Book excerpt: The impacts of crop rotations and N fertilization on the activities of 12 enzymes involved in C, N, and P cycling was studied in soils of two long-term cropping systems at the Northeast Research Center(NERC) in Nashua and the Clarion-Webster Research Center (CWRC) in Kanawha, established 1979 and 1954, respectively, in Iowa. The activities of glycosidases ([alpha]- and [beta]-glucosidases, [alpha]- and [beta]-galactosidases), amidohydrolases (amidase, arylamidase, L-glutaminase, L-asparaginase, L-aspartase), and phosphatases (acid and alkaline phosphatases, and phosphodiesterase) were assayed in soils sampled in 1996 and 1997 from four replicated cropping systems taken in corn, soybean, oats, or meadow (alfalfa) that received 0 or 180 kg N ha−1 before corn. Crop rotations significantly affected enzymes activities in soils at the NERC site in both years, and glycosidases only in soils at the CWRC site. The effect of N fertilization was inconsistent. Greatest activity values were in multicropping systems in meadow or oats, and the least in continuous corn or soybean systems. Enzyme activities were significantly correlated with C[Subscript mic], and arylamidase and amidohydrolases significantly correlated with N[Subscript mic], in both years and sites. The amounts of N mineralized during 24 weeks of incubation at 30°C were significantly correlated with the activities of arylamidase and amidohydrolases at both sites. The activity of arylamidase was also significantly correlated with amounts of N mineralized in soils from six regions of North Central U.S. An alkaline hydrolysis method for determining the total N potentially hydrolyzable in soils was evaluated. It involves determination of the NH4-N produced by direct steam distillation of 1 g field-moist soil and 1M KOH, NaOH, or LiOH or with PO4-B3O-- buffer (pH 11.8) successively every 5 min for 40 min. Calculated total hydrolysable N (N[Subscript max] or N[Subscript o]) values differed among soils, ranging from 401 to 1667 mg kg−1 soil and accounted for 12-56% of organic N in the soils. The N[Subscript max] or N[Subscript o] values obtained with KOH or NaOH were significantly correlated with the values obtained by using selected N mineralization indexes, and with the activities of amidohydrolases and arylamidase in the soils.

Book excerpt: Studies were undertaken to investigate the long-term effects of lime application and tillage systems (no-till, ridge-till, and chisel plow) on soil microbial biomass C (C[subscript mic]) and N (N[subscript mic]) and the activities of glycosidases ([alpha]- and [beta]-glucosidases, [alpha]- and [beta]-galactosidases and [beta]-glucosaminidase); phosphatases (acid and alkaline phoshatases and phosphodiesterase); amidohydrolases (L-asparaginase, L-glutaminase, amidase, urease, and L-aspartase); and arylamidase at their optimal pH values. With the exception of acid phosphatase, which was significantly but negatively correlated, all other enzyme activities were significantly and positively correlated with soil pH values at four sites in Iowa. [Delta] activity/[Delta] pH values showed that among the enzymes studied [beta]-glucosidase, L-glutaminase, and acid phosphatase are the most sensitive to pH changes and could be used as tools for monitoring ecosystems health and function. The effect of crop rotations and N fertilization on [beta]-glucosaminidase activity and its relationship to N mineralization were studied in soils of two long-term field experiments in Iowa. The activity of [beta]-glucosaminidase was significantly affected by crop rotations and N fertilization, and was significantly correlated with C[subscript org] and N[subscript org], C[subscript mic], and N[subscript mic] in soils, and with cumulative N mineralized during 24 weeks of incubation at 30°C. Studies to evaluate the effects of 23 trace elements on the activity of [beta]-glucosaminidase in three Iowa surface soils showed that at 5 mmol kg−1 soil, the activity of this enzyme was inhibited by 18, and activated by 5, of the trace elements tested, with Ag(I) and Hg(II) being the most effective inhibitors. Also, the activity of this enzyme was significantly affected by tillage systems (no-till, chisel plow, and moldboard plow) and four residue placements (bare, normal, mulch, and double mulch). Other studies showed that the amounts of N mineralized in 56 surface soils, obtained from six states in the North Central region of the United States, by two biological and three chemical methods were significantly correlated with [beta]-glucosaminidase activity, and with organic C and total N. The amounts of N mineralized during 14 days of incubation of field-moist soils under waterlogged conditions at 30°C were the most significantly correlated with [beta]-glucosaminidase activity (r=0.86***).

Differences in litter composition between crop and tree litter, and microclimate modifications in alley cropping systems may introduce spatial variation in soil organic carbon (SOC) and nitrogen (N) fractions and soil microbial properties. The objectives of this study were: (1) to assess the effects of litter quality, soil properties and microclimate differences on soil C and N mineralization in established alley cropping systems; (2) to determine the spatial variability of soil C and N fractions in these systems; and (3) to evaluate the effects of these systems on spatial differences in soil microbial activity and functional diversity. To address the first objective, microcosm and litterbag experiments were conducted in 2001 and 2002, respectively. Bulk soils were collected from the 0-20 cm depth at three sites: a 21-yr old pecan (Carya illinoinensis)/bluegrass (Poa trivials) intercrop (Pecan site) in north-central Missouri, a 12-yr old silver maple (Acer saccharinum)/soybean (Glycine max) - maize (Zea mays) rotation (Maple site) in northeastern Missouri and a warm season grass restored prairie (MDC site) in southwestern Missouri. Seven litters with varying composition were collected including pecan, silver maple, chestnut and walnut leaf litter (tree litter) and maize, soybean and bluegrass hay residues (crop litter).
