The shortnose sturgeon (Acipenser brevirostrum LeSueur, 1818) is a vulnerable species that exists along the eastern coast of North America. The impact of heating rate (0.1, 0.2 and 0.25 °C/min) on the CTMax, associated hematological responses, stress response genes and oxygen consumption were determined. Heating rate did not alter the CTMax value of shortnose sturgeon. Neither heating rate nor thermal stress affected plasma sodium and chloride levels, or the expression of stress response genes, such as heat shock proteins. However, thermal stress (regardless of heating rate) increased both plasma potassium and lactate concentrations. Glucose levels were increased at heating rates of 0.2 and 0.25°C/min, but not at 0.1°C/min. Book excerpt: The environment during early life history strongly impacts phenotypic development in all organisms, which further influences developmental trajectory and ecological fitness later in life. Depending on the developmental stage and magnitude of change in the environment, phenotypes may become irreversible and thus have a long-lasting effect later in life. This thesis was designed to better understand how changes in the environment may influence plasticity and variation of metabolic phenotypes of Lake Sturgeon (Acipenser fulvescens) within the first year of life. Broadly speaking, the thesis tested two hypotheses that 1) all measured phenotypes would be plastic; and 2) durations of environmental effects on phenotypic development would be correlated with distinct developmental windows. Studies were developed to examine 1) short-term effects of temperature or diet on metabolic phenotypes such as metabolic rate, energy density, fatty acid profiles, and growth (Chapters 2 and 3) and 2) longer-term effects of temperature or diet during early life on these metabolic phenotypes (Chapters 4, 5 and 6). The first experimental chapter (Chapter 2) examined ontogenetic development of metabolic rate and demonstrated that dietary shifts between Artemia to bloodworm resulted in cessation of growth with elevated routine metabolic rate. Chapter 3 examined how fatty acid profiles and plasma cortisol concentration were influenced by environmental temperature and showed that decreasing temperature led to increases in mono- and polyunsaturated fatty acids in both phospholipids and triglycerides, and food deprivation resulted in lack of difference between baseline and peak cortisol concentrations. Chapter 4 examined how temperature during early life influenced plasticity of growth and showed that temperature post-dietary transition resulted in a transient effect on growth and energy metabolism without long-term effects post-winter. Chapter 5 examined how temperature during early life could influence growth and fatty acid metabolism when fish were exposed to colder temperatures later in life and demonstrated that elevated temperatures resulted in a longer-term effect on growth but lack of transcriptional responses of desaturating fatty acids when exposed to a cold temperature (3.5°C) later in life. The final experimental chapter, Chapter 6 examined longer-term effects of diet at the onset of exogenous feeding on metabolism and growth and demonstrated that an enriched diet resulted in prolonged effects on growth, digestive enzyme activity and survival prior to a simulated overwintering. This doctoral thesis research revealed that all measured metabolic phenotypes were plastic, but subtle changes in temperature and diet during early life history resulted in transient or prolonged effects on growth and metabolism in age-0 lake sturgeon. Results will aid our understanding of cohort and population dynamics as well as contribute to the development of conservation strategies for lake sturgeon, a species at risk or endangered across its natural range.

The first study describes metabolic, hormonal and haematological effects during a 24 h salinity challenge. The second study examines the potential whole-animal effects elicited by salinity exposure via recovery from exercise. The results of these experiments show that, while juvenile shortnose exhibit hydromineral imbalance (i.e. water loss, salt gains) and metabolic depression in high salinity in the first 24 h, these changes have little effect on the animals' ability to recover from exhaustive exercise. This is interesting as hydromineral imbalances have been shown to negatively affect swimming in other fishes. Temperature, dissolved oxygen and substrate are all important factors for developing metabolic phenotypes in fish. In this thesis, effects of temperature, dissolved oxygen (DO) and substrate on standard metabolic rate (SMR), forced maximum metabolic rate (FMR), metabolic scope (MS), energy density (ED), enzyme activity associated with ATP regeneration (pyruvate kinase, lactate dehydrogenase and cytochrome c oxidase), condition factor (K), hepatosomatic index (HSI) and critical thermal maxima (CTmax) of developing age-0 Lake Sturgeon, Acipenser fulvescens, including a simulated overwintering event, were examined. This research revealed that temperature, dissolved oxygen and substrate all significantly influenced development of metabolic phenotypes in Lake Sturgeon. While reduced temperature significantly increased SMR, increased temperature substantially decreased the survival rate. Lower DO (80%) did not impact FMR and MS in the short term, however, prolonged exposure to mild hypoxia, particularly during the stressful condition of overwintering, significantly reduced FMR and MS. Inclusion of substrate was found to significantly enhance K and SMR, and it was suggested that substrate be used at the early yolk-sac stage to enhance growth and survival. Further, a strong linear relationship between dry to wet mass ratio and ED was found, which can be used practically to estimate energetic status of Lake Sturgeon. This research provides information regarding environment-phenotype interaction during the first year of life in Lake Sturgeon. This information will contribute to a better understanding of critical factors influencing the successful recruitment of populations that are endangered throughout most of their natural range.

