Book excerpt: L'étude du micro-environnement apparaît comme un défi majeur tant dans un contexte de régénération osseuse que dans le développement de nouvelles thérapies ciblant les tumeurs osseuses primitives. Avec plus d'un million de greffes réalisées chaque année en Europe, l'os est le tissu le plus transplanté. Afin de traiter un certain nombre de ces affections osseuses, des avancées cliniques telles que la régénération, ont pu être développées. L'approche combinatoire, cellules souches mésenchymateuses (CSM) et biomatériaux de phosphate de calcium (BCP), a montré son potentiel dans différentes études in vitro et in vivo. Néanmoins, le mécanisme d'ostéoinduction est à ce jour inconnu. Les travaux présentés dans cette thèse identifient les populations cellulaires, macrophages et ostéoclastes, intervenant dans le processus de régénération osseuse à partir des CSM mais aussi d'un autre type cellulaire, les monocytes. Le développement de nouvelles molécules de traitement suivent, dans la majorité des cas, un même processus de recherche. Ces étapes demandent de nombreux tests ; dans un premier temps, sur des cellules primaires ou des lignées cultivées en 2D, et par la suite sur des modèles in vivo adaptés à la pathologie concernée. Malgré ce lourd processus, seul un très faible pourcentage de ces composés est mis sur le marché. La nécessité de créer un intermédiaire entre les 2 systèmes de tests in vitro et in vivo semble essentiel. Dans cette étude, nous avons ainsi mis au point un modèle de culture 3D miniaturisé de cellules engagées dans la différenciation ostéoblastique et ostéoclastique. Par la suite, ce modèle a été modifié pour l'étude des cellules d'ostéosarcome (tumeur osseuse primitive) et le développement de nouvelles molécules thérapeutiques. Pour cela, les cellules d'ostéosarcome ont été cultivées sur la structure 3D hybride constituée des CSM et BCP. Cet outil a été utilisé par la suite pour le test de différentes stratégies de vectorisation de molécules : le ciblage à l'os et le ciblage des cellules tumorales.

Ce travail de recherche, à forte composante méthodologique, a eu pour objectifs i) la modélisation qualitative et quantitative de la morphogenèse des propriétés texturales de l'ilion humain; ii) la compréhension des relations intimes entre enveloppe corticale – le "contenant" - et architecture structurale – le "contenu"; iii) l'extraction - de nature non invasive et à haute résolution - et l'interprétation biomécanique de la signature endo-structurale de différents éléments du squelette appendiculaire de Mammifères, actuels et fossiles. Basée sur un large échantillon d'images numériques de référence 2-3D, l'application de notre protocole analytique à des spécimens fossiles sélectionnés, représentant taxons très variés du point de vue de la taille et du style de vie-contraintes environnementales-comportement locomoteur et issus de contextes géo-chronologiques différenciés, confirme la potentialité informative de ce genre d'approche dans une perspective paléobiomécanique.

Nos résultats montrent que l'architecture etla nature de cette matrice permettent de guider la différenciation ostéoblastique descellules humaines mésenchymateuses issues de la moelle osseuse. L'organisationcellulaire en agrégats observée stimule les interactions cellulaires, et plusparticulièrement la formation de jonctions communicantes de type GAP et l'activitédes Connexines 43. Nous avons en également étudié la fonction des Pannexines 1et 3 dans la culture 3D. En conclusion, l'ensemble de nos travaux démontre que lesinteractions cellule-cellule constituent des événements majeurs dans cesmécanismes de régénération tissulaire. Les données cellulaires et expérimentalestémoignent de l'intérêt d'utiliser la totalité de la suspension de moelle osseuse pourfavoriser à la fois l'ostéoformation et la vascularisation du tissu.
