Book excerpt: Controlled synthesis of metal nanoparticles is an important area of study because of the unique size and shape dependent properties of such nanoparticles. For example, gold and silver nanoparticles show distinct surface Plasmon resonance (SPR) bands varying with the size, shape and surface morphology of the nanoparticles. The assembly of these nanoparticles usually exhibit interesting collective optical properties different from their individual components. However, controlled synthesis and assembly of metal nanoparticles with high yield and enhanced optical properties is still challenging. In this thesis, I presented the synthesis of ultrathin gold/silver nanowires and sub-50 nm gold triangular nanoprisms by adopting silver seeds and benzyl dimethyl hexadecyl ammonium chloride (BDAC) in a seed-mediated growth method. Furthermore, I developed a templated surfactant-assisted seed growth method to synthesize gold nanoshells with varying surface morphologies by changing the type of surfactants and ions in the growth solution. The optical properties of the nanoshells could be controlled over a wide wavelength range by varying the surface morphology. By using a combination of BDAC surfactant and template surfactant-assisted seed growth method, isotropic shell-type gold nanoparticle clusters, or raspberry-like meta-molecules (raspberry-MMs), were successfully synthesized. The raspberry-MMs exhibited interesting far field and near field optical properties. The raspberry-MMs showed unusually strong magnetic resonances, yielding broad SPR bands in the visible and near-IR region. Both experimental data and finite-difference time-domain (FDTD) simulations showed that the magnetic dipole can be even larger than that of the electric dipole resonance in large raspberry-MMs. Moreover, I utilized Raman spectroscopy as a tool to probe the near field optical properties of the raspberry-MMs. Due to the existence of a large number of hotspots at the gaps among gold beads within individual raspberry-MMs, individual raspberry-MMs proved to be efficient Raman substrate. Importantly, the contribution from hotspots created at the gap between two adjacent raspberry-MMs is negligible compared to that from a large number of hotspots on individual raspberry-MMs. Consequently, the Raman signal intensity of the analyte molecules on the raspberry-MM dimers exhibited quite narrow distribution and is weakly dependent on the distance between the two raspberry-MMs. This paved the way for fabricating large-area raspberry-MM films which can serve as macroscopic efficient and reproducible Raman substrate. The robustness and tunability of the synthetic method presented in this thesis, the strong magnetic responses of the raspberry-MMs and the efficient Raman enhancement from single raspberry-MM, raspberry-MM dimer and films can lead to large-scale manufacture and wide applications of magnetic metamaterials as well as commercialization of uniform Raman substrate.

Book excerpt: Nanoparticles have garnered much attention in pharmaceutical and biomedical fields because their small size and high surface area facilitate drug absorption, improve access to cells and organs, and enhance optical imaging. However, delivery of nanoparticles to the body is not always feasible or effective. Here, nanoparticle assemblies (flocs or clusters) for pulmonary drug delivery and biomedical imaging in cells are shown to facilitate delivery, interactions with cells, and manipulation of optical properties of inorganic/organic nanocomposites. The formation of aggregates by physical techniques and their mechanisms are described in detail. For pulmonary delivery, particles with aerodynamic diameters between 1-5 [mu]m deposit efficiently in the deep lungs. However, crystalline, non-porous, poorly water soluble drugs of this size require long dissolution times, limiting absorption by the body. Therefore, drug dissolution must be "decoupled" from deposition to improve absorption. To address this challenge, drug nanoparticles were dispersed within 4-[mu]m water droplets when administered via nebulization or as micron-sized flocs using a pressurized metered dose inhaler (pMDI). Upon deposition in aqueous media, the aerosolized nanoparticle assemblies dissociated into constituent nanoparticles, raising the available surface area for dissolution and increasing dissolution rates, relative to solid particles. Poorly water soluble drug nanoparticles were prepared using a controlled precipitation (CP) or thin film freezing (TFF) process, in which stable nanoparticles (30-300 nm in diameter) with high potencies (>90 wt% drug) were produced by rapidly nucleating drug solutions in the presence of strongly adsorbing polymers or by freezing, respectively. Amorphous, nanoparticles prepared by CP produced stable aqueous dispersions with high fine particle fractions (FPF) of 77% and total emitted doses (TED) of 1.5 mg/min upon nebulization. CP and TFF also produced anisotropic particles (aspect ratios >5), which formed stable suspensions in a hydrofluoroalkane propellant. Inefficient packing of anisotropic particles formed loose, open flocs that stacked upon each other to prevent settling. Upon pMDI actuation, atomized propellant droplets shear apart and template portions of the floc to yield porous particles with high FPFs (49-64%) and TEDs (2.4 mg/actuation). The controlled assembly of gold nanoparticles into clusters is also of great interest for biomedical imaging and therapy because clusters exhibit improved near infrared absorbance (where blood and tissue are most transparent), relative to single spherical particles, and can biodegrade into clearable particles. Gold nanoparticles (5 nm) were assembled into clusters between 30 to 100 nm in diameter with high gold loadings, resulting in strong NIR absorbance. The assembly was kinetically controlled with weakly adsorbing polymers by manipulating electrostatic, van der Waals, steric, and depletion forces. Furthermore, clusters assembled with a biodegradable polymer deaggregated back into primary particles in physiological media and within cells. This kinetic assembly platform is applicable to a wide variety of fields that require high metal loadings and small particle sizes.