Understanding such effects is particularly important for sedentary species that cannot make large-scale movements in response to changes in environmental conditions. I examined how metabolic rates responded to temperature in the bluebanded goby (Lythrypnus dalli) and the Woolly Sculpin (Clinocottus analis). Oxygen consumption and metabolic rate increased significantly with temperature for both species, but L. dalli was much more sensitive to the effects of temperature than C. analis (Q 10 values were 5.21 and 3.02, respectively). Thermal fluctuations likely play a significant role in the ecology of these species and continued increases in seawater temperature will either necessitate an increase in foraging and food consumption or drive costly trade-offs between metabolism and processes such as growth and reproduction.

Book excerpt: "Climate change has emerged as an increasingly critical threat to freshwater systems that may affect freshwater fishes through direct effects on their thermal physiology, but also indirectly by altering the magnitude of biotic interactions and/or the relative abundance of co-habiting species. Understanding the direct and indirect effects of temperature represents a significant knowledge gap that may be vital to effective conservation strategies for freshwater fishes. This is especially true for already imperiled species because these taxa tend to be more sensitive to changes in their environment and are already contending with other, and potentially interactive, stressors. In this thesis, I used a combination of ecophysiology and field-based techniques to examine effects of elevated water temperature on Pugnose Shiner, Notropis anogenus, an endangered fish species in Canada. In Chapter 1, I investigated effects of temperature on critical thermal maximum (CTmax) and standard metabolic rate (SMR) in juvenile Pugnose Shiner reared for 4 months across five different temperatures. CTmax was measured under normoxia and acute exposure to hypoxia to test for oxygen sensitivity of upper thermal limits in this species. CTmax increased with elevated water temperature. Agitation temperature (temperature at which fish show behavioural signs of thermal stress) also increased with rearing temperature and occurred on average 4.32°C above acclimation temperatures. CTmax was lower when fish were acutely exposed to hypoxia during CTmax trials and showed no increase between 19 and 25°C. Surprisingly, CTmax under acute hypoxia increased sharply at the highest temperature. SMR increased between 16 and 25°C, and then declined significantly at 28°C, indicating that the ability of Pugnose Shiner to reverse thermodynamic effects on SMR is limited and there was no evidence of long-term thermal compensation. Interestingly, gill size (e.g. total gill filament length, hemibranch area) increased with long-term exposure to high temperature, which may increase oxygen uptake capacity and help to fuel increased metabolic demands. In Chapter 2, I explored the role of temperature, other abiotic factors, and co-occurring species in shaping the fine-scale distribution of Pugnose Shiner within Thompson’s Bay in the upper St. Lawrence River, Ontario. Multivariate analyses and occupancy modelling were used to assess how environmental variables and species co-occurrence accounted for variation in the distribution of Pugnose Shiner. Environmental variables, including temperature, were significant predictors of community composition. Pugnose Shiner was associated with clear, cool sites far from the inner bay with high vegetation density, including Chara vulgaris. Pugnose Shiner was negatively associated with Brook Silverside (Labidesthes sicculus), Largemouth Bass (Micropterus salmoides), Pumpkinseed (Lepomis gibbosus), and juvenile Lepomis spp. This has important conservation implications because some native centrarchids are increasing in abundance with climate warming, which may threaten persistence of rare and imperiled cyprinids, including Pugnose Shiner. Although Pugnose Shiner showed little evidence of thermal compensation (SMR increased with acclimation temperature), it did show acclimation capacity in terms of thermal tolerance and increased gill size. Captive rearing may provide opportunities for thermal hardening in reintroduction efforts, and targeted hunting of expanding species may buffer Pugnose Shiner from the negative effects of elevated water temperature. Overall, my research highlights the importance of examining multiple traits at multiple scales of biological organization to quantify effects of elevated water temperature on freshwater fishes and guide conservation recommendations for imperiled species"--
