L'un des problèmes sanitaires émergeant est la libération des bactéries pathogènes et résistantes aux antibiotiques dans l'environnement aquatique. Les décharges plasma à la pression atmosphérique sont d'un grand intérêt pour de nombreuses applications. L'une de ces applications est la décontamination microbiologie des liquides. Les travaux développés dans cette thèse participent à mieux appréhender l'intérêt du plasma pour la décontamination microbiologie de liquides et pour la dégradation des gènes de résistance aux antibiotiques. La source utilisée est un multi-jets plasma généré à partir d'un mélange hélium-oxygène à la pression atmosphérique. Différents diagnostics ont été réalisés pour la caractérisation de la décharge plasma. Les mesures de courant et de tension réalisées nous ont permis de déterminer l'énergie du plasma. La spectroscopie d'émission optique mise en oeuvre nous a permis de mettre en évidence et d'identifier les espèces réactives de l'oxygène et de l'azote (RONS) dans le plasma. La création des RONS tels que le peroxyde d'hydrogène, les nitrites et les nitrates dans le liquide modifie la chimie du liquide. Dans ce contexte nous avons quantifié la production de ces espèces dans le liquide. L'interaction plasma-bactérie pour la décontamination microbiologie des liquides a été évaluée à travers trois études distinctes. La première consiste à comprendre l'influence de la phase de croissance des bactéries sur l'efficacité du procédé de décontamination par plasma. Nous avons montré que les bactéries en phase exponentielle sont plus sensibles au traitement plasma que les bactéries issues de la phase stationnaire. De plus, les cellules en phase exponentielle sont plus susceptibles de réduire leur activité métabolique en passant à l'état de bactéries viables mais non-cultivables (VBNC) après un court temps d'exposition au plasma tandis que celles en phase stationnaire acceptent un temps plus long d'exposition au plasma avant d'entrer dans cet état. Pour mieux appréhender les raisons qui peuvent expliquer ces différences, nous avons évalué l'effet du plasma sur l'intégrité de la membrane cellulaire et de l'ADN. Dans la seconde étude, nous avons évalué l'effet de la présence du plasmide sur la sensibilité des bactéries au traitement par plasma. Cette étude a révélé que les bactéries portant un plasmide sont plus sensibles au plasma que celles qui en sont dépourvues. Les bactéries soumises à une intégration de plasmide ont subi plus de dommages membranaires et un taux de ROS intracellulaires plus élevé après exposition au plasma. Nous suggérons que la présence de plasmides présente des inconvénients pour les bactéries en diminuant leur croissance et en imposant un état d'affaiblissement. La dernière étude consiste à évaluer l'effet du plasma sur la dégradation des gènes de résistance aux antibiotiques et sur la réduction des mécanismes de transfert horizontal des gènes (HGT) entre les bactéries donneuses traitées par plasma et celles réceptrices non traitées par plasma. Nous avons démontré que le plasma réduit le nombre de cellules donneuses, ce qui par conséquent diminue la fréquence de transfert conjugatif. Nous avons également conclu que le transfert de plasmides pouvait se produire chez les bactéries VBNC. Toutefois, la prolongation du temps d'exposition entraîne une inhibition de HGT. Bien que le plasma semble être une solution pour la décontamination des liquides et la dégradation des gènes de résistance aux antibiotiques, il serait pertinent de réaliser des études intégrant des conditions qui se rapprochent de l'environnement réel avec la présence de matière organique et en augmentant le volume du liquide traitement.

Taking advantages of the plasma plume atmosphere, plasma spray can be used for surface modification and treatment, especially for activation of polymer surfaces. I addition, plasma spray can be used to deposit nanostructures as well as advanced coating structures for new applications in wear and corrosion resistance.
