Book excerpt: A 61-day study was designed to assess the effects of the level of dietary linolenic acid (LNA) and its ratio to linoleic acid (LA) on plasma lipoproteins (LP) and the fatty acid composition of the platelet phospholipids (PL) in twelve normolipidemic men. The study consisted of four diet periods: a 7-day pre-emperimental period and two 18-day experimental periods separated by an 18-day washout period. Diets supplied 53% of total energy as carbohydrate, 14% as protein and 33% as fat. Added fat accounted for 79% of the total fat or 29% of total energy. A mixture of fats (MF) was provided during the pre-experimental and washout periods. Subjects were randomly assigned to receive two or three experimental diets containing: i) 100% low linolenic canola oil (LLNA), ii) 85% regular canola oil and 15% sunflower oil (CAN), or iii) 67% regular canola oil, 15% flax oil, and 18% sunflower oil (FLAX). The experimental diets provided similar amounts of LA but different amounts of LNA. The LA/LNA ratios and the LNA levels of the diets were 6.3, 4.3 and 2.5, and 4%, 6% and 11% respectively. Fasting 12-hour blood samples were analyzed for plasma lipids and LP. Book excerpt: This study investigated the effect of a-linolenic acid enriched virgin olive oil vs. canola and virgin olive oil on body composition, serum lipids, and lipid peroxidation and adipose tissue lipoprotein lipase activity, in terms of their relation to atherosclerosis. Twenty Six Sprague Dawley rats were fed ad-libitum three types of diets, each containing 40% of dietary calories from fat being either canola, virgin olive, or a-linolenic acid enriched virgin olive oil, for seven weeks, with free access to water. At the end of feeding period, the rats were killed by decapitation and blood samples were analyzed for serum triglycerides (TG), total and HDL-Cholesterol (TC and HDL-C), glucose, insulin, and by products of lipid peroxidation, malondialdehyde (MDA) and 4-hydroxy-2(E)-nonenal (4-HNE). Carcasses were analysed for water, fat. and protein content. Individual fat pads were dissected out and weighed. Fat cell number, tissue homogenate and heparin releasable adipose tissue lipoprotein lipase activity were assayed from the right epididymal pad. Weight and fat content was determined for feces and liver. All groups showed no significant difference in the nutritional parameters. Results showed that total cholesterol and triglycerides were higher in the virgin olive oil group compared to canola and enriched virgin olive oil that were similar. HDL-C/total cholesterol ratio was not different between the canola and enriched virgin olive oil group but was significantly lower in the virgin olive oil group. Liver percent fat was significantly higher in the virgin olive oil fed rats as compared to the canola oil fed rats. The higher content of oleic acid and saturates in virgin olive oil may have enhanced the accumulation of fat in the liver. The above results suggest that a-linolenic acid enrichment of virgin olive oil can mimic the effect of canola oil on blood lipids, hence could be responsible for reducing triglycerides and total cholesterol, and increasing HDL-C/total cholesterol ratio when compared to virgin olive oil. The effects of a-lino!enic acid may be explained by its conversion into EPA, which has been previously reported to affect plasma lipids. Further investigations are needed to elucidate the effect of a-linolenic acid from vegetable oils before recommending to include it in daily diet.
