Book excerpt: This book covers the current understanding of the role of activation-induced cytidine deaminase (AID) in the generation of antibody response to antigenic challenge. Since the discovery of AID, and the genetic demonstration of its role in somatic hypermutation and class-switch recombination of antibody genes, much has been learned about the biochemistry of this enzyme. However, some key questions remain hotly contested, such as: how does this enzyme get to the antibody locus leaving the rest of the genome intact, and why are DNA repair pathways which normally repair deamination events co-opted into actually fixing mutations into the genome? These questions, among others, will be addressed in this monograph from various perspectives. Being leading experts in their respective fields, the contributors of this highly valued title summarize current research in the field of AID and put forth hypotheses in order to provide a platform for future experiments./a

Book excerpt: Living cells have evolved many ways of coping with metabolic events and environmental influences that damage DNA. These mechanisms, and the frequent progression to cancer that results when they go awry, are rev iewed in this volume by authors from over sixty of the world's leading laboratories. The topics discussed include DNA repair, mutagenesis and other damage-tolerance functions, checkpoint control, apoptosis, and adaptation. They draw from studies on human and yeast cells. Current, but with a valuable historical perspective, this volume has the depth and lasting value typical of this most prestigious series and is esse ntial reading for investigators of DNA replication, cell cycle control, and tumorigenesis.

Book excerpt: In recent years, the field of Toxinology has expanded substantially. On the one hand it studies venomous animals, plants and micro organisms in detail to understand their mode of action on targets. While on the other, it explores the biochemical composition, genomics and proteomics of toxins and venoms to understand their three interaction with life forms (especially humans), development of antidotes and exploring their pharmacological potential. Therefore, Toxinology has deep linkages with biochemistry, molecular biology, anatomy and pharmacology. In addition, there is a fast developing applied subfield, clinical toxinology, which deals with understanding and managing medical effects of toxins on human body. Given the huge impact of toxin-based deaths globally, and the potential of venom in generation of drugs for so-far incurable diseases (for example, Diabetes, Chronic Pain), the continued research and growth of the field is imminent. This has led to the growth of research in the area and the consequent scholarly output by way of publications in journals and books. Despite this ever growing body of literature within biomedical sciences, there is still no all-inclusive reference work available that collects all of the important biochemical, biomedical and clinical insights relating to Toxinology. The Handbook of Toxinology aims to address this gap and cover the field of Toxinology comprehensively.

Book excerpt: The vertebrate immune system is distinctive among defense systems of multicellular organisms. In addition to nonspecific immunity, it generates a randomized array of millions of antigen receptors (immunoglobulins and T-cell receptors). A subset of these receptors are critical for binding to invading microbes or biochemicals from them to tag the microbes for elimination. Three site-directed DNA modification processes are critical to this process in vertebrates. V(D)J recombination generates the array of exons that encode the antigen binding pockets. Recent work summarized in this volume describes the dissection of this process at the biochemical level. The mechanism of the reaction is now understood in considerable detail. The proteins that catalyze many steps of the process have now been identified by biochemical and genetic recon stitution and by analysis of genetic mutants defective in V(D)J recombination. Class switch recombination is the process by which the variable domain exon of the heavy chain is changed from IgM to IgG, IgA. or IgE. Recent progress is described in the de velopment of an extrachromosomal substrate assay system. Molecular genetic analysis of the process in transgenics is defining some of the cis sequence requirements. Biochemical assays for defining enzymatic components are also described. In addition to exciting progress in V(D)J recombination and class switch recombination, one chapter describes recent pro gress in somatic hypermutation.

Available in PDF, EPUB and Kindle. Book excerpt: Ageing is of perennial interest as a universal feature in all human societies. The genetic background and biochemical bases of ageing processes are currently being revealed in unprecedented detail. It is emerging that one of the main hurdles to be overcome in achieving a long and healthy lifespan is the maintenance of a properly functioning immune system. The main cause of death in people who have achieved "successful ageing" (which mostly means not having succumbed to cancer or cardiovascular disease) is infectious disease, caused by immunosenescence. This book contains chapters by many of the leaders in the field of immune-related issues in ageing and remediation.

A wide spectrum of distinct CRISPR/Cas systems has been identified in at least half of the available prokaryotic genomes. On-going structural and functional analyses have resulted in a far greater insight into the functions and possible applications of these systems, although many secrets remain to be discovered. In this book, experts summarize the state of the art in this exciting field.

Stress response is a set of reactions that allow the organism to adjust and survive adverse conditions. Stress can be physical, such as extreme temperature, radiation, injury, or psychological, caused by perceived danger or deprivation. Every living cell has biochemical mechanisms to cope with physical stress. These mechanisms show a degree of similarity among several types of living organisms. Stress Response and Immunity: Links and Trade Offs explores the functional and evolutionary connections between stress response and immunity. The book introduces the reader to the concept of stress and subsequently examines the connection between stress response and immunity at various evolutionary stages of living organisms - from bacteria to humans. The book also features chapters dedicated to the role of tumor suppressor genes and the immune system of the brain. The information presented in this reference demonstrates the profound effects of physical and psychological stress on human health. Readers with basic knowledge of molecular biology will learn about the interesting facets of stress responses and the evolutionary trade offs observed in different life forms.
