Book excerpt: This book brings together lmportant work on the structure and function of the dehydrogenases requiring nicotinamide coenzymes. The structure and function of lactate dehydrogen ase, alcohol dehydrogenase, and glyceraldehyde 3-phosphate dehydrogenase are compared. These are the relatively simple oligomeric dehydrogenases for which very extensive structural and mechanistic information is available. Kinetic studies are dealt with in general terms, with selected examples. Genetic and structural relationships revealed by primary structure are discussed. General features of the specificity of dehydrogen ase reactions are described. The pyridine nucleotide - disulphide oxidoreductases exemplify a type of multicomponent enzyme complex. Lipoamide dehydrogenase, glutathione reductase, and thioredoxin reduct ase are described and compared. The fatty acid synthesizing systems of some organ isms are nonaggregated, whereas in other organisms they are multienzyme complexes containing, in animal tissues for example, two closely related multifunctional polypeptide chains. The dehydrogenase activities of the fatty acid syn thesizing systems afford many points of interest, which are described and discussed. Hydroxymethylglutaryl-CoA reductase occupies a key position in the synthesis of isoprenoid compounds, including cholesterol. It probably occurs in all forms of life; in animal cells it is a microsomal enzyme. Its properties are reviewed.

Book excerpt: The book takes in consideration some dehydrogenases, enzymes with different functions in the cells by using different substrates, such as hydroxysteroid dehydrogenases, aldehyde dehydrogenases, glucose-6-phosphate dehydrogenase, pyruvate dehydrogenase complex, glutamate dehydrogenase, succinate dehydrogenase. They are examined from the following points of view: biochemistry, physiological functions and role in some diseases and in the development of tumours. For these reasons, the book is divided into three sections: 1. Dehydrogenases and cancer 2. Dehydrogenases and some diseases 3. Dehydrogenases and physiological role

Available in PDF, EPUB and Kindle. Book excerpt: The dynamic field of flavin and flavoprotein biochemistry has seen rapid advancement in recent years. This comprehensive two volume set provides an overview of all aspects of contemporary research in this important class of enzymes. Topics treated include flavoproteins involved in energy generation, signal transduction and electron transfer (including respiration); oxygen activation by flavoproteins; the biology and biochemistry of complex flavoproteins; flavin and flavoprotein photochemistry/photophysics as well as biotechnological applications of flavoproteins. Recent developments in this field include new structures (including those of large membrane-integral electron transfer complexes containing FMN or FAD), elucidation of the role of flavoproteins in cell signalling pathways (including both phototaxis and the circadian cycle) and important new insights into the reaction mechanisms of flavin-containing enzymes. This volume focussing on oxidases, dehydrogenases and related systems is an essential reference for all researchers in biochemistry, chemistry, photochemistry and photophysics working on flavoenzymes.

Book excerpt: The Sixth International Workshop on the Enzymology and Molecular Biology of Carbonyl Metabolism was held outside of Dublin, Ireland at the end of June, 1992. Prof. Keith Tipton, Chairman of the Biochemistry Department at Trinity College, kindly agreed to host the meeting. On behalf of all of us who attended I wish to extend our sincere thanks to the whole Tipton family for making us feel so welcome in Ireland. It has been a decade since the frrst workshop was held in Bern, Switzerland. The scope of the meetings reflected somewhat the changes that have occurred in biochemistry during the past decade. At the first meeting primarily enzymes and their properties were discussed. At this last meeting many of the talks centered on gene regulation as well as more traditional aspects of enzymology and metabolism. During the past decade site directed mutagenesis to probe for the active site of an enzyme has become part of traditional enzymology; this was virtually unheard of at our frrst meeting. Many of the presenters now used this tool to study some aspect of structure and function of one of the three carbonyl metabolizing enzymes.

Book excerpt: Since 1982, our ever-expanding group of investigators has been meeting in exotic parts of the world to discuss aspects of three enzyme systems. The 1996 meeting was no exception. Nearly 90 scientists from 15 countries met in the small city of Deadwood, South Dakota, for four days of stimulating talks and posters and incredible scenery. Once more this meeting reflected the changing trends in biochemical research. At the 1982 meeting most of the speakers discussed isolating new enzymes and trying to characterize them. At this meeting many speakers discussed interpretations of three-dimensional struc ture or regulatory elements of the genes controlling for the tissue-specific expression of the enzyme. Hopefully, readers will find the proceedings of the meeting to be of interest. Though they reflects the scientific information that was presented at the meeting, they do not indicate the level of personal interactions that went on during the meeting. Once again, the willingness of the participants to discuss unpublished data and to share thoughts about the future directions of their research helped make this, like our previous seven meetings, a special scientific experience for those who attended.

Available in PDF, EPUB and Kindle. Book excerpt: Paves the way for new industrial applications using redox biocatalysis Increasingly, researchers rely on the use of enzymes to perform redox processes as they search for novel industrial synthetic routes. In order to support and advance their investigations, this book provides a comprehensive and current overview of the use of redox enzymes and enzyme-mediated oxidative processes, with an emphasis on the role of redox enzymes in chemical transformations. The authors examine the full range of topics in the field, from basic principles to new and emerging research and applications. Moreover, they explore everything from laboratory-scale procedures to industrial manufacturing. Redox Biocatalysis begins with a discussion of the biochemical features of redox enzymes as well as cofactors and cofactor regeneration methods. Next, the authors present a variety of topics and materials to the research and development of full-scale industrial applications, including: Biocatalytic applications of redox enzymes such as dehydrogenases, oxygenases, oxidases, and peroxidases Enzyme-mediated oxidative processes based on biocatalytic promiscuity All the steps from enzyme discovery to robust industrial processes, including directed evolution, high-throughput screening, and medium engineering Case studies tracing the development of industrial applications using biocatalytic redox reactions Each chapter ends with concluding remarks, underscoring the key scientific principles and processes. Extensive references serve as a gateway to the growing body of research in the field. Researchers in both academia and industry will find this book an indispensable reference for redox biotransformations, guiding them from underlying core principles to new discoveries and emerging industrial applications.
