Book excerpt: La presente memoria ha abordado cl análisis de la fracción proteica de distintos productos lácteos mediante técnicas analíticas de alta resolución. Se ha estudiado la determinación de la relación proteína dc suero/proteína total de la leche. Se ha optimizado un método de espectroscopía ultravioleta de la 4" derivada (UV 4"' DS) que ha permitido la determinación de la citada relación sin influencia del tratamiento térmico ni del grado de proteolisis de la leche. Con este método puede detectarse la adición de suero a la leche UHT en una proporción de un 5% en proteína. También se han evaluado un método de electroforesis capilar en zona libre (CE) y un método de electroforesis capilar con SDS (SDS-CE). Se han comparado los resultados obtenidos mediante los métodos anteriores en muestras de leche sometidas a diferentes tratamientos térmicos. obteniéndose valores similares. Se han evaluado algunas técnicas aplicables a la detección de la adición de caseína o caseinato a quesos fundidos, Quarg y Mozzarella. La cuantificación dc la K-CN intacta ha demostrado ser un procedimiento adecuado para detectar la caseína ácida en los quesos elaborados con cuajo mediante CE e isoelectroenfoque en gradiente de pH inmovilizado (IPG-IEF). Se ha evaluado la determinación de lisinoalanina mediante cromatografía de líquidos de alta eficacia (HPLC) para detectar la adición de caseinato a quesos Mozzarella, comprobándose que puede ser utilizada como índice cualitativo. Se ha desarrollado un método de CE mediante el cual pueden cuantificarse simultáneamente la ß-Lg y la para-K-CN en muestras de leche y queso. El método ha permitido separar algunos productos de degradación que aparecen en la leche cruda refrigerada debido a la actividad proteolítica de bacterias psicrotrofas sobre la molécula de K-CN. Mediante la combinación de la HPLC y la espectrometría de masas (ESI-MS) se han caracterizado cinco péptidos trípticos que confirmaron la formación de para-K-CN y otros péptidos relacionados por la acción de la proteasa extracelular de Pseudomonas fluorescens B52. Además, el método de CE desarrollado permitió la caracterización de los quesos frescos por su proceso tecnológico de elaboración y por el tipo de leche utilizada.
