Obesity (Body Mass Index> 30 kg/m2) is an inflammatory disease that has its origin in diet. Obesity is the condition of accumulation of lipids in the adipose tissue and in non-adipose tissues such as the liver and muscle (eg. steatosis). The WHO-coordinated Cardiovascular Diseases and Alimentary Comparison Study (CARDIAC), a worldwide epidemiological study, found that those people who consumed soy had a significantly reduced risk of obesity than those who did not consume soy in their diets. The effects of fermented soy, soy, and casein diets on body weight of adipose mass and lipid composition of the liver in obese Zucker rats was studied. Thirty-nine male Zucker fatty rats, which are prone to hyperphagia, obesity, and insulin resistance, were randomized and placed in four groups, each group being fed a different diet. The diets were: 1) Casein diet (n=9); 2) Soy diet (n=10); 3) Fermented soy diet (n=10); 4) Mixture of soy and fermented soy (n=10). The differences in weight gain, food intake, changes in adipose mass and fat accumulation in the liver were examined. After feeding diets for ten weeks, the rats were euthanized and necropsy was performed. Weights of the removed organs were recorded. There were no effects of diets on food intake, body weight or adipose mass. All three soy diet groups had a significantly lower liver weight and liver to body weight ratio compared to the casein diet group. The casein diet group also had a significantly lower percentage of polyunsaturated fatty acids in the livers than both the soy and fermented soy diet groups. However, the casein diet group had a significantly higher percentage of monounsaturated fatty acids in the livers than all three of the soy diet groups. Both the fermented soy and soy diet groups had significantly higher amounts of omega-6 fatty acids in the livers than the casein diet group. Overall, fermented soy and soy diets did have an effect on hepatic lipid metabolism in Zucker fatty rats when compared to the casein diet.

Book excerpt: Dietary fat has long been implicated in the etiology of metabolic and cardiovascular disease, and both the amount of fat and the fatty acid composition of the diet play a role in disease progression. Although national health organizations have set guidelines for the recommended intake of dietary fats, questions remain regarding the optimal dietary lipid profile for maintaining health and improving disease conditions. Whether certain types of fatty acids from plant-based oils can improve metabolic and vascular disease has been studied and debated, but not fully determined. In this study, we investigated the role of dietary fatty acids from plant-based oils, and examined their effects on metabolic and vascular disease parameters. Obese fa/fa Zucker rats were fed a diet containing flaxseed oil, which resulted in smaller adipocytes and decreased adipose tissue T-cell infiltration. Obese-prone Sprague Dawley rats were fed high-fat diets with different proportions of mono- and polyunsaturated fats. Changes were observed in adipose tissue levels of fatty acid synthase, adiponectin and fatty acid receptors GPR41 and GPR43, but other metabolic and inflammatory mediators in adipose tissue and serum remained stable. A systematic review and meta-analysis on the impact of n3 fatty acids on major cardiovascular endpoints showed that little evidence exists to support their role in peripheral arterial disease. Then again, very few studies on this topic have been conducted. To address this research gap, a clinical trial was designed to investigate the effects of a dietary intervention on blood vessel properties in people with peripheral arterial disease. Participants in the Canola-PAD Study consumed 25 g/day of canola oil or a Western diet oil mixture as part of their usual diet for 8 weeks. Although the intervention altered phospholipid fatty acids, vascular function, the lipid profile and inflammatory markers stayed relatively stable. Overall, this research demonstrates that dietary fatty acids from plant-based oils can be immunomodulatory, but at the physiological doses tested they are not potent mediators of functional changes in obesity or vascular physiology.

As such, this study examined the metabolic effects of a plant-based n3PUFA, stearidonic acid (SDA), in polygenic obese rodents. Methods, Lean (LZR) and obese Zucker (OZR) rats were provided either a standard westernized control diet (CON) with a high n6PUFA to n3PUFA ratio (i.e., 16.2/1.0) or experimental diet modified with flaxseed (FLAX), menhaden (FISH), or SDA oil that resulted in n6PUFA to n3PUFA ratios of 1.7/1.0, 1.3/1.0, and 1.0/0.8, respectively. Results, After 12 weeks, total adiposity, dyslipidemia, glucose intolerance, and hepatic steatosis were all greater, whereas n3PUFA content in liver, adipose, and muscle was lower in OZR vs. LZR rats. Obese rodents fed modified FISH or SDA diets had lower serum lipids and hepatic fat content vs. CON. The omega-3 index (i.e., ÓEPA + DHA in erythrocyte membrane) was 4.0, 2.4, and 2.0-fold greater in rodents provided FISH, SDA, and FLAX vs. CON diet, irrespective of genotype. Total hepatic n3PUFA and DHA was highest in rats fed FISH, whereas both hepatic and extra-hepatic EPA was higher with FISH and SDA groups. Conclusions, these data indicate that SDA oil represents a viable plant-derived source of n3PUFA, which has therapeutic implications for several obesity-related pathologies.
