No primeiro caso, preocupou-se em estabelecer condicoes para cultivo in vitro de explantes de Passiflora edulis f. flavicarpa Deg. (maracuja amarelo). A enxertia foi empregada visando estudar a compatibilidade de porta-enxertos para maracuja amarelo e maracuja guacu (Passiflora alata Ait).Os explantes para cultivo in vitro foram retirados de plantas adultas e de plantulas de maracujazeiro amarelo. Foram feitos tres ensaios, sendo que no primeiro, o meio de cultura foi o de NITSCH & NITSCH (1969). Observou-se que explantes retirados de plantulas nao apresentaram problemas de contaminacao interna, encontrando-se, ao contrario, grande percentagem de infeccao em explantes provenientes de ramos de plantas adultas. No segundo ensaio foram testadas seis variacoes do meio Nitsch & Nitsch, utilizando-se explantes de maracuja amarelo com um ano de idade e mantidos em casa de vegetacao. Verificou-se que meio basico sem fito-hormonios, permitiu pequeno crescimento dos explantes que cessou pronunciadamente ao final do primeiro mes de cultivo. Quando se acrescentou 6 BA (2mg/l) ou 2 i-P (2mg/l) houve desenvolvimento apenas de parte aerea. Ao se acrescentar 6 BA (0,5mg/l) e IAA (1,0mg/l) formaram-se pequenos callus e desenvolveu parte aerea. A combinacao meio basico acrescida de cinetina (2mg/l) e IAA (0,1mg.l apresentou formacao de callus mais volumosos e menor desenvolvimento de parte aerea. Quando se acrescentou 2,4-D (3mg/l) e leite de coco (100mg/l) houve formacao de callus.
