Abnormal infiltration of neutrophils is the hallmark of inflammatory disease. The activation and regulation of neutrophil chemotaxis must be precise in order to prevent abnormal situations leading to disease states. In this dissertation, I use zebrafish as an italicin vivoitalic model to delineate the cell signaling and cytoskeletal regulation of neutrophil interstitial migration and innate immune response. In particular, I elucidate two negative regulators of neutrophil motility, the SH2-domain-containing inositol 5-phosphatase (SHIP) and microtubules. Phosphoinositide 3-kinase (PI3K), which generates the phosphatidylinositol (3,4,5)-trisphosphate [PI(3,4,5)P(3)], is necessary for neutrophil motility italicin vivoitalic; however, the role of SHIP enzymes, which hydrolyze PI(3,4,5)P(3) to phosphatidylinositol 3,4-bisphosphate [PI(3,4)P(2)], is not well understood. In Chapter 2, I demonstrate that SHIPs control neutrophil inflammation and limit neutrophil motility by modulating PI3K signaling. The role of microtubules in regulating leukocyte migration is also not clear. In Chapter 3, I illustrate that microtubule depolymerization inhibits polarized PI3K activation at the leading edge of neutrophils, inducing PI3K independent motility but negatively impacting neutrophil directional migration. Our data also suggest that microtubules exert their effects on neutrophil polarity and motility at least in part by the negative regulation of both Rho and Rac activities. Next, in Chapter 4, I demonstrate that the routine heat shock method used for inducible gene expression in animal models with common heat shock promoters can alter neutrophil function in zebrafish. My findings suggest that caution must be taken when employing a heat shock-dependent inducible system to study the innate immune response and that data arising from such experiments need to be carefully scrutinized. Together, my thesis research provides a better understanding of the signaling mechanisms involved in regulating neutrophil migration and function italicin vivoitalic.

Book excerpt: Abstract: Signaling through the B cell antigen receptor (BCR) is essential for several processes of B cell biology, such as proliferation, differentiation and homeostasis. Accordingly, the signaling events emanating from the BCR have to be tightly regulated in order to ensure an effective immune response and at the same time avoid pathological conditions like autoimmunity and lymphomagenesis. Mature B cells express two different BCR isotypes, IgM and IgD, which reside in distinct, isotype-specific protein islands on the B cell surface. These structures dependent on the actin cytoskeleton, the disruption of which induces robust BCR signaling, even in the absence of antigen. We here have studied the isotype-specific regulation of the BCR by the cytoskeleton. Using several different genetically engineered mouse models we found that cytoskeleton disruption induced signaling (CDIS) is mediated by the IgD-BCR and the co-receptor CD19. CDIS differs from canonical, antigen-induced BCR signaling in many facets including a lack of BCR proximal tyrosine phosphorylation and induction of a calcium flux originating exclusively from the extracellular space. CDIS can also be enhanced by anti-CD19 stimulation in the absence of a complete IgD- BCR structure as long as the Ig[beta] signaling subunit of the BCR is still expressed on the B cell surface. Together these findings reveal a functional difference between the IgM-BCR and IgD-BCR as well as Ig[alpha] and Ig[beta] in their interactions with CD19 and the actin cytoskeleton. Furthermore, our results provide explanations for the multitude of immunological phenotypes observed in mutants of actin-binding proteins

Book excerpt: The high affinity immunoglobulin E (IgE ) receptor (Fc[epsilon]RI) is a member of a family of related antigen receptors with conserved structure and similar roles in initiating intracellular signaling in response to an outside stimulus. In mammalian cells, Fc[epsilon]RI is found primarily on the surface of mast cells and basophils. Activation of this receptor by multivalent antigen triggers a series of intracellular signaling events that ultimately lead to the secretion of preformed mediators, such as histamine and serotonin, which are responsible for allergic reactions. Our recent efforts in the study of immunoreceptor signaling have focused on the participation of the plasma membrane and the actin cytoskeleton. However, tools for investigating biomolecular dynamics and interactions that occur within close proximity to the plasma membrane of living cells are somewhat limited. Micro-fabricated surfaces have been used for applications in cell and tissue engineering, although their full potential has not been explored, particularly in the area of molecular cell biology. We have recently developed a polymer lift-off method for localizing the initial stimulus to micron size patches of antigens such that assembly of signaling components could be visualized with selective fluorescent labeling and confocal microscopy. Employing these technologies we have investigated fundamental mechanisms in immune cell signaling, specifically IgE receptor (Fc[epsilon]RI) signaling involved in allergic responses on mast cells. In past years, our group has observed spatially and temporally resolved responses, including receptor phosphorylation, selective accumulation of Lyn kinase and other membrane components. In this work, I have applied these tools to investigate functional and structural implications of the actin cytoskeleton regulation of Fc[epsilon]RI signaling and the cytoskeletal dependence of activated receptor internalization. I have shown that particular actin binding proteins, paxillin, talin and vinculin co-redistribute at the patterned antigen and clustered receptors in a similar time course as polymerized actin. These proteins have been extensively characterized in the context of integrin signaling and focal adhesion assembly, but little is known about their involvement in immune cell signaling. Our biochemical data suggests that these proteins have functional relevance in some of the early and late events of the signaling cascade. In addition, I show that the GTPase dynamin 2, which is commonly associated with receptor endocytosis, is selectively recruited to the sites of receptor clustering. Protein knockdown of dynamin 2 decreases the accumulation of F-actin at the clustered receptors, implicating dynamin in the regulation of Fc[epsilon]RI mediated actin redistribution. These results point a possible role for dynamin-actin interactions in regulating activated IgEFc[epsilon]RI internalization.
