Book excerpt: The work presented in this thesis describes the electrochemical study of ions and molecules of biological importance, both at the ITIES using liquid-liquid electrochemistry, and at the sample solution-PVC membrane phase boundary using potentiometric ISEs. Urea-/thiourea-functionalized calix[4]arene compounds are evaluated as anion selective receptors in PVC membrane ISEs. These ionophores have been characterized as HPO42- selective. Detection limits on the order of 10-4 to 10-5 M, and Nernstian slope values are reported for this anion. Selectivity evaluation revealed a non-Hofmeister selectivity sequence as ISEs remained selective to phosphate over a range of common interfering anions. Both calix[4]arene compounds were also characterized using liquid-liquid electrochemistry. These studies showed that anion transfer is assisted by the presence of the urea- or thiourea-calix[4]arene in the organic phase. Diffusion coefficients were calculated on the order of 10-6 and 10-7 cm2sec-1 in relation to the facilitated transfer of HPO42-. Association constants of 2 x 1010 and 2 x 1013 M-2 were estimated for the complexed HPO42- anion. The ITIES was employed for the study and detection of insulin, a protein of high physiological importance. A response to insulin was obtained down to 2 {uF06D}M concentration, and was significantly affected by the pH conditions and nature of the organic phase electrolyte employed. Based on the results obtained, it is suggested that the response is largely due to the interaction of positively charged insulin adsorbed at the interface with hydrophobic anions of the organic phase. Characterization of an acridine-functionalized calix[4]arene compound was also performed at the ITIES. This compound behaves as an ionophore for a range of anions and cations, based on the possible binding sites the structure contains for both types of interactions. Electrochemical instabilities observed in some cases may be attributed to the localization of the calix[4]arene compound at the interface, whereby the hydrophilic acridine moieties veer towards the aqueous phase. CV and impedance spectroscopy results showed that a reduction in ion-transfer resulted upon addition of dsDNA to the aqueous phase. Based on the arcridine functionality of the calix[4]arene compound, which is a well-known DNA intercalator, it is believed that adsorption of DNA at the interface via intercalation of the acridine moieties with dsDNA may account for the reduction in signal observed. These results provide a novel approach for the study of this biologically important substance.

QDs have two components: an inorganic core and an optically active coated shell. Moreover, surface coatings can be applied to QDs to modify the particle as needed for experiments. Hydrophilic coatings prevent leaking of metal cargo from the core, enhancing the solubility in biological contexts and bind molecules, such as receptor–ligands, antibodies, therapeutic, and diagnostic macromolecules for enhanced effects. Their high surface-to-volume ratio allows multiple functional groups to attach onto the surface of the particles at constant surface volume. Silicon-, gallium-, indium-, or germanium-based; cadmium-based; and carbon-based QDs have already been used in many applications, such as imaging probes for the engineering of multifunctional nanodevices.
