Book excerpt: Estudos preliminares no que se refere ao explante, tipo e concentrações de reguladores vegetais são necessário para se definir um protocolo de conservação in vitro de germoplasma, bem como garantir a fidelidade na resposta morfogenética. Avaliou-se a influência dos reguladores de crescimento vegetal ácido nafatalenoacético (ANA) e ácido giberélico (AG3) no estiolamento in vitro e posterior regeneração de plantas do abacaxizeiro hídrico PE x SC-60 (Perolera x Smooth Cayenne). Os resultados revelaram que o ANA favoreceu quanto ao estiolamento, números de gemas e diâmetro de haste estiolada, no entanto ele não foi eficiente quanto à resposta organogênica, independentemente da concentração do AG3. Foram testados também os reguladores vegetais benzilaminopurina (BAP) e paclobutrazol (PBZ) sob a taxa de multiplicação in vitro do abacaxizeiro hídrico de PE x SC-60,quando da utilização de hastes estioladas provenientes do meio de cultura MS (Murashige & Skoog,1962)sem adição de reguladores. Verificou-se que o PBZ não foi eficiente na regeneração de brotos, enquanto que a concentração de 0,5mg. L-1 de BAP propocionou uma taxa de multiplicação de 36,5 brotos por explante. Estudou-se ainda o efeito do PBZ no crescimento in vitro do referido hídrico, visando à conservação de germoplasma. Os resultados mostraram a viabilidade de se manter, por um ano, plantas deste hídrico em condições de crescimento reduzido, em ausência total deste regulador ou quando as plantas foram estabelecidas na concentração de 0,5mg. L-1 de PBZ, porém subcultivadas posteriormente para meio sem regulador.

Dessa forma são importantes, tanto o estabelecimento de protocolos de propagação eficientes de variedades silvestres, como ações de conservação de germoplasma. A micropropagação e a conservação in vitro são técnicas de cultura de tecidos com potencial para dar suporte tanto ao melhoramento genético quanto á conservação de germoplasma. Em vista disso o objetivo desse trabalho foi avaliar a eficiência do cultivo de segmentos nodais de genótipos da variedade silvestre Ananas comosus var. ananassoides com vista á micropropagação e conservação in vitro, e avaliar a viabilidade e a posterior taxa de multiplicação de diferentes genótipos de Ananas comosus var. comosus conservados in vitro. Os resultados obtidos de dez genótipos de abacaxi Ananas comosus var. ananassoide (25, 203, 205, 206, 207, 232, 233, 465, 472, e 772) mostraram a eficiência no uso de segmentos nodais para incrementar a micropropagação de genótipos silvestres, sendo que dos dez avaliados o 203 foi o que apresentou o melhor desempenho, tanto na formação de segmentos nodais, como na regeneração de plantas. já a viabilidade dos acessos conservados após um período de quatro meses a um ano de incubação sob condições de crescimento mínimo mostrou que a relação entre o tempo de conservação e o estado fisiológico da planta é resultado de um forte componente genético associado ás condições de cultivo estabelecidas para a conservação in vitro.

Moreover, horticulture provides more than twice the number of jobs compared to traditional cereal crop production, and the shifting of conventional agriculture towards high-value horticulture has increased employment opportunities in developing countries. To exploit this potential, researchers need a vast array of horticultural genetic resources and information on new traits. Horticultural crops, which are only a part of PGRFA (Plant Genetic Resources for Food and Agriculture), are characterized by a wide and varied range of species. In fact, there are five major horticultural crop groups: fruit and nut crops, vegetables, food legumes, roots and tubers, and lastly the ornamental and medicinal group. In this context, the present book provides a comprehensive overview of the current state of conservation and utilization of horticultural genetic resources, addressing contemporary approaches to conservation in connection with different technologies, including biotechnological approaches as practised in India and in some cases, globally. It includes a brief chapter on the unique nature of horticultural genetic resources, providing a rationale for viewing them as being distinct from field crop genetic resources. Subsequent chapters share insights on protocols for the conservation of selected horticultural crops ex situ, and focus on the increased need to complement these efforts with in situ conservation approaches. Geospatial tools are also briefly described, emphasizing their utility with regard to mapping and managing resources. The book also explores the wild gene pool in horticulture crops; discusses legal aspects related to horticultural genetic resources and biotechnological aspects; and describes the key aspects of sustainable management and replenishment.
