We have used the model plant species Arabidopsis thaliana to investigate control of cell proliferation during development by three transcription factors: SPATULA (SPT), encoding a bHLH transcription factor and FOUR LIPS (FLP) and its paralog MYB88, both encoding R2R3 MYB transcription factors. Detailed analysis of embryo and root development showed that loss of SPT function causes an increase in quiescent center size in both the embryonic and postembryonic stem cell niche. spt mutants exhibit other pleotropic developmental phenotypes, including longer hypocotyls, more flowers, shorter internodes and an extended flowering period. FOUR LIPS (FLP) and MYB88 have overlapping functions in restricting Guard Mother Cell (GMC) division during stomatal development. In our current study, we have identified another role for FLP during female gametogenesis.

The model plant Arabidopsis thaliana was used to investigate how AHL-mediated changes in transcription influence development. YUCCA8 (YUC8), a gene involved in auxin biosynthesis, and members of the SMALL AUXIN UP RNA (SAUR) 19 subfamily, which act at the end of the auxin signaling pathway and promote hypocotyl cell expansion, were misregulated in SUPPRESSOR OF PHYTOCHROME B4-#3 (SOB3)/AHL29 mutant seedlings. Specifically, transcripts for these genes were depleted in the short-hypocotyl SOB3-D over-expression line, but elevated in the tall, dominant-negative sob3-6 mutant. Further, ectopic expression of SAUR19 partially rescued the short-hypocotyl phenotype characteristic of SOB3-D, and SOB3 bound the promoters of YUC8 and SAUR19 subfamily members. This indicated that SOB3 represses hypocotyl elongation by inhibiting the transcription of both YUC8 and SAUR19 subfamily members. Although YUC8 and SAUR19 are both primarily associated with auxin, links between SOB3, these genes, and other signaling pathways were also identified. Light intensity also impacted the severity of hypocotyl phenotypes in SOB3 mutants. A link between SOB3 and light-mediated signal transduction was also identified at the molecular level, as expression of YUC8 and SAUR22 were affected by both light fluence rate and the transcription factor PIF4. SOB3 phenotypes and SAUR22 expression were also affected by brassinosteroid signaling, as evidenced by growth in the presence of brassinolide, an active form of the hormone, or the biosynthesis inhibitor brassinazole. Transgenic seedlings expressing a mutant form of BZR1 which codes for a constitutively active version of the protein also had higher levels of SAUR22, revealing a link between SOB3 and brassinosteroid signaling at the molecular level. Finally, physical interactions between histone H2B and SOB3 were also found to be essential for modulation of hypocotyl growth. Therefore, interactions with histones must also be important for regulation of gene expression by SOB3.

Book excerpt: The formation of the apical-basal axis during Arabidopsis thaliana embryogenesis is a crucial step that directs the overall body plan of the seedling and directs the establishment of stem cell populations at either end of the embryo that give rise to all of the adult structures. The establishment of the apical-basal axis occurs with the first division of the zygote and remains plastic until the transition stage, where apical-basal polarity becomes fixed. The molecular mechanisms and genes responsible for the formation and fixation of the apical-basal axis remain largely unknown. This dissertation describes the advances made toward the elucidation of two major pathways that contribute to the formation and fixation of the apical-basal axis. Transcriptional repression plays a major role in developmental processes in general and this dissertation describes the role of the TOPLESS (TPL) gene in repression of the basal embryonic gene program in the developing apical domain of the embryo. We provide a framework for the function of TPL in transcriptional repression through chromatin remodeling. Furthermore, we show that the PLETHORA (PLT) family of transcription factors are direct targets for TPL mediated repression and necessary for the shoot to root transformation seen in the tpl-1 mutant. Secondly, using the tpl-1 mutant as a tool for forward genetics, the HD-ZIP Class III transcription factor family members are identified as determinants of apical fate in the globular stage embryo. Additionally, the HD-ZIP III genes are shown to have a mutually antagonistic relationship with the PLT gene family. Lastly, we show that the HD-ZIP III genes are true apical determinants and able to cause a homeotic conversion of the basal pole to a second apical pole.
