Book excerpt: Biodegradable nanomaterials have attracted broad interests because of their significant biomedical applications. However, it remains a challenge to develop simple and robust approaches for the preparation of biodegradable nanomaterials with well-controlled structures and optimal properties. Nanoparticles typically have high tendency to aggregate, but such aggregation behavior may impair their performance as drug delivery scaffolds. Our research group recently demonstrated an efficient synthesis of well-defined biodegradable nanoparticles by thiol-ene cross-linking in transparent miniemulsion. However, the resulting polylactide-based nanoparticles can easily aggregate in various liquid media. This thesis research is to achieve nanoparticles with minimal aggregation tendency by covalent stabilization of surfactants on the surface of nanoparticles. Reactive surfactants were designed, prepared, and used in the preparation of nanoparticles. The nanoparticles with fixed surfactant moieties on surface exhibited remarkably high colloidal stability.
