Chronic dietary supplementation with polyphenols, from pomegranates, has been shown to accelerate recovery following eccentric exercise, but the optimal dose is unknown. The purpose of this study was to determine the effect of dietary supplementation with different doses of pomegranate juice concentrate (PJC) on muscular strength, power, and soreness throughout a 96-hour time period following an acute bout of eccentric exercise. Healthy recreationally active males (n=45) were assigned to one of three treatment groups: Once-daily PJC (1x), twice-daily PJC (2x), or placebo (PLA) supplementation over a period of eight days. A 1x dose of PJC provided approximately 650 mg GAE. On day four of each treatment, subjects performed downhill running intervals ( -10% grade) over a 40-minute period followed by 40 repetitions of eccentric elbow flexion at 100% of concentric 1-RM. Muscle soreness of arms and legs, maximal isometric strength of the elbow flexors (EF) and knee extensors (KE), vertical jump height (VJ[subscript height]) maximal cycling power (P[subscript max]), and 10-meter sprint velocity (V[subscript 10m]) were assessed pre-exercise and 2, 24, 48, 72, 96 hours post-exercise. Additionally, maximal instantaneous power (IP[subscript max]), maximal velocity (V[subscript max]), maximal torque (T[subscript max]), and torque at 0° (T0) were assessed on the inertial load power cycle pre-exercise and 24, 48, 72, 96 hours post-exercise. Book excerpt: Abstract: Exercise-induced muscle damage (EIMD) occurs following strenuous and/or unaccustomed exercise including resistance training. EIMD is associated with elevated blood creatine kinase (CK) concentrations, limitations in range of motion (ROM), a reduction in muscular power, and delayed onset muscle soreness (DOMS). EIMD may negatively affect training quality and performance, thus methods to mitigate EIMD may be useful for resistance training populations including both athletes and recreationally active individuals. Omega-3 (n-3) fatty acids, particularly eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), are known for their anti-inflammatory and anti-nociceptive properties. The current literature on supplementation with n-3 fatty acids and EIMD is not conclusive, however, there are data showing supplementation can ameliorate EIMD and facilitate recovery in both untrained and resistance-trained individuals following resistance exercise, particularly at higher doses. Presently, a characterization of efficacious doses of n-3 supplementation is currently lacking. Therefore, the purpose of this study was to investigate the effect of 33 days of six and eight grams of n-3 supplementation on markers of muscle damage and recovery after an acute bout of eccentric resistance exercise in resistance trained males. It was hypothesized that both six and eight grams of n-3 supplementation would attenuate EIMD and promote recovery following eccentric resistance exercise. In this double-blind randomized control study, 26 resistance trained males (23 ± 4 years; 173.6 ± 20.5 cm.; 81.9 ± 9.7 kg; 14.2 ± 3.7% BF) supplemented with six grams of n-3 fatty acids (n=10), eight grams n-3 fatty acids (n=7), or placebo (n=9) for 33 days total. On day 30, participants completed an acute bout of eccentric resistance exercise (10 sets of 8 repetitions barbell back squat at 70% 1RM with a four second eccentric, followed by 5 sets of 10 repetitions per side body weight split squat jumps). Participants were instructed to refrain from additional recovery methods post-exercise. Outcome measures included vertical jump height (VJH), perceived muscle soreness (PMS), hip and knee range of motion (ROM), repetitions to fatigue (RTF) at 70% 1RM barbell back squat, and serum creatine kinase (CK). Vertical jump height, PMS, hip and knee ROM, and capillary blood samples were taken immediately pre-exercise bout (day 30), 1 day (1d), 2 days (2d) and 3 days (3d) post-exercise, while number of RTF were completed at the familiarization session (day 21, 22, or 23) and 3d post-exercise. There was a significant difference in VJH from pre to 1d post-exercise (p=0.009, 1d to 2d (p=0.02), and 2d to 3d post-exercise (p=0.002) in all groups. Perceived muscle soreness significantly increased from pre to 1d, 2d, and 3d hour post-exercise (p 0.05), and significantly decreased at 3d post-exercise compared with 1d and 2d (p 0.05) in all groups. Results failed to show a significant difference in subjective DOMS ratings between the two groups following supplementation. DOMS ratings' were significantly lower for both groups after performing the second exercise trial compared to the first. Book excerpt: Unaccustomed eccentric exercise can lead to decreases in muscle force production, increases in soreness, swelling, stiffness, and discomfort. The purpose of this study was to test the dose response of pomegranate juice concentrate on serum markers of muscle permeability, inflammation, and total anti-oxidant status. To test this hypothesis, 45 healthy recreationally active males (22.3 ± 0.5 y, 73.8 ± 1.71 kg, 174.9 ± 0.9 cm) were recruited from the local Austin community for participation