Light governs their photosynthesis and other biological functions. Specifically, cryptochromes, photoreceptors found universally, primarily detect UV-A/blue light and are known to repair DNA. This study focuses on five algal cryptochromes, especially CRY-DASH1 (Drosophila, Arabidopsis, Synechocystis, Homo) in the green alga, Chlamydomonas reinhardtii. Located in the chloroplast, CRY-DASH1's expression peaks around midday. A mutant lacking CRY-DASH1 showed decreased growth but enhanced photosynthesis efficiency. Furthermore, the mutant revealed increased pigment amounts, hyper-stacking of the thylakoid membrane and elevated levels of photosystem II proteins like D1 and CP43, but not of their transcripts. These data suggest that CRY-DASH1 plays its role as regulator at the posttranscriptional/translational level. CRY-DASH1 has an absorption peak in the UV-A range and supplementation of white light with UV-A increases photoautotrophic growth of wild type but not of the mutant lacking CRY-DASH1. Comparative analysis of the chloroplast proteome of wild type and of the mutant revealed that multiple proteins, including key enzymes for the synthesis of chlorophylls and carotenoids, as well as acyl-lipid metabolism and photoprotection, are upregulated in the mutant. In contrast, we found that essential proteins from the central carbon metabolism, are downregulated. Additionally, we discovered that the mutant revealed a downregulation of certain enzymes involved in histidine metabolism which leads to a reduction in the quantity of free histidine. Another cryptochrome, CRY-DASH2, when mutated, exhibited similar growth reduction and pigment increase. The study ends pondering if CRY-DASH1 and CRY-DASH2 might have overlapping functionalities.

Book excerpt: Cryptochromes are known as blue light sensory flavoproteins in plants, insects, fungi, and bacteria and as part of the circadian oscillator, the latter mainly in mammals. An animal-like cryptochrome (aCRY) from Chlamydomonas reinhardtii was recently characterized as a blue and red light photoreceptor that plays a crucial role in the regulation of light-dependent genes involved in chlorophyll and carotenoid biosynthesis, light-harvesting complex assembly, nitrogen metabolism, cell cycle control and the circadian clock. In this work, aCRY was found to be mainly a soluble protein, which is also attached to membranes to some extent. Soluble aCRY accumulates in vegetative cells and mature zygotes, but not in pregametes, gametes and early zygotes. The degradation of soluble aCRY in pregametes, gametes and early zygotes is processed by the proteasome pathway. In contrast to soluble aCRY, membrane-associated aCRY accumulates in vegetative cells, pregametes, gametes, and late zygotes, but not in early zygotes. Based on an available acry mutant, acry mutant strains of mating type plus and minus with similar genomic background as wild-type strains were generated by backcrossing. Also complemented strains of these mutants were generated. Based on these strains, aCRY was found to act as a negative regulator in mating ability and in maintenance of mating ability, in contrast to the blue light photoreceptor phototropin that acts positively in these processes. This work also showed that mainly blue, and to a lesser degree red light are involved in zygote germination, during which the zygote undergoes meiosis. In addition to phototropin, both aCRY and plant cryptochrome were found to positively regulate zygote germination. These results clearly suggest that not only phototropin but also aCRY and plant cryptochrome are key regulators of the sexual cycle of C. reinhardtii.

This volume presents 15 chapters by world experts on their subjects, ranging from reviews of algal diversity and genetics to in-depth reviews of special algal groups such as diatoms (which account for over 30% of marine carbon capture). Other chapters chart the ways in which this carbon capture occurs or how there are a multiplicity of ways in which algae intercept sun light and deploy this energy for carbon capture. A fascinating aspect here is the way in which sun light is harvested. A special chapter is devoted to the very recent and exciting possibility that algae use coherent light energy transformation to enhance the efficiency of light capture, an aspect of quantum physics that has implications for future developments at several levels and a variety of industries. Just how and why algae use Chlorophyll a as the major light capture pigment is discussed in several chapters. However, attention is also given to those cyanobacteria, which have been found to use the special Near-Infra Red absorbing chlorophylls mentioned above. And attention is also given to those algae that employ phycobiliproteins to fill in the “green window”, i.e., the spectral region from 400 – 650 nm, which is not efficiently covered by chlorophyll and carotenoid pigments. Photoinhibition and photoprotection is the subject area of several chapters and one which it is essential to understand a we work towards greater efficiency of algal photosynthesis.
