entre les déchets et principalement les conditions d’incubation testées (incubations sur solide, en suspension, en aérobiose ou anaérobiose, avec ou sans ajout d’éléments nutritifs, ...) étaient prépondérants. Dans les déchets minéraux, l’activité microbienne n’est généralement pas favorisée par les conditions très alcalines du milieu et le manque de ressources nutritives (notamment de matière organique). Néanmoins, selon les conditions d’incubation, l’analyse moléculaire de l’ADN des populations par DGGE montre que des populations sont très bien établies et se développent en fonction des modifications physico-chimiques des matrices. Ces modifications peuvent être liées directement ou indirectement à l’activité des micro-organismes et entraîner une augmentation de la solubilisation des éléments métalliques polluants (Pb, Cu, Cr, Zn). L’étude a en effet montré que l’évolution physico-chimique de ces déchets influence leur évolution microbiologique et inversement.
