Available in PDF, EPUB and Kindle. Book excerpt: My personal history in the field of cytokines had an initial period of several years during which my student and then colleague, Werner Muller, tried in vain to attract me to them. My interest always vanished when I was confronted with complex data pointing to func tional redundancy of cytokines in cell culture systems. When gene targeting in the mouse germline became possible, this frustration came to an end. We and others immediately embarked on analyzing the in vivo function of cytokines and the problem of functional redundancy with this powerful new approach. The early cytokine gene knockouts performed by colleagues in Wiirzburg (IL-2) and by ourselves (IL-4 and IL-l 0) seemed to give clear answers and at the same time led to surprises: Each of these cytokines apparently had its own special and irreplaceable function, and this function could be quite distinct from what had been anticipated from functional experiments in vitro. Al though the latter finding is of course a wonderful incentive for fur ther research, the former is pleasing in a general sense since it highlights the value of each of those one hundred thousand genes or so in our genome, cherished by evolution to become respectable mem bers of the community. Even in the present era of "genomics" there will be no way around the careful functional analysis of each gene by itself.

Available in PDF, EPUB and Kindle. Book excerpt: Cancer is fundamentally a disease of abnormal cell proliferation: Cancer cells multiply when and where they should not. This proliferation entails escape from normal bounds imposed by the tissue environment, the internal biology of the cell (DNA damage, chromosomal imbalances, disorganized mitotic spindles), and the proliferative history of the cell (normal generational times). Some of the key oncogenic events in cancer directly perturb proteins that regulate progression through the cell division cycle, others alter cell cycle progression indirectly, through effects on signaling pathway that impinge on the cell cycle. This biology is fundamentally important in cancer therapy. Many of the workhorse treatments for cancer rely on killing proliferating cells. Furthermore, there is growing recognition that stem cell-transit amplifying cell hierarchies may persist or be generated during tumorigenesis, generating important functional heterogeneity in cell cycle control among tumor cells, with far-reaching scientific and clinical implications. This volume outlines major cell cycle perturbations that drive tumorigenesis and considers the prospects for using such knowledge in cancer therapy.

Book excerpt: The development of the young brain after birth and the emergence of cognitive capacities, mind, and individuality rest on the maturation of a dense net of synaptic connections between neurons. Memory Makes the Brain describes the dramatic, competitive elimination of surplus synapses that occur in the young, maturing brain — in a process called synaptic pruning that was discovered by pediatric neurologist Peter Huttenlocher in the 1970's at the University of Chicago. Explaining similarities between developmental pruning and learning processes in the adult brain, neurobiologist Christian Hansel offers a unique perspective on brain adaptation and plasticity throughout lifetime, at times weaving in personal accounts and memories. The cellular plasticity machinery that enables learning is known to be affected in brain developmental disorders such as autism. Memory Makes the Brain explains how both maturation and adult synaptic plasticity are deregulated in autism, and how we begin to trace back autism-typical behavioral abnormalities to such synaptopathies.

Available in PDF, EPUB and Kindle. Book excerpt: Free radicals are constantly formed in living cells and removed by antioxidant defenses. Antioxidant enzymes are the main line of defense against free radicals in animal and plant cells. Uncontrolled generation ROS are involved in a number of human disease states, including diabetes and cancer due to disturbance in cellular and molecular processes including cell growth, differentiation and proliferation. When cells are exposed to oxidative stress a defense system endorses the expression and regulation of number of antioxidant enzymes as a defense mechanism to protect them from the damage induced by free radicals. Based on this fact, the book "Antioxidant Enzymes" was designed to overview the importance of the antioxidant enzymes in human and plant cells against toxic free radicals, their relationship with several pathophysiologic processes and their possible therapeutic application.

Each chapter presents different technological approaches for cost effective liquid biofuel production from agroresidues/biomass. Two chapters cover future direction and the possibilities of biomass-based biofuel production at the industrial level. The book provides a genetic and metabolic engineering approach for improved cellulase production and the potential of strains that can ferment both pentose and hexose sugars. The book also gives direction on how to overcome challenges for the further advancement of lignocellulosic biomass-based biofuel production.

Available in PDF, EPUB and Kindle. Book excerpt: Cyclin Dependent Kinase 5 provides a comprehensive and up-to-date collection of reviews on the discovery, signaling mechanisms and functions of Cdk5, as well as the potential implication of Cdk5 in the treatment of neurodegenerative diseases. Since the identification of this unique member of the Cdk family, Cdk5 has emerged as one of the most important signal transduction mediators in the development, maintenance and fine-tuning of neuronal functions and networking. Further studies have revealed that Cdk5 is also associated with the regulation of neuronal survival during both developmental stages and in neurodegenerative diseases. These observations indicate that precise control of Cdk5 is essential for the regulation of neuronal survival. The pivotal role Cdk5 appears to play in both the regulation of neuronal survival and synaptic functions thus raises the interesting possibility that Cdk5 inhibitors may serve as therapeutic treatment for a number of neurodegenerative diseases.

These multitasking organelles are also part of species-specific pathways such as the glyoxylate cycle in yeast and plants or the synthesis of ether lipid in mammals. Proteins required for the biogenesis of peroxisomes typically assemble in large molecular complexes, which participate in membrane formation, protein transport, peroxisome duplication and - inheritance during cell division. Peroxisomal function is essential for life. Mutations in PEX genes, encoding for biogenesis factors, are often associated with lethal disorders. The association of peroxisomes with other organelles suggests an extensive participation in organellar crosstalk. This book represents a state-of-the-art review in the field of peroxisome research encompassing the cell and molecular biology of peroxisome biogenesis and its diseases, the protein complexes involved in this process and the modern technologies applied to study them. The book is intended for graduate students, researchers and lecturers in biochemistry, molecular and cell biology with a biomedical background.

It is now a very widely-used term, but there is still a lot of confusion surrounding what it actually is and does.Epigenetics is a new textbook that brings together the structure and mac
