Book excerpt: Beet curly top virus (BCTV), transmitted by the beet leafhopper (Circulifer tenellus), is the causal agent of curly top disease. The disease can cause substantial yield losses in processing tomato production in California, such as in the major outbreak in 2013. In this study, a loop-mediated isothermal amplification (LAMP) assay was developed to rapidly and specifically detect BCTV in infected leaf tissue and viruliferous leafhoppers. The LAMP primer set, ID1R4, was designed from a conserved region of the BCTV genome identified in 67 isolates. A master mix containing phenol red was used that allows visual identification of a positive (BCTV) detection due to a red to yellow color change in the LAMP assay following 40 min incubation at 65C. When used in this LAMP assay, the ID1R4 primer set mediated detection of five of the six BCTV strains tested, including the major strains that are associated with curly top disease of processing tomatoes in California. Compared with a qPCR test developed for BCTV in this study, the LAMP assay was 10-fold less sensitive for the detection of BCTV DNA in extracts prepared from infected tomato leaf tissue. A commercially available buffer (GEB2) was identified that allowed for rapid preparation of sap from infected leaves that could be used as a template for detection of BCTV in the LAMP assay. Extracts prepared from beet leafhoppers by grinding in sterile water and STE buffer were suitable for the detection of BCTV DNA in the LAMP assay. A tomato line, Line 20 #12, with resistance to BCTV, and a susceptible control line, were infected with BCTV-Svr-Cfh (US-SVR-Cfh) by agroinoculation and compared in terms of time and intensity of detection of the virus in the LAMP assays. Curly top symptoms were considerably less severe in the resistant line 36 days post inoculation (dpi); however, in LAMP assays, viral DNA was detected in leaves of the resistant and susceptible lines beginning at 7 dpi and the intensity of the color change was similar throughout the course of the experiment. We conclude that the LAMP assay developed for detection of BCTV-infected plants and viruliferous beet leafhoppers in this study is rapid and simple when compared to existing qPCR, PCR, and ELISA methods, and can be a useful tool for routine detection and improved disease management.

To make matters worse there are no effective antiviral chemicals available and, although naturally resistant host genotypes exist, they are so rare that conventional breeding techniques cannot be used reliably to create resistant plants. The most effective option to combat phytopathogenic viruses is through biotechnological intervention, such as the use of genetic engineering to develop transgenic plants or the topical use of RNA silencing technologies to prevent or modulate the severity of the viral infection. Since the first report on the virus resistance of transgenic tobacco plants in 1986, enormous progress has been made in this field. In addition great strides have been made in our ability to genetically manipulate plants and viruses leading to a plethora of novel applications.
