Available in PDF, EPUB and Kindle. Book excerpt: This volume provides a comprehensive review of resistance induced by photodynamic therapy (PDT) in tumor cells. Understanding the underlying mechanisms in this process leads to the improvement of therapeutic modality, in combination with chemotherapy, immunotherapy, and radiotherapy. Photodynamic therapy is a minimally invasive therapeutic procedure that can exert a selective or preferential cytotoxic activity toward malignant cells. The procedure involves administration of an intrinsically non-toxic photosensitizing agent (PS) followed by irradiation at a wavelength corresponding to a visible absorption band of the sensitizer. In the presence of oxygen, a series of events lead to direct tumor cell death, damage to the microvasculature, and induction of a local inflammatory reaction. Studies reveal that PDT can be curative, particularly in early stage tumors and this volume explores the potential of PDT, but also reveals strategic approaches to overcome resistance in tumor cells.

Book excerpt: Porphyrin-lipids are ideal building blocks for the formation of self-assembled nanoparticles in multimodal imaging and therapeutics. In this study, a ~120 nm porphyrin-lipid shelled nanoemulsion loaded with paclitaxel (PLNE-PTX) was designed for combination of photodynamic therapy (PDT) and chemotherapy. The stable PLNE-PTX had sufficient loading efficiency of PTX (~3.1 %) and porphyrin (~18.3 %), and efficient tumor accumulation in mice (~5.4 ID %/g). For in vivo studies, the combination of PDT and chemotherapy (~223.2 mm3) illustrated an increased tumor growth inhibition compared with PDT (~566.9 mm3) or chemotherapy alone (~544.3 mm3) on day 16. Furthermore, a 4-fold decreased PTX dosage in combination treatment was achieved compared to chemotherapy alone, which could lower PTX toxicities. No significant side effects were observed from blood tests and H stains in mice after PLNE-PTX injection. Thus, PLNE-PTX is a suitable candidate for the synergistic and safe combination of PDT and chemotherapy for tumor treatment.

Available in PDF, EPUB and Kindle. Book excerpt: Cancer develops from the outgrowth of a clonal population of cells with a genetic pathology to evade cell death and exponential proliferation. It has become a global burden with increasing mortality rates. Lung cancer is a major contributor to cancer fatalities. Conventional therapies have shown advances in treating lung cancer, but the successful eradication of cancer lies in targeting both cancer and cancer stem cells. Cancer stem cells (CSCs) are a ration of cells found within the tumour bulk, capable of cancer initiation, therapy resistance, metastasis and cancer relapse. Photodynamic therapy (PDT) has proven effective in treating lung cancer. PDT exerts selective cell death mechanisms toward cancerous cells. With the use of a photosensitizer (PS) which becomes excited upon irradiation with laser light at a specific wavelength, the PS forms reactive oxygen species (ROS) in turn killing neoplastic cells. Leading therapeutic sequel can be obtained by transcending PDT though combination therapies such as immunotherapy and nanotechnology which will enable PDT to target lung CSCs preventing lung cancer recurrence.

Available in PDF, EPUB and Kindle. Book excerpt: As a new concept of cancer treatment, photodynamic therapy (PDT) has gained great attention in the last few decades. Compared to classical treatments such as surgery, chemotherapy and radiotherapy, PDT is a noninvasive, localized treatment of lesions that shows fewer side effects and has low systemic toxicity. In Chapter One, the basic mechanisms, applications and functional nanomaterials-based drug delivery systems for photodynamic therapy of cancer are reviewed. Chapter Two summarizes the application of different carbon based nanomaterials as agents for PDT and discusses current state-of-the-art use of fullerenes and their derivatives, carbon nanotubes and graphene quantum dots in PDT. Chapter Three covers the benefits and pitfalls of using chemi- and bioluminescent systems as intracellular excitation sources in PDT. Bioluminescence is a widespread natural phenomenon, which consists on emission of light resulting from the oxidation of a substrate in a reaction catalyzed by an enzyme in a biological system. Chapter Four addresses in the synthesis, characterization, and photodynamic activity of a novel hydrophobic photosensitizer 5,10,15,20-tetrakis(benzo[b]thiophene) Porphyrin (BTP) and water soluble photosensitizer 5,10,15,20-Tetrakis(7- sulfonatobenzo[b]thiophene) Porphyrin (SBTP). The authors lab is engaged in the synthesis of PDT molecules incorporating benzothiophene moiety to the meso-position of porphyrin molecules. Chapter Five discusses the Guidelines for Gastroenterological Endoscopy in Patients on Oral Antithrombotic Treatment established by the Japan Gastroenterological Endoscopy Society (JGES).These guidelines classify endoscopic interventions according to the risk of hemorrhage and specify the management of various antithrombotic drugs. Endoscopic mucosal resection (EMR) and endoscopic submucosal dissection (ESD) are classified as high-risk procedures for hemorrhage, but PDT is not included in the guidelines. Because PDT causes ischemic necrosis of tumor tissue, the authors say this could possibly be performed safely in patients on oral antithrombotic therapy. The authors of Chapter Six developed a method of the initial concentration of protoporphyrin-IX in the operated removing glioblastoma tissue using the calibration curve of the fluorescent intensity and the known Pp-IX concentration in order to estimate of the more correlated with the cure rate.

Available in PDF, EPUB and Kindle. Book excerpt: One of the major advances of the last decade concerning the treatment of patients with soft tissue sarcomas is that an increased number of patients are being discussed in multidisciplinary teams prior to the initial treatment. The present volume on soft tissue sarcomas in the series Cancer Treatment and Research reflects the multidisciplinary approach with a focus on recent developments. The availability of new histopathologic techniques has reduced the number of unclassified sarcomas and has furhter increased the importance of the histo pathologist in providing estimates of the prognosis of the patient as well as data for the planning of treatment strategy. Further data for this strategy will be provided by diagnostic imaging. In this field, the role of magnetic reson ance imaging has been further defined. Of utmost importance is the recent trend toward consensus in staging. The modification of the staging system of the American Joint Commission for Cancer Staging and End Results Report ing brings the possibility of a single staging system within reach in the next decade. As surgery still provides the only chance for cure, the importance of being the most sparing as possible is obvious. For this reason, radiotherapy has been applied with success. The introduction of relatively new radiation tech niques is therefore being observed with interest.

Book excerpt: The book covers the principles and applications of the photodynamic therapy (PDT), a new promising platform for veterinarians, to treat intricate diseases like cancer and microbial infections. The therapy involves three combined fundamental elements: 1) the use of photosensitizer drugs; 2) light emitted by different sources and devices and 3) molecular oxygen. This association leads to microbial or cancer cells death obtained by oxidative stress.Antimicrobial PDT is a minimally invasive and cost-effective therapy, and due to its action mechanism, microbial resistance is not observed rendering it a huge differential when compared to antimicrobial drugs such as antibiotics. Antineoplastic PDT acts with high selectivity in cancer cells promoting their death with low adverse effects to the patient ́s health and immune integrity. The book is pioneer in introducing this therapeutic modality for veterinarians. It addresses from basics to clinical practice showing the PDT history and fundamentals for non-experts and reporting a collection of basic and clinical studies about PDT in cancer and infections to ensure its safe use by practitioners. Illustrations are provided to give the reader examples of successful procedure in treating pets or other animals. Future perspectives and innovative applications involving nanotechnology and advanced drug delivery are also presented.Photodynamic therapy is an innovative and advanced tool for veterinarian practitioners, students and researchers looking for expanding their competence in Veterinary Medicine.