in this study. Subjects were disqualified from participation in the study if in the past 6 months they were engaged in an exercise training program. Subjects were placed into either the placebo group, the once-daily, or twice-daily pomegranate juice concentrate supplementation group. Subjects performed a total of 8 days of supplementation. On day 4, all subjects came to the laboratory and underwent an eccentric exercise protocol consisting of 2 minutes of downhill running at -10% grade at 7.5 mph repeated 10 times, resulting in ~20 minutes of total downhill running. Thereafter, subjects performed 50 eccentric elbow extensions each lasting 5 seconds using a weight equal to their concentric one-repetition maximum. Blood measures were made pre-exercise (baseline), and 2, 24, 48, 72, and 96 hours post exercise and analyzed for interleukin-6, creatine kinase, myoglobin, and total anti-oxidant status. Creatine kinase was significantly elevated at 96 hours post exercise, but with no significant differences between treatments. Myoglobin was significantly elevated above baseline at 2 and 96 hours, but with no differences between treatments. There was no effect for time or treatment on the total anti-oxidant status of the serum. Il-6 was significantly higher at 2 and 24 hours after exercise, but with no difference between treatments. The percent increase in interleukin-6 from baseline was significantly lower in the twice-daily POM supplementation group versus placebo (124.3 ± 9.4, 188.6 ± 16.0% of baseline; respectively) during the 2-96 hours following eccentric exercise, but no statistical difference between 1x and 2x or 1x and placebo was observed. This suggests that 8 days of supplementation with pomegranate juice concentrate twice a day significantly reduces the percent increase in a marker of inflammation (interleukin-6) during the 96 hours following eccentric exercise; however, neither supplement was different than the placebo in regards to all other measures.

DOMS affects athletic performance, therefore therapy is of interest to athletes. The purpose of this study was to examine the effects of localized vibration (biomechanical muscle stimulation, BMS) on biomarkers of DOMS following eccentric cycling, and to test changes in muscle length and soreness. We investigated if BMS reduces DOMS and enhances muscle function following eccentric exercise and if creatine kinase (CK), lactate, and pain were affected. Twenty-eight recreationally active men (18 - 40 years old) were randomized into control or BMS groups. Subjects performed 5 minutes of eccentric cycling, then either received BMS, or rested. Outcome variables, plasma creatine kinase, blood lactate, pain scores (Likert), muscle length, muscle pain (self-reported), and pressure algometry were collected at four intervals (baseline, pre-cycling, 24 hours post-cycling, and 48 hours post). A main effect of time was found for quadriceps pain threshold (F(3, 78) = 3.02, p = 0.04). A main effect of time was found regarding an increase in lactate (p = .016; T0 to T1) and decrease in lactate (p = .025; T1 to T3). A main effect of time for Likert pain score was found pre-cycling versus post-cycling (p = .0001). There were no significant interactions between group (BMS or control) and time. This study does not support our hypotheses regarding localized vibration and recovery from DOMS. However, we found significance regarding a main effect of time for pain threshold in the quadriceps, Likert pain score, and lactate level.

DOMS is associated with impaired muscular performance. Dietary interventions may reduce inflammation and improve physical performance. The first aim of this investigation was to determine if Acai supplementation reduces markers of oxidative stress and the inflammatory process caused by eccentric exercise. The second aim was to determine if Acai supplementation reduces muscle soreness and improves muscle function. Individuals were counterbalanced/stratified into the Acai group or a placebo group. Supplementation started 48 hours prior to downhill running. Markers of oxidative stress and inflammation, and range of motion, muscle soreness perception, agility, and vertical jump displacement were assessed at baseline, after, 24, and 48 hours after downhill running. Twenty collegiate athletes and non-athletes (21±2 years old) completed the protocol. The Acai group (N = 10) reported significantly less muscle soreness in the quadriceps muscle (p = .011) compared to the placebo group (N = 10). In addition, there was a significant difference (p = .023) in the group by time interaction in the quadriceps muscle soreness. Furthermore, the Acai group scored slightly different on range of motion, agility, vertical jump displacement, creatine kinase, and c-reactive levels compared to the placebo group throughout all 4 time periods. Acai has demonstrated to be an effective supplement to decrease quadriceps muscle soreness after downhill running. Furthermore, the consistency of results throughout the 4 time periods may suggest its potential to slightly change performance and levels of oxidative stress and inflammation.