Book excerpt: The optical properties of metallic particles change dramatically as the size shrinks to the nanoscale. The familiar mirror-like sheen of bulk metals is replaced by the bright, sharp, colorful plasmonic resonances of nanoparticles. The resonances of plasmonic metal nanoparticles are highly tunable throughout the visible spectrum, depending on the size, shape, local dielectric environment, and proximity to other optical resonances. Fundamental and applied research in the nanoscience community in the past few decades has sought to understand and exploit these phenomena for biological applications. In this work, discrete nanoparticle assemblies were produced through biomolecular interactions and studied at the single particle level with darkfield spectroscopy. Pairs of gold nanoparticles tethered by DNA were utilized as molecular rulers to study the dynamics of DNA bending by the restriction enzyme EcoRV. These results substantiated that nanoparticle rulers, deemed "plasmon rulers", could measure the dynamics of single biomolecules with high throughput, long lifetime, and high temporal resolution. To extend these concepts for live cell studies, a plasmon ruler comprised of peptide-linked gold nanoparticle satellites around a core particle was synthesized and utilized to optically follow cell signaling pathways in vivo at the single molecule level. The signal provided by these plasmon rulers allowed continuous observation of caspase-3 activation at the single molecule level in living cells for over 2 hours, unambiguously identifying early stage activation of caspase-3 in apoptotic cells. In the last section of this dissertation, an experimental and theoretical study of electomagnetic coupling in asymmetric metal nanoparticle dimers is presented. A "heterodimer" composed of a silver particle and a gold particle is observed to have a novel coupling between a plasmon mode (free electron oscillations) and an inter-band absorption process (bound electron transitions). The modes in such asymmetric "plasmonic molecules" are possibly characterized by unique spatial and spectral profiles, and polarization behavior, allowing one to tune the optical response of the nanostructure in the near and far fields.

Book excerpt: Noble metal nanoparticles exhibit unique optical properties arising from the resonant oscillations of their conduction electrons with light. This phenomenon is called localized surface plasmon resonance (LSPR). The LSPR frequency is extremely sensitive to the size, shape, refractive index at the metal-dielectric interface, and other nearby metal nanoparticles. In an assembly of proximal nanoparticles, the LSPR of individual particles can couple to yield enhanced light scattering and large spectral shifts, which are useful for many applications including diagnostics and sensing. This dissertation presents a complex nanostructure comprising a core gold nanoparticle surrounded by multiple satellite gold nanoparticles for biosensing application. Chapter 2 introduces the fabrication and characterization of the core-satellite assemblies via a layer-by-layer process. Using ATP-aptamer as the linker, we demonstrated the detection of ATP based on the disassembly of the nanostructure, which can be readily captured by darkfield microscopy. The detection limit, dynamic range, and sensitivity can be tuned by controlling the size of the assembly. We found that the aptamer-linked nanoparticle assemblies were selective to only ATP, and not other adenine-containing compounds. Additionally, sensing of ATP in buffer and in bulk cell lysates was demonstrated. Chapter 3 presents the methodology for detecting ATP directly from lysed cells, down to the single-cell level without the need for purification or extraction. The intracellular ATP levels of two ovarian cancer cell lines were quantified to elucidate the differences and cellular distribution, and the potential of the stick-and-peel platform for mapping the microenvironment of 2D heterogeneous surfaces was demonstrated. In chapter 4, the optical properties of nanoparticle assemblies were tuned by changing the morphology of the nano building block, where the incorporation of gold nanoshells as satellites led to an extended redshift of LSPR to a much longer wavelength compared with using solid gold nanoparticles as satellites. This tunability in the LSPR of the assemblies allows for color-based analysis and color-coding of the plasmonic sensors. Lastly, Chapter 5 outlines the development of a multiplexed assay using the nanoparticle assemblies. Two types of assemblies, targeting either ATP or a nucleic acid (DNA-210), were fabricated with different DNA linkers in the same sensing area. The multiplexing was demonstrated by the selective disassembly process. Moreover, the ability to tune the optical properties of nanostructures using different morphologies was integrated; two different morphology of nanostructures, i.e. solid-solid and solid-shell nanostructures, for two targets, ATP and DNA-210, respectively, were fabricated. Based on a difference in scattered color, two types of biosensors among thousands of nanoparticle assemblies can be easily identified. Finally, we demonstrated duplex detection based on the change in the scattering intensity and the color read-out. Reflecting on the contributions of our work, this dissertation advances the fundamental knowledge and practical design of chip-based sensing platforms comprising complex plasmonic nanostructures. The work contributes to the sensing field by addressing some of the challenges in point-of-care or point-of-need measurement applications and provides an alternative bioanalytical tool for single-cell based analysis.
