Dans le but d'apporter notre contribution au développement de ce type de supports, nous avons préparé une famille de supports à base de silice passivée avec une phase stationnaire de type polymère greffée par la bêta-cyclodextrine. Les cyclodextrines appartiennent à la famille des molécules dites cages car elles comportent au sein de leur structure une cavité indéformable et hydrophobe qui leur permet d'emprisonner ou d'encapsuler d'autres molécules. Notre choix s'est porté sur les poly(alkylamines) comme polymères. Ces polymères sont des polybases de structure relativement simple, qui présentent de fortes interactions avec les gels de silice. Nous avons utilisé la poly(éthylèneimine), la poly(allylamine) et la poly(vinylamine). Ce dernier polymère n'étant pas commercial, a été synthétisé. Ces polymères ont été fonctionnalisés par la bêta-cyclodextrine. Ces polymères greffés ont alors servi de matériaux de base dans la préparation de nouveaux supports pour la chromatographie liquide haute performance. Les phases stationnaires préparées nous ont permis de séparer des dérivés disubstitués du benzène (nitrophénol, acide nitrobenzoïque, crésol) et d'autres molécules telles que la caféine, la théophylline, l'indométhacine le mécanisme de rétention est basé non seulement sur la formation de complexe d'inclusion, mais aussi sur des interactions de type acide-base et des interactions stériques. Nous avons tenté d'établir une corrélation entre les caractéristiques physique et chimique de nos supports et leur comportement chromatographique. La quantité et la nature du polymère déposé, la teneur en cyclodextrine, ainsi que certains paramètres chromatographiques (la nature et la proportion de la phase éluante, longueur de la colonne...) ont été étudiés.

Book excerpt: La chromatographie à contre courant (CCC) est une technique de séparation qui utilise une phase stationnaire liquide et une phase mobile également liquide. Des systèmes biphasiques liquides sont mis en œuvre. Il est très difficile de maintenir stationnaire une phase liquide, ce qui explique que les colonnes de CCC soient des machines complexes avec des parties tournantes. En effet des forces centrifuges sont utilisées pour atteindre le résultat : une phase stationnaire 100% liquide. L’objectif de ce travail est de présenter l'intérêt qu'il y a à travailler avec une phase stationnaire liquide. Un premier chapitre présente la technique CCC, son principe, les appareils et les paramètres utilisés et les applications principales. Un second chapitre présente les propriétés des systèmes de solvants biphasiques. Un système liquide biphasique particulier est très souvent utilisé. C'est le système eau-acétate d'éthyle, méthanol-heptane. Des mélanges particuliers de ces quatre solvants ont été décrits, référencés de A à Z et appelés "Systèmes Arizona". La composition de chacune des phases de ces mélanges Arizona a été déterminée. Les changements de compositions ont été associés aux proportions volumiques initiales des quatre solvants et à la nature de l'alcane utilisé (pentane, hexane, heptane ou octane). La stabilité de ces systèmes à court et long terme a également été étudiée. Dans un troisième et dernier chapitre, l'intérêt d'une phase stationnaire liquide est exploité en l'utilisant pour faire des séparations chromatographiques d’une façon qui ne serait pas possibles avec une phase stationnaire classique (solide). Deux méthodes ont été explorées. La méthode dite d'élution-extrusion et la méthode dite à phase stationnaire mobile. La méthode d'élution-extrusion consiste à pousser la phase stationnaire liquide hors de la machine tout en conservant la séparation des solutés initiée dans la phase stationnaire par la phase d'élution initiale. La méthode à phase stationnaire mobile consiste à pousser les deux phases liquides dans la colonne CCC dans le même sens mais à des vitesses différentes. Ces deux méthodes ont permis de réduire le temps nécessaire pour récupérer le soluté test cholestérol avec une résolution à la ligne de base de 16 heures à une heure. Ce travail devrait permettre à l’utilisateur de la technique chromatographie à contre courant de mieux comprendre l'intérêt qu'il y a à travailler avec une phase stationnaire liquide

The physicochemical properties of ionic liquids can be "tuned" by the modification of their structural moieties. Understanding of structure-property relationship is important if one wants to introduce a desired property in ionic liquid for a specific application. The initial part of this thesis will discuss the understanding of structure-property relationship of dicationic ionic liquids. Ionic liquids with high thermal stability are useful in many applications from simple solvents for high temperature reactions to high vacuum lubricants for space applications. This thesis presents the design, synthesis, and characterization of different thermally stable dicationic ionic liquids. ILs with TGA thermal stabilities in the range of 330 to 467 °C and inverse gas chromatography stabilities up to 400 °C are reported. The thesis also provides an understanding of the thermal behavior of ionic liquids at higher temperatures. The first, in depth thermal decomposition study of dicationic ionic liquids by using electrospray ionization mass spectrometer is presented, and it discusses the mechanism of thermal degradation of ionic liquids and their degradation products at high temperatures. Two important applications of thermally stable ionic liquids, stationary phases for gas chromatography and solvents for high temperature reactions are discussed in this thesis. Applications of thermally stable dicationic ionic liquid gas chromatography stationary phases for the high temperature separation of high molecular weight compounds is presented. Applications of ionic liquid stationary phases in the separation of structural isomers of toxic environmental pollutants, compounds with different functional groups and polarities are reported. A fundamental investigation of the effect of structural modifications such as cations, linkage chains, and anions on the selectivities of ionic liquids is discussed. The last chapter of the thesis presents the application of thermally stable monocationic phosphonium ionic liquid as a solvent in a high temperature synthesis. The advantages of higher thermal stability and the catalytic properties of ionic liquid were utilized for the "rapid" deprotection of tert-butoxy amino acids and peptides at higher temperatures.
